Inhibitory effect of triptolide on lymph node metastasis in patients with non-Hodgkin lymphoma by regulating SDF-1/CXCR4 axis in vitro

Objective:To evaluate the effect and safety of foot baths with Tangbi Waixi Decoction(TW)in treating patients with diabetic peripheral neuropathy(DPN).Methods:It is a multicenter double-blinded randomized controlled *** with DPN were recruited between November 18,2016 and May 30,2018 from 8 hospitals in *** patients received basic treatments for glycemic *** received foot baths with TW herbal granules either 66.9 g(intervention group)or 6.69 g(control group)for 30 min once a day for 2 weeks and followed by a 2-week rest,as a therapeutic *** the Toronto Clinical Scoring System total score(TCSS-TS)6 points,the patients received a total of 3 therapeutic courses(for 12 weeks)and were followed up for 12 *** primary outcome was change in TCSS-TS score at 12 and 24 *** outcomes included changes in bilateral motor nerve conduction velocity(MNCV)and sensory nerve conduction velocity(SNCV)of the median and common peroneal *** was also ***:Totally 632 patients were enrolled,and 317 and 315 were randomized to the intervention and control groups,*** the 12-week intervention,patients in both groups showed significant declines in TCSSTS scores,and significant increases in MNCV and SNCV of the median and common peroneal nerves compared with pre-treatment(P2 weeks and the increase of SNCV of median nerve at 24 weeks in the control group were greater than those in the intervention group(P0.05),and no serious adverse event was related with ***:Treatment of TW foot baths was safe and significantly benefitted patients with DPN.A low dose of TW appeared to be more effective than a high dose.(Registry *** CTR-IOR-16009331)

Background Pediatric antineutrophil cytoplasmic antibody-associated vasculitis(AAV)is a life-threatening systemic vasculitis featured by liability to renal ***,there are few studies on the risk factors and predictive models for renal outcomes of AAV in *** Data from 179 AAV children in multiple centers between January 2012 and March 2020 were collected *** risk factors and predictive model of end-stage renal disease(ESRD)in AAV were *** Renal involvement was the most typical manifestation(95.5%),and the crescent was the predominant pathological lesion(84.9%).The estimated glomerular filtration rate(eGFR)was evaluated in 114 patients,of whom 59.6%developed ESRD,and the median time to ESRD was 3.20 *** eGFR[P=0.006,odds ratio(OR)=0.955,95%confidence interval(CI)=0.924–0.987]and the percentages of global glomerulosclerosis(pGGS;P=0.018,OR=1.060,95%CI=1.010–1.112)were independent risk factors for ESRD of renal *** on the pGGS and eGFR at renal biopsy,we developed three risk grades of ESRD and one predictive *** Kaplan‒Meier curve indicated that renal outcomes were significantly different in different risk grades(P2)in external *** Renal involvement is the most common manifestation of pediatric AAV in china,of which more than half deteriorates into *** predictive model based on eGFR at renal biopsy and the pGGS may be stable and accurate in speculating the risk of ESRD in AAV children.

We operated a p-type point contact high purity germanium(PPCGe)detector(CDEX-1B,1.008 kg)in the china Jinping Underground Laboratory(CJPL)for 500.3 days to search for neutrinoless double beta(0νββ)decay of^(76)Ge.A total of 504.3 kg⋅day effective exposure data was *** anti-coincidence and the multi/single-site event(MSE/SSE)discrimination methods were used to suppress the background in the energy region of interest(ROI,1989–2089 keV for this work)with a factor of 23.A background level of 0.33 counts/(keV⋅kg⋅yr)was *** lower limit on the half life of^(76)Ge 0νββdecay was constrained as T_(1/2)^(0ν)>1.0×10^(23)yr(90%C.L.),corresponding to the upper limits on the effective Majorana neutrino mass:2–7.5 eV.

