Polyester(PEt) was pre-activated by atmospheric air plasma and coated by various inorganic oxide nanoparticles(MOx) such as titanium dioxide(tiO2), zinc oxide(ZnO), and silicon oxide(SiO2), using poly(vinylidene f.uoride)(PVDf. and chitosan(Ct) as binders. the resulting PEt-PVDf.MOx-Ct composites were thermally compressed and then characterized by scanning electron microscopy, f.urier inf.ared spectroscopy, thermal gravimetric analysis, and f.ame retardancy(f.) ability tests. PEt modif.cations resulted in more thermally stable and less harmf.l composites with weaker hazardous gas release. this was explained in terms of.structure compaction that blocks pyrolysis gas *** incorporation was f.und to reduce the material susceptibility to oxidation. this judicious procedure also allowed improving f.ame retardancy ability, by lengthening the combustion delay and slowing the f.ame propagation. Chitosan also turned out to contribute to a possible synergy with the other polymers present in the synthesized materials. these results provide valuable data that allow understanding the f. phenomena and envisaging low-cost high f. materials f.om biodegradable raw materials.

A new institutional clinical trial assessed the improvement of.sleep disorders in 40 children with autism treated by immediate-release melatonin f.rmulation in dif.erent regimens(0.5 mg, 2 mg, and 6 mg daily) f.r one month. the objectives of.present study were to(i) prepare low-dose melatonin hard capsules f.r pediatric use controlled by two complementary methods and(ii) carry out a stability study in order to determine a use-bydate. Validation of.preparation process was claimed as ascertained by mass unif.rmity of.hard *** analysis by attenuated total ref.ectance f.urier transf.rmed inf.ared(AtR-f.IR) of.melatonin/microcrystalline cellulose mixture allowed to identif. and quantif. relative content of.active pharmaceutical ingredients and excipients. Absolute melatonin content analysis by high perf.rmance liquid chromatography in 0.5 mg and 6 mg melatonin capsules was 93.6% ± 4.1% and 98.7% ± 6.9% of.theoretical value, respectively. f.rced degradation study showed a good separation of.melatonin and its degradation products. the capability of.the method was 15, conf.rming a risk of.f.lse negative tability test and dissolution test were compliant over 18 months of.storage with European Pharmacopoeia. Preparation of.melatonin hard capsules was completed manually and melatonin in hard capsules was stable f.r 18 months, in spite of.low doses of.active ingredient. AtR-f.IR of.ers a real alternative to HPLC f.r quality control of.highdose melatonin hard capsules bef.re the release of.clinical batches.

Radicle emergence and reserves mobilization are two distinct programmes that are thought to control germination. Both programs are inf.uenced by abscissic acid （ABA） but how this hormone controls seed germination is still poorly known. Phenotypic and microscopic observations of.the embryo axis of.Medicago truncatula during germination in mitotic inhibition condition triggered by 10 μM oryzalin showed that cell division was not required to allow radicle emergence. A suppressive subtractive hybridization showed that more than 10% of.up-regulated genes in the embryo axis encoded proteins related to cell-wall biosynthesis. the expression of.α-expansins, pectin-esterase, xylogucan-endotransglycosidase, cellulose synthase, and extensins was monitored in the embryo axis of.seeds germinated on water, constant and transitory ABA. these genes were overexpressed bef.re completion of.germination in the control and strongly inhibited by ABA. the expression was re-established in the ABA transitory-treatment af.er the seeds were transf.rred back on water and proceeded to germination. this proves these genes as contributors to the completion of.germination and strengthen the idea that cell-wall loosening and remodeling in relation to cell expansion in the embryo axis is a determinant f.ature in germination. Our results also showed that ABA controls germination through the control of.radicle emergence, namely by inhibiting cell-wall loosening and expansion.

the vast majority of.f.xp3^(+)regulatory t cells(tregs)are generated in the thymus,and several f.ctors,such as cytokines and unique thymic antigen-presenting cells,are known to contribute to the development of.these thymus-derived tregs(ttregs).Here,we report the existence of.a specif.c subset of.f.xp3^(+)tregs within the thymus that is characterized by the expression of.IL-1R2,which is a decoy receptor f.r the inf.ammatory cytokine *** f.ow cytometric analysis of.the thymocytes f.om f.xp3^(hCD2)xRAG1^(Gf.) reporter mice revealed that the IL-1R2^(+)tregs are mainly RAG1^(Gf.-)and CCR6^(+)CCR7^(-),demonstrating that these tregs are recirculating cells entering the thymus f.om the periphery and that they have an activated *** the spleen,the majority of.IL-1R2^(+)tregs express neuropilin-1(Nrp-1)and Helios,suggesting a thymic origin f.r these ***,among all tissues studied,the highest f.equency of.IL-1R2^(+)tregs was observed in the thymus,indicating pref.rential recruitment of.this treg subset by the *** f.tal thymic organ cultures(f.OCs),we demonstrated that increased concentrations of.exogenous IL-1β blocked intrathymic treg development resulting in a decreased f.equency of.CD25^(+)f.xp3^(+)ttregs and an accumulation of.CD25^(+)f.xp3^(-)treg ***,the addition of.IL-1R2^(+)tregs,but not IL-1R2^(+)tregs,to reaggregated thymic organ cultures(RtOCs)abrogated the IL-1β-mediated blockade,demonstrating that these recirculating IL-1R2^(+)tregs can quench IL-1 signaling in the thymus and thereby maintain thymic treg development even under inf.ammatory conditions.

