Human mesenchymal stem cells,such as human adipose-derived stem cells(hASCs),are typically cultured on a two-dimensional(2 D)monolayer material surface,on which 2 D culturing methods are easily performed and ***,hASCs usually suffer from decreased pluripotency and differentiation ability when cultured with a 2 D monolayer culturing method compared to hASCs cultured with a three-dimensional(3 D)culturing method,such as suspension *** this study,we evaluated whether the pluripotency and differentiation ability of hASCs can be reversibly changed during sequential cultivation with 2 D and 3 D culturing *** hASCs cultivated with a 3 D culturing process after 2 D culture showed at least 2-fold enhanced pluripotency(Sox2,Nanog,and OCT4)compared with that of hASCs cultured with the 2 D culture process ***,hASCs obtained from the 3 D culture process expressed increased levels of differentiation markers of chondrocytes and osteoblasts compared with hASCs obtained from the 2 D culture process when hASCs were induced to ***,their pluripotency and differentiation ability were extensively reduced when hASCs were shifted from 3 D culture to 2 D culture and vice versa,which indicates that hASCs show reversibility in terms of their pluripotency and differentiation ability depending on their environment in 2 D and 3 D *** reversibility of pluripotency and differentiation ability were found to last for at least 5 passages in culture during the alternative and sequential culture of cells with 2 D and 3 D culturing *** study revealed the importance of the culture microenvironment in maintaining the pluripotency and differentiation ability of hASCs,which may reduce the effects of the aging process in *** discuss whether the environment of stem cell culture(i.e.,2 D or 3 D cultivation)can affect stem cell fate in terms of pluripotency and differentiation reversibility.

A prospective cohort of men with localized prostate cancer on active surveillance protocol in Hong kong,China:what did we learn?

This study aimed to report the outcomes of active surveillance(AS)in the management of low-risk prostate cancer(PCa).It recruited87 men who were prospectively followed up according to the Prostate Cancer Research International Active Surveillance(PRIAS)protocol with local adaptation at SH Ho Urology Centre,Prince of Wales Hospital,Hong kong,*** investigated the predictorsof disease progression and found that baseline prostate-specific antigen density(PSAD)and the presence of the highest ProstateImaging-Reporting and Data System(PI-RADS)score 5 lesion on magnetic resonance imaging(MRI)are significantly correlatedwith disease ***,men with PSAD>0.2 ng ml^(−2)or PI-RADS 4 or 5 lesions had significantly worse upgradingfree survival compared to those with PSAD≤0.2 ng ml−2 and PI-RADS 2 or 3 *** study concludes that AS is a safe andeffective management strategy for selected patients to defer radical treatment and that most disease progression can be detectedafter the first repeated *** combination of PSAD>0.2 ng ml^(−2)and PI-RADS 4 or 5 lesions may serve as a useful predictorof early disease progression and provide a guide to optimize follow-up protocols for men in different risk groups.

Rare finding of primary aortoduodenal fistula on single-photon emission computed tomography/computed tomography of gastrointestinal bleeding: A case report

BACKGROUND Primary aortoduodenal fistula is a rare cause of gastrointestinal(GI)bleeding consisting of abnormal channels between the aorta and GI tract without previous vascular intervention that results in massive intraluminal *** SUMMARY A 67-year-old man was hospitalized for coffee ground vomiting,tarry stools,and colic abdominal *** was repeatedly admitted for active GI bleeding and hypovolemic *** and spontaneously stopped bleeders were undetectable on multiple GI endoscopy,angiography,computed tomography angiography(CTA),capsule endoscopy,and ^(99)mTc-labeled red blood cell(RBC)*** patient received supportive treatment and was discharged without signs of ***,he was re-admitted for bleeder *** CTA after a bleed revealed a small aortic aneurysm at the renal level contacting the fourth portion of the duodenum.A ^(99)mTc-labeled RBC singlephoton emission CT(SPECT)/CT scan performed during bleeding symptoms revealed active bleeding at the duodenal *** to his clinical symptoms(intermittent massive GI bleeding with hypovolemic shock,dizziness,dark red stool,and bloody vomitus)and the abdominal CTA and ^(99)mTc-labeled RBC SPECT/CT results,we suspected a small aneurysm and an aortoduodenal *** duodenal excision and duodenojejunal anastomosis were performed.A 7-mm saccular aneurysm arising from the anterior wall of the abdominal aorta near the left renal artery was *** intravascular stenting of the abdominal aorta was performed and his symptoms *** Our findings suggest that ^(99)mTc-labeled RBC SPECT/CT scanning can aid the diagnosis of a rare cause of active GI bleeding.

