Increasing evidence indicates a role of leptin in immune response, but it remains largely unclear whether leptin signaling is involved in regulating NK cell development in the bone marrow （BM）. In this study, we have characterized NK cell differentiation and maturation in the BM of leptin-receptor deficient db/db mice at a prediabetic stage. Although the BM cellularity was similar to the control value, the total number of NK cells was severely reduced in mutant mice. Flow cytometric analysis of db/db BM cells revealed significantly decreased frequencies of developing NK cells at various stages of differentiation. BM db/db NK cells displayed markedly increased apoptosis but maintained normal cell cycling status and proliferative capacity. Moreover, recombinant leptin could significantly enhance the survival of NK cells from wild-type mice in cultures. Further examination on NK cell functional activity showed that db/db NK cells exhibited normal intrinsic cytotoxicity with significantly increased IL-10 production. Taken together, our findings suggest that leptin signaling regulates NK cell development via enhancing the survival of immature NK cells in mouse BM.

Inhibitory effect of different Dendrobium species on LPS-induced inflammation in macrophages via suppression of MAPK pathways

Dendrobii Caulis(DC), named ‘Shihu' in Chinese, is a precious herb in traditional Chinese medicine. It is widely used to nourish stomach, enhance body fluid production, tonify "Yin" and reduce heat. More than thirty Dendrobium species are used as folk medicine. Some compounds from DC exhibit inhibitory effects on macrophage inflammation. In the present study, we compared the anti-inflammatory effects among eight Dendrobium species. The results provided evidences to support Dendrobium as folk medicine, which exerted its medicinal function partially by its inhibitory effects on inflammation. To investigate the anti-inflammatory effect of Dendrobium species, mouse macrophage cell line RAW264.7 was activated by lipopolysaccharide. The nitric oxide(NO) level was measured using Griess reagent while the pro-inflammatory cytokines were tested by ELISA. The protein expressions of inducible NO synthase(iNOS), cyclooxygenase-2(COX-2) and mitogen-activated protein kinases(MAPKs) phosphorylation were evaluated by Western blotting analysis. Among the eight Dendrobium species, both water extracts of D. thyrsiflorum ***(DTW) and D. chrysotoxum Lindl(DCHW) showed most significant inhibitory effects on NO production in a concentration-dependent manner. DTW also significantly reduced TNF-α, MCP-1, and IL-6 production. Further investigations showed that DTW suppressed iNOS and COX-2 expression as well as ERK and JNK phosphorylation, suggesting that the inhibitory effects of DTW on LPS-induced macrophage inflammation was through the suppression of MAPK pathways. In conclusion, D. thyrsiflorum *** was demonstrated to have potential to be used as alternative or adjuvant therapy for inflammation.

Sequential transcatheter arterial chemoembolization and portal vein embolization before right hemihepatectomy in patients with hepatocellular carcinoma

