microRNA-let-7a promotes E2F-mediated cell proliferation and NFKB activation in vitro

Epigenetic factors, including altered microRNA （miRNA） expression, may contribute to aberrant immune cell function in systemic lupus erythematosus （SLE）. miRNA-let-7a （let-7a） has been shown to directly alter cell cycle progression and proinflammatory cytokine production. Due to the crucial role of let-7a in cell division and inflammation, we investigated let-7a-mediated proliferation and NFKB translocation in J774A. 1 macrophages and mES 13 mesangial cells in vitro. In immune-stimulated cells transfected with let-7a, cell proliferation was significantly increased over time. There was a significant increase in the number of immune-stimulated cells in S and G2 phases. Immune-stimulated cells overexpressing let-7a had increased nuclear translocation of NFKB. Bioinformatical analysis revealed that the E2F family, critical regulators of the G1-S transition, has potential binding sites for let-7a in their mRNA transcripts. Let-7a overexpression significantly increased the expression of the cell cycle activator E2F2 and increased retinoblastoma protein （Rb） phosphorylation in immune-stimulated cells. The cell cycle inhibitor E2F5 was significantly decreased in let-7a-transfected cells that were immune-stimulated. Bioinformatical analysis revealed E2F2 and NFKB are transcription factors predicted to regulate the let-7a promoter. We analyzed transcriptional regulation of let-7a by real-time RT-PCR using chromatin immunoprecipitation with E2F2 and NFKB antibodies. There was an increase in E2F2 and NFKB binding in DNA enriched for the let-7a promoter in immune-stimulated cells. Silencing E2F2 or NFKB significantly decreased let-7a expression and IL-6 production in immune-stimulated cells. Taken together, our results suggest that overexpression of let-7a may contribute to hyperplasia and the proinflammatory response in SLE.

Epigallocatechin-3-gallate （EGCG） attenuates nflammation in mRL/Ipr mouse mesangial cells

Epigallocatechin-3-gallate （EGCG）, a bioactive component of green tea, has been reported to exert anti-inflammatory effects on immune cells. EGCG is also shown to activate the metabolic regulator, adenosine 5＇-monophosphate-activated protein kinase （AmPK）. Reports have also indicated that EGCG inhibits the immune-stimulated phosphoinositide 3-kinase （PI3K）/Akt/mammalian target of rapamycin （mTOR） pathway. The PI3K/Akt/mTOR pathway has been implicated in mesangial cell activation in lupus. mesangial cells from mRUIpr lupus-like mice are hyper-responsive to immune stimulation and overproduce nitric oxide （NO） and other inflammatory mediators when stimulated. In our current studies, we sought to determine the mechanism by which EGCG attenuates immune-induced expression of pro-inflammatory mediators. Cultured mesangial cells from mRL/Ipr mice were pre-treated with various concentrations of EGCG and stimulated with lipopolysaccharide （LPS）/interferon （IFN）-γ. EGCG activated AmPK and blocked LPS/ IFN-γ-induced inflammatory mediator production （iNOS expression, supernatant NO and interleukin-6）. Interestingly, EGCG attenuated inflammation during AmPK inhibition indicating that the anti-inflammatory effect of EGCG may be partially independent of AmPK activation. Furthermore, we found that EGCG effectively inhibited the immune-stimulated PI3K/Akt/mTOR pathway independently of AmPK, by decreasing phosphorylation of Akt, suggesting an alternate mechanism for EGCG-mediated anti-inflammatory action in mesangial cells. Taken together, these studies show that EGCG attenuated inflammation in mRL/Ipr mouse mesangial cells via the PI3K/Akt/mTOR pathway. Our findings suggest a potential therapeutic role for the use of EGCG to regulate inflammation and control autoimmune disease.

Heat shock protein 90 inhibition by 17-DmAG lessens disease in the mRL/Ipr mouse model of systemic lupus erythematosus

Elevated expression of heat shock protein 90 （HSP90） has been found in kidneys and serum of systemic lupus erythematosus （SLE） patients and mRLImp-FasIprIFasJpr（mRLIIpr） autoimmune mice. We investigated if inhibition of HSP90 would reduce disease in mRL/ Ipr mice. In vitro, pretreatment of mesangial cells with HSP90 inhibitor Geldanamycin prior to immune-stimulation showed reduced expression of IL-6, IL-12 and NO. In vivo, we found HSP90 expression was elevated in mRL/Ipr kidneys when compared to C57BL/6 mice and mRIJIpr mice treated with HSP90 inhibitor 17-DmAG. mRIJIpr mice treated with 17-DmAG showed decreased proteinuria and reduced serum anti-dsDNA antibody production. Glomerulonephritis and glomerular IgG and C3 were not significantly affected by administration of 17-DmAG in mRIJIpr. 17-DmAG increased CD8＋ T cells, reduced double-negative T cells, decreased the CD4/CD8 ratio and reduced follicular B cells. These studies suggest that HSP90 may play a role in regulating T-cell differentiation and activation and that HSP90 inhibition may reduce inflammation in lupus.