The proton-proton(pp)fusion chain dominates the neutrino production in the *** uncertainty of the predicted pp neutrino flux is at the sub-percent level,whereas that of the best measurement is O(10%).In this study,for the first time,we measure solar pp neutrinos in the electron recoil energy range from 24 to 144 keV using the PandaX-4T commissioning data with 0.63 tonne×year *** pp neutrino flux is determined as(8.0±3.9(stat)±10.0(syst))×1010 s^(-1)cm^(-2),which is consistent with the Standard Solar Model and existing measurements,corresponding to an upper flux limit of 23.3×10^(10)s^(-1)cm^(-2)at 90%C.L..

Aims:Surveys and research on the applications of the hepatic venous pressure gradient(HVPG)are important for understanding the current status and future development of this technology in *** article aimed to investigate the status of hepatic venous pressure gradient measurement in china in ***:We investigated the overall status of HVPG technology in china-including hospital distribution,hospital level,annual number of cases,catheters used,average cost,indications,and current challenges by using online *** counting the number and percentages of cases of these results,we hope to clarify the current status of HVPG measurements in ***:According to the survey,85 hospitals in china used HVPG technology in 2022 distributed across 29 provinces.A total of 4989 HVPG measurements were performed in all of the surveyed hospitals in 2022,of which 2813 cases(56.4%)were measured *** average cost of HVPG measurement was 5646.8±2327.9 *** the clinical teams who performed the measurements(sometimes multiple per hospital),94.3%(82/87)used the balloon method,and the majority of the teams(72.4%,63/87)used embolectomy ***:This survey clarified the clinical application status of HVPG in china and confirmed that some medical institutions in china have established a foundation for this *** is still necessary to continue promoting and popularizing this technology in the future.

Objective:To assess the efficacy and safety of mulberry twig alkaloids(Sangzhi alkaloids,SZ-A)for treatment of type 2 diabetes in a randomized,double-blind,placebo-controlled multicenter clinical ***:A total of 200 patients were randomized to receive SZ-A(n=100)or placebo(n=100)for 16 *** data analysis system for electronic data capture clinical trial central randomization system was used for randomization and dispensing of *** primary outcome was the change in glycosylated hemoglobin(Hb A1c)*** secondary outcome included the proportions of cases with Hb A1c2h)),body weight,and body mass index(BMI).Adverse events(AEs),severe adverse events(SAEs),treatment-related adverse events(TAEs),gastrointestinal disorders(GDs),blood pressure,routine blood tests,and liver and kidney function were ***:Compared with baseline,the change of Hb A1c at week 16 was-0.80%(95%CI:-0.98%to-0.62%)and-0.09%(95%CI:-0.27%to 0.09%)in SZ-A group and placebo group,*** proportion of patients with Hb A1c21.6%and 29.9%vs.10.8%).The observed values and changes in FBG,1 h-PBG,2 h-PBG,and AUC_(0-2h) differed significantly between groups(P0.05).The incidence rates of AEs,TAEs,and GDs differed significantly between groups(P=0.010,P=0.005,and P=0.006,respectively),whereas the incidence rates of SAEs showed no significant differences between groups(P=1.000).Conclusion:SZ-A are effective and safe for treatment of type 2 diabetes.

Objective:This cross-sectional study aimed to determine the epidemiology of olfactory and gustatory dysfunctions related to COVID-19 in ***:This study was conducted by 45 tertiary Grade-A hospitals in *** and offline questionnaire data were obtained from patients infected with COVID-19 between December 28,2022,and February 21,*** collected information included basic demographics,medical history,smoking and drinking history,vaccination history,changes in olfactory and gustatory functions before and after infection,and other postinfection symptoms,as well as the duration and improvement status of olfactory and gustatory ***:Complete questionnaires were obtained from 35,566 *** overall incidence of olfactory and taste dysfunction was 67.75%.Being female or being a cigarette smoker increased the likelihood of developing olfactory and taste *** received four doses of the vaccine or having good oral health or being a alcohol drinker decreased the risk of such *** infection,the average olfactory and taste VAS scores were 8.41 and 8.51,respectively;after infection,they decreased to 3.69 and 4.29 and recovered to 5.83 and 6.55 by the time of the *** median duration of dysosmia and dysgeusia was 15 and 12 days,respectively,with 0.5%of patients having symptoms lasting for more than 28 *** overall self-reported improvement rate was 59.16%.Recovery was higher in males,never smokers,those who received two or three vaccine doses,and those that had never experienced dental health issues,or chronic accompanying ***:The incidence of dysosmia and dysgeusia following infection with the SARS-CoV-2 virus is high in *** and prognosis are influenced by several factors,including sex,SARS-CoV-2 vaccination,history of head-facial trauma,nasal and oral health status,smoking and drinking history,and the persistence of accompanying symptoms.