以Francis背景下通过CRISPR/Cas9技术获得的敲除突变体ossaur55-1、ossaur55-2为对象,研究其是否影响水稻株型、叶色、单株穗质量、结实率等农艺性状的变化,探究基因OsSAUR55是否通过参与GA途径调控水稻株型,为相关激素途径调控水稻株型提供理论依据。调查野生型(WT)与敲除突变体成株期的主要农艺性状,包括株高、穗长、单株穗质量、十粒长、十粒宽、剑叶长、剑叶宽等;利用叶绿素计和光合速率测定仪测定野生型和突变体齐穗期剑叶的叶绿素含量和净光合速率;通过实时荧光定量PCR技术(qPCR)测定基因OsSAUR55在水稻种子萌发的不同时间点和不同组织中的表达情况;通过原核表达法观察蛋白OsSAUR55的亚细胞定位情况;采用液相色谱-串联质谱法(LC-MS/MS)测定野生型和突变体的内源激素(GA和IAA)含量;进一步分析GA途径相关基因的表达情况。结果显示,在成株期,突变体ossaur55-1和ossaur55-2都有显著的表型变化,其中突变体的株高比野生型明显变矮、叶色更绿、叶绿素含量增加、光合作用效率降低、结实率降低、千粒质量降低、单株穗质量降低;定量结果显示,OsSAUR55在胚芽中表达量最高,在剑叶中基本不表达;共聚焦结果显示,OsSAUR55可能定位于细胞膜和细胞核;激素检测结果表明,突变体内源激素GA4的含量显著降低;基因的表达量分析发现,突变体中GA合成途径中基因KAO、GA13ox1、GA20ox1、ko1的表达量显著高于野生型,GA代谢途径中GA2ox5、GA2ox8、GA2ox9的表达量也显著高于野生型。初步推测,敲除基因OsSAUR55导致内源GA4含量降低,可能通过GA途径调控水稻株型,进而影响水稻产量。

Prevalence and Parental Awareness of Hearing Loss in Children with Down Syndrome

Background： To establish the prevalence of hearing deficit in children with Down syndrome （DS） in Hong kong as measured by brainstem auditory evoked potentials （BAEP）. The secondary objective is to examine the agreement between BAEP and clinical questioning in detecting hearing deficit in DS. Methods： Consecutive DS patients attending the Down＇s Clinic in a regional pediatric referral center were recruited into this cross-sectional study. BAEP data performed within 12 months were retrieved. The care-taker was interviewed with a structured questionnaire to detect any symptom of hearing impairment. BAEP findings and clinical questionings were compared in an agreement analysis using quadratic weighted kappa statistics. Results： Filly DS patients （35 male, 15 female, mean age 11.70 years ± 5.74 standard deviation） were recruited. Eighteen patients （36.0%） were identified having hearing deficit by BAEP. Among patients with hearing impairment, 13 patients （72.2%） had a conductive deficit, and most have mild to moderate hearing loss. Five patients （27.8%） had sensorineural deficit and most have moderate to severe degree. Eight （44.4%） had bilateral hearing deficit. Care-takers of 13 patients （26.0%） reported symptoms of hearing impairment, with 9 （69.2%） having mild symptoms, 3 （23.1%） had moderate symptoms and 1 （7.7%） had severe symptoms. The weighted kappa was 0.045 （95.0% confidence interval - 0.138-0.229）, indicating very poor strength of agreement between BAEP and clinical questioning. For patients with conductive hearing impairment, only 1 patients （7.7%） recalled history ofotitis media. Conclusions： The estimated point prevalence of hearing impairment in Chinese DS children in Hong kong is 36%. Our finding of poor strength of agreement between objective testing and symptom questioning reflects significant underestimation of hearing impairment by history taking alone.