Turbulent fluxes at the air-sea interface were estimated with data collected in 2011-2020 with a low-profile platform named OCARINA during eight experiments in five regions:2011,2015,and 2016 in the Iroise Sea;2012 in the tropical Atlantic;2014 in the Chilie-Peru upwelling;2017 and 2018 in the mediterranean Sea,and 2018 and 2020 in *** observations were carried out with moderate winds(2-10 m s^(-1))and average wave heights of 1.5 *** this study,the authors used the fluxes calculated by the bulk method using OCARINA-sampled data as the *** data can validate the fluxes estimated from ERAS reanalysis *** OCARINA and ERA5 data were taken *** do this,the authors established an algorithm to extract the OCARINA data as closely as possible to the reanalysis data in time and *** measurements of the OCARINA platform can conclude on the relevance of the widely used reanalysis data.

Surgeon preferences in the treatment of thumb carpometacarpal osteoarthritis

Importance of urease activity on pathogenic differences among Brucella species was evaluated. In cell-free extracts, the B. suis urease showed 12 times greater specific activity than the B. melitensis urease. When Fisher-344 rats were inoculated intraperitoneally (IP), at 1 week post-inoculation (PI), B. melitensis wild type 16 m was recovered from spleens and livers in greater numbers than B. suis wild type 1330. At 8 weeks PI, spleens were clear of B. melitensis, whereas B. suis remained. The wild type and the urease deficient strains of B. suis did not differ from each other in terms of recovery from spleen or liver. Our observations suggest that B. melitensis induces greater acute infectivity in Fisher-344 rats, whereas B. suis causes chronic infectivity;and urease activity has no influence on Brucella infection using an IP route.

Excitons,bound electron–hole pairs,in two-dimensional hybrid organic inorganic perovskites(2D HOIPs)are capable of forming hybrid light-matter states known as exciton-polaritons(E–Ps)when the excitonic medium is confined in an optical *** the case of 2D HOIPs,they can self-hybridize into E–Ps at specific thicknesses of the HOIP crystals that form a resonant optical cavity with the ***,the fundamental properties of these self-hybridized E–Ps in 2D HOIPs,including their role in ultrafast energy and/or charge transfer at interfaces,remain ***,we demonstrate that>0.5µm thick 2D HOIP crystals on Au substrates are capable of supporting multiple-orders of self-hybridized E–P *** E–Ps have high Q factors(>100)and modulate the optical dispersion for the crystal to enhance sub-gap absorption and *** varying excitation energy and ultrafast measurements,we also confirm energy transfer from higher energy E–Ps to lower energy E–***,we also demonstrate that E–Ps are capable of charge transport and transfer at *** findings provide new insights into charge and energy transfer in E–Ps opening new opportunities towards their manipulation for polaritonic devices.

Compared to reforming reactions using hydrocarbons,ethanol steam reforming(ESR)is a sustainable alternative for hydrogen(H_(2))production since ethanol can be produced sustainably using *** work explores the catalyst design strategies for preparing the Ni supported on ZSm-5 zeolite catalysts to promote ***,two-dimensional ZSm-5 nanosheet and conventional ZSm-5 crystal were used as the catalyst carriers and two synthesis strategies,i.e.,in situ encapsulation and wet impregnation method,were employed to prepare the *** on the comparative characterization of the catalysts and comparative catalytic assessments,it was found that the combination of the in situ encapsulation synthesis and the ZSm-5 nanosheet carrier was the effective strategy to develop catalysts for promoting H_(2) production via ESR due to the improved mass transfer(through the 2-D structure of ZSm-5 nanosheet)and formation of confined small Ni nanoparticles(resulted via the in situ encapsulation synthesis).In addition,the resulting ZSm-5 nanosheet supported Ni catalyst also showed high Ni dispersion and high accessibility to Ni sites by the reactants,being able to improve the activity and stability of catalysts and suppress metal sintering and coking during ESR at high reaction ***,the Ni supported on ZSm-5 nanosheet catalyst prepared by encapsulation showed the stable performance with~88% ethanol conversion and~65% H_(2) yield achieved during a 48-h longevity test at 550-C.

A Rare Case of Aortic Valve Endocarditis and Acute meningitis Due to Haemophilus influenzae

HACEK organisms represent a rare but important group of causative pathogens in endocarditis. These bacteria have historically been associated with culture-negative endocarditis;however, modern laboratory techniques have made this less common. In this case, we present a 74-year-old man who presented with acute onset altered mentation, fever, and sepsis. He was ultimately found to have Haemophilus influenzae meningitis, cerebral empyema, aortic valve endocarditis, psoas myositis, and L2 - L3 diskitis with osteomyelitis. Although HACEK organisms are commonly found in the oropharynx and upper respiratory tract in humans, our patient did not report recent preceding dental or ENT procedures. H. influenzae is responsible for approximately 0.16% of all cases of bacterial endocarditis, representing a very limited subset. Although generally considered low virulent pathogens, this case demonstrates the unusual extent of infection from a HACEK organism, H. influenzae, causing aortic valve endocarditis as well as atypical non-cardiac sequelae, including acute meningitis.