目的:观察雷公藤内酯醇对人淋巴瘤细胞系Raji细胞体外增殖和细胞凋亡的影响。方法:实验于2005-04/11在华中科技大学同济医学院附属协和医院血液病研究所实验室完成。①人淋巴瘤细胞系Raji(中国科学院上海生物细胞研究所)。雷公藤内酯醇(Sigma公司)分子式C20H24O6,相对分子量360.4,纯度99%,用二甲基亚砜稀释,微孔滤膜(0.22μm)除菌,等量分装,-20℃保存,使用前解冻。②将Raji细胞放入RPMI-1640培养基中常规培养,每48h换液传代1次。实验前24h半量换液,取处于对数生长期、细胞活性大于98%的细胞进行实验。③采用四甲基偶氮唑蓝比色法检测雷公藤内酯醇对Raji细胞增殖的影响。将Raji细胞按2×108L-1接种于96孔培养板,200μL/孔。实验共分5组:雷公藤内酯醇12.5,25,50,100nmol/L浓度组分别加入适量雷公藤内酯醇使其终浓度为12.5,25,50,100nmol/L,空白对照组未加入雷公藤内酯醇,5个平行孔/组。分别于培养12,24,48,72h后每孔加入5g/L的四甲基偶氮唑蓝20μL继续培养。用全自动酶标仪测定490nm波长处吸光度值。细胞增殖抑制率(%)=(1-雷公藤内酯醇各浓度组吸光度值/空白对照组吸光度值)×100%。④采用AnnexinV/PI双标流式细胞术检测雷公藤内酯醇对Raji细胞凋亡的影响。将Raji细胞按2×108L-1接种于6孔培养板,实验分组同上,分别于孵育24,48h后收集细胞,调整各组细胞数为2×108L-1。取195μL细胞悬液,加入异硫氰酸荧光素标记的AnnexinV室温孵育,洗涤后加入碘化丙锭5μL,流式细胞仪检测细胞凋亡率。结果:①雷公藤内酯醇对Raji细胞增殖的影响:不同浓度的雷公藤内酯醇对人淋巴瘤细胞系Raji的增殖均具有抑制作用,且呈时效和量效关系。与空白对照组比较,培养12,24,48,72h后雷公藤内酯醇12.5,25,50,100nmol/L浓度组的细胞增殖抑制率均明显升高(t=18.314～77.366,P均24h时的半数抑制浓度为43.06nmol/L。②雷公藤内酯醇对Raji细胞凋亡的影响:经雷公藤内酯醇作用后,随着药物浓度的增加和作用时间的延长,Raji细胞凋亡率逐渐增高,且呈时效和量效关系。与空白对照组比较,培养24,48h后雷公藤内酯醇12.5,25,50,100nmol/L浓度组的细胞凋亡率均明显升高(t=-23.657～-37.895,P均<0.05)。结论:雷公藤内酯醇对人淋巴瘤细胞系Raji细胞具有明显的生长抑制作用及诱导凋亡发生,且存在浓度和时间依赖关系。提示雷公藤内酯醇可能通过诱导细胞凋亡来有效抑制人淋巴瘤细胞的增殖,从而有望为治疗淋巴瘤提供新的策略。
