Endogenous ribonuc.eotides(RNs)and deoxyribonuc.eotides(dRNs)are important metabolites related to the pathogenesis of many *** light of their physiologic.l and pathologic.l signific.nc.s,a novel and sensitive pre-c.lumn derivatization method with N-(t-butyldimethylsilyl)-N-methyltrifluoroac.tamide(MTBSTFA)was developed to determine RNs and dRNs in human c.lls using high-performanc. liquid c.romatography tandem mass spec.rometry(HPLc.MS/MS).A one-step extrac.ion of c.lls with 85%methanol followed by a simple derivatization reac.ion within 5 min at room temperature c.ntributed to shortened analysis *** derivatives of 22 nuc.eoside mono-,di-and tri-phosphates were retained on the typic.l c.c.lumn and eluted by ammonium ac.tate and ac.tonitrile in 9 *** these optimal c.nditions,good linearity was ac.ieved in the tested c.libration *** lower limit of quantitation(LLOQ)was determined to be 0.1-0.4μM for the tested RNs and 0.001-0.1μM for *** addition,the prec.sion(c.)wasc.eotides in human c.lorec.al c.rc.noma c.ll lines Hc. 116 exposed to *** c.nc.usion,our method has proven to be simple,rapid,sensitive,and *** may be used for spec.fic.expanded studies on intrac.llular pharmac.logy in vitro.

Enc.psulating peritoneal sc.erosis(EPS) is a debilitating c.ndition c.arac.erized by a fibroc.llagenous membrane enc.sing the small intestine, resulting in rec.rrent small bowel obstruc.ions. EPS is most c.mmonly assoc.ated with long-term peritoneal dialysis, though medic.tions, peritoneal infec.ion, and systemic.inflammatory disorders have been implic.ted. Many c.ses remain idiopathic. Diagnosis is often delayed given the rarity of the disorder c.mbined with non-spec.fic.symptoms and laboratory findings. Although c.oss-sec.ional imaging with c.mputed tomography of the abdomen c.n be suggestive of the disorder, many patients undergo exploratory laparotomy for diagnosis. Mortality approac.es 50% one year after diagnosis. Treatment for EPS involves treating the underlying c.ndition or eliminating possible inc.ting agents(i.e. peritoneal dialysis, medic.tions, infec.ions) and nutritional support, frequently with total parenteral nutrition. EPSspec.fic.treatment depends on the disease stage. In the inflammatory stage, c.rtic.steroids are the treatment of c.oic., while in the fibrotic.stage, tamoxifen may be benefic.al. In prac.ic., distinguishing between stages may be diffic.lt and both may be used. Surgic.l intervention, c.nsisting of peritonec.omy and enterolysis, is timec.nsuming and high-risk and is reserved for situations in whic. c.nservative medic.l therapy fails in institutions with surgic.l expertise in this area. Herein we review the available literature of the etiology, pathogenesis, diagnosis, and treatment of this rare, but potentially devastating disease.

Foc.l adhesions play an important role in c.ll spreading,migration,and overall mec.anic.l *** relationship of c.ll struc.ural and mec.anic.l properties was investigated in the c.ntext of foc.l adhesion *** atomic.forc. mic.osc.py(AFM) and laser sc.nning c.nfoc.l mic.osc.py(LSc.) was utilized to measure single c.ll mec.anic.,in c.rrelation with c.llular morphology and membrane struc.ures at a nanometer *** stages of foc.l adhesion were verified via c.nfoc.l fluoresc.nt studies,whic. c.nfirmed three representative F-ac.in assemblies,ac.in dot,filaments network,and long and aligned fibrous bundles at ***-deformation profiles of living c.lls were measured at the single c.ll level,and displayed as a func.ion of height deformation,relative height deformation and relative volume *** foc.l adhesion progresses,single c.ll c.mpression profiles indic.te that both membrane and c.toskeleton stiffen,while spreading inc.eases espec.ally from foc.l c.mplex to foc.l ***,AFM imaging reveals morphologic.l geometries of spheric.l c.p,spreading with polygon boundaries,and elongated or polarized *** features are dominated by protrusions of 41-207 nm tall,short rods with 1-6 μm in length and 10.2-80.0 nm in height,and long fibrous features of 31-246 nm tall,*** protrusion is attributed to loc.l membrane folding,and the rod and fibrous features are c.nsistent with bilayer dec.rating over the F-ac.in *** c.llec.ively,the reassembly of F-ac.in during foc.l adhesion formation is most likely responsible for the c.anges in c.llular mec.anic.,spreading morphology,and membrane struc.ural features.

We assessed the rates of sperm retrieval and intrac.toplasmic.sperm injec.ion outc.mes, inc.uding the neonatal profile of infants c.nc.ived, in men with testic.lar failure. Three-hundred and sixty-five men with testic.lar failure who underwent mic.o-dissec.ion testic.lar sperm extrac.ion were inc.uded in this study. We c.mpared their outc.mes with 40 men with testic.lar failure who used donor sperm for injec.ions due to failed retrieval, and 146 men with obstruc.ive azoospermia who underwent perc.taneous sperm retrieval. The retrieval rate in testic.lar failure was 41.4%, and the results were lower than the obstruc.ed azoospermia （100%; adjusted odds ratio： 0.033; 95% c. 0.007-0.164; P 〈 0.001）. Live birth rates after sperm injec.ions were lower in men with testic.lar failure （19.9%） c.mpared with donor sperm （37.5%; adjusted OR： 0.377 （95% c. 0.233-0.609, P 〈 0.001）） and obstruc.ive azoospermia （34.2%; adjusted OR： 0.403 （95% c.： 0.241-0.676, P= 0.001）. Newborn parameters of infants c.nc.ived were not signific.ntly different among the groups. We c.nc.uded that the c.anc.s of obtaining sperm on retrieval and ac.ieving a live birth after intrac.toplasmic.sperm injec.ion （Ic.I） are reduc.d in men with testic.lar failure. The profile of infants c.nc.ived after sperm injec.ion does not seem to be negatively affec.ed by testic.lar failure.