Prevalence and Parental Awareness of Hearing Loss in Children with Down Syndrome

Background： To establish the prevalence of hearing deficit in children with Down syndrome （DS） in Hong kong as measured by brainstem auditory evoked potentials （BAEP）. The secondary objective is to examine the agreement between BAEP and clinical questioning in detecting hearing deficit in DS. Methods： Consecutive DS patients attending the Down＇s Clinic in a regional pediatric referral center were recruited into this cross-sectional study. BAEP data performed within 12 months were retrieved. The care-taker was interviewed with a structured questionnaire to detect any symptom of hearing impairment. BAEP findings and clinical questionings were compared in an agreement analysis using quadratic weighted kappa statistics. Results： Filly DS patients （35 male, 15 female, mean age 11.70 years ± 5.74 standard deviation） were recruited. Eighteen patients （36.0%） were identified having hearing deficit by BAEP. Among patients with hearing impairment, 13 patients （72.2%） had a conductive deficit, and most have mild to moderate hearing loss. Five patients （27.8%） had sensorineural deficit and most have moderate to severe degree. Eight （44.4%） had bilateral hearing deficit. Care-takers of 13 patients （26.0%） reported symptoms of hearing impairment, with 9 （69.2%） having mild symptoms, 3 （23.1%） had moderate symptoms and 1 （7.7%） had severe symptoms. The weighted kappa was 0.045 （95.0% confidence interval - 0.138-0.229）, indicating very poor strength of agreement between BAEP and clinical questioning. For patients with conductive hearing impairment, only 1 patients （7.7%） recalled history ofotitis media. Conclusions： The estimated point prevalence of hearing impairment in Chinese DS children in Hong kong is 36%. Our finding of poor strength of agreement between objective testing and symptom questioning reflects significant underestimation of hearing impairment by history taking alone. 以Francis背景下通过CRISPR/Cas9技术获得的敲除突变体ossaur55-1、ossaur55-2为对象,研究其是否影响水稻株型、叶色、单株穗质量、结实率等农艺性状的变化,探究基因OsSAUR55是否通过参与GA途径调控水稻株型,为相关激素途径调控水稻株型提供理论依据。调查野生型(WT)与敲除突变体成株期的主要农艺性状,包括株高、穗长、单株穗质量、十粒长、十粒宽、剑叶长、剑叶宽等;利用叶绿素计和光合速率测定仪测定野生型和突变体齐穗期剑叶的叶绿素含量和净光合速率;通过实时荧光定量PCR技术(qPCR)测定基因OsSAUR55在水稻种子萌发的不同时间点和不同组织中的表达情况;通过原核表达法观察蛋白OsSAUR55的亚细胞定位情况;采用液相色谱-串联质谱法(LC-MS/MS)测定野生型和突变体的内源激素(GA和IAA)含量;进一步分析GA途径相关基因的表达情况。结果显示,在成株期,突变体ossaur55-1和ossaur55-2都有显著的表型变化,其中突变体的株高比野生型明显变矮、叶色更绿、叶绿素含量增加、光合作用效率降低、结实率降低、千粒质量降低、单株穗质量降低;定量结果显示,OsSAUR55在胚芽中表达量最高,在剑叶中基本不表达;共聚焦结果显示,OsSAUR55可能定位于细胞膜和细胞核;激素检测结果表明,突变体内源激素GA4的含量显著降低;基因的表达量分析发现,突变体中GA合成途径中基因KAO、GA13ox1、GA20ox1、ko1的表达量显著高于野生型,GA代谢途径中GA2ox5、GA2ox8、GA2ox9的表达量也显著高于野生型。初步推测,敲除基因OsSAUR55导致内源GA4含量降低,可能通过GA途径调控水稻株型,进而影响水稻产量。

Seropharmacological Study on Osteogenic Effects of Post-Absorption Ingredients of An Osteoprotective Herbal Formula

Objective： To further investigate the in vitro effects of an osteoprotective herbal formula ＂ELP＂（Herba Epimedii, Fructus Ligustri Lucidi and Fructus Psora/eae） using seropharmacological approach. Methods： Rats were fed with ELP or its individual component herbs for 2 days. The serum containing the post-absorbed ingredients of the herbal items were collected for cell culture using UMR106 cell, RAW264.7 cell and mesenchymal stem cell （MSC） isolated from the bone marrow of the rats. The effects of the herbal-containing serum on cell toxicity were detected by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay; bromodeoxyuridine assay was conducted to measure the cell proliferation of UMR106 cell and MSC; cell activity was measured using colorimetric method, and mRNA expression of runt-related transcription factor 2 （Runx2）, alkaline phosphatase （ALP） and osteopontin （OPN） of UMR106 and MSC as well as matrix metalloproteinase 9 （MMP-9）, tartrate-resistant acid phosphatase （TRAP） and cathepsin K of RAW264.7 were analyzed using real-time reverse-transcription polymerase chain reaction. Results： ELP and its component serum exhibited no cytotoxic effects on the cells. The ELP-containing serum increased the proliferation of UMR106 cell and MSC by 25.7% and 14.4%, respectively and the alkaline phosphatase activity of MSC was increased by 42.6%. On the contrary, it inhibited the RAW264.7 cell differentiation by 29.2%. ELP serum upregulated the Runx2 expression of UMR and MSC by 1.18 fold and 1.27 fold, respectively. It also upregulated ALP and OPN expression in MSC by 1.69- and 2.12-fold, respectively. On the other hand, ELP serum down-regulated MMP-9 and cathepsin K expression of RAW264.7 cell by 0.46- and 0.36-fold, respectively. Conclusions： The serum of the animals fed with ELP contains active ingredients which are effective in promoting osteogenesis and inhibiting osteoclastogenesis.