在景观尺度,德国的GISCAME把权属、经营措施等森林经理学方法对森林功能与服务的影响加入到森林多目标规划中,对多权属区域森林多目标权衡分析具有重要意义。以生物量规划为例,该平台引入一种提高生物量和生态服务价值在时间和空间上动态权衡的土地利用策略模型,通过生长与收获模型结合的评估方法,解决了小班经营决策和区域规划目标不统一的问题,用情景分析来检验不同措施选择在应对气候变化和生物量增加要求上的差异。从森林经营管理的角度来看,该系统在生长与收获模型、森林调查数据、权属转换率、可及性、保护等级、皆伐或者间伐经营措施改变等方面还存在不确定性假设问题。为将GISCAME平台引进到我国森林经营管理中还需要加强嵌套精准模拟和景观资源可及性等问题研究,系统探索森林权属功能潜力及森林保护等级功能潜力数量差异,为森林景观精准规划与经营管理提供科学参考。

Expression of.tissue f.ctor in pancreatic adenocarcinoma is associated with activation of.coagulation

AIM： to study expression of.tissue f.ctor （tf. in pancreatic cancer and its role in the development of.thromboembolism.MEtHODS： tf.expression was studied in eight human pancreatic carcinoma cell lines by Northern blot and indirect immunof.uorescence. Expression of.alternatively spliced tf.（astf. was assessed by Rt-PCR. In addition, tf.expression was determined by immunof.uorescence in pancreatic tissues of.19 patients with pancreatic adenocarcinoma （PCa）, 9 patients with chronic pancreatitis （CP） and 20 normal controls. Plasma samples （30 PCa-patients, 13 CP-patients and 20 controls） were investigated f.r soluble tf.levels and coagulation activation markers [thrombin-antithrombin Ⅲ complex （tAt）, prothrombin f.agment 1 ＋ 2 （f. ＋ 2）]. RESULtS： All pancreatic carcinoma cell lines expressed tf.（8/8） and most of.them expressed astf.（6/8）. tf.expression at the protein level did not correlate with the dif.erentiation of.the carcinoma cell line. All but two pancreatic cancer tissue samples stained positive f.r tf.（17/19）. In all samples of.CP weak staining was restricted to pancreatic duct cells, whereas only a f.w subendothelial cells were positive in 9/20 of.normal controls. tf.and tAt levels in PCa patients were signif.cantly elevated compared to controls whereas elevated f. ＋ 2 levels did not reach statistical signif.cance compared to controls. In CP patients tAt and f. ＋ 2 levels proved to be signif.cantly elevated compared to controls, although tAt elevation was less pronounced than in PCa patients. CONCLUSION： We conclude that in addition to the upregulated expression of.tf.on the cell membrane, soluble tf.might contribute to activation of.the coagulation system in pancreatic cancer.

A direct real-time polymerase chain reaction assay f.r rapid high-throughput detection of.highly pathogenic North American porcine reproductive and respiratory syndrome virus in China without RNA purif.cation

Background： Porcine reproductive and respiratory syndrome virus （PRRSV）, and particularly its highly pathogenic genotype （HP-PRRSV）, have caused massive economic losses to the global swine industry. Results： to rapidly identif. HP-PRRSV, we developed a direct reaL-time reverse transcription polymerase chain reaction method （dRt-PCR） that could detect the virus f.om serum specimen without the need of.RNA purif.cation Our dRt-PCR assay can be completed in 1.5 h f.om when a sample is received to obtaining a result. Additionally, the sensitivity of.dRt-PCR matched that of.conventional reverse transcription PCR （cRt-PCR） that used purif.ed RNA the lowest detection limit of.HP-PRRSV was 6.3 tCIDs0 using dRt-PCR. We applied dRt-PCR assay to 144 f.eld samples and the results showed strong consistency with those obtained by cRt-PCR. Moreover, the dRt-PCR method was able to tolerate 5-20% （v/v） serum. Conclusions： Our dRt-PCR assay allows f.r easier, f.ster, more cost-ef.ective and higher throughput detection of.HP-PRRSV compared with cRt-PCR methods. to the best of.our knowledge, this is the f.rst report to describe a real-time Rt-PCR assay capable of.detecting PRRSV in crude serum samples without the requirement f.r purif.ing RNA. We believe our approach has a great potential f.r application to other RNA viruses.

this study aims to investigate the ability of.an imidazolium biobased Zwitterionic Ionic Liquids(ZILs)in enhancing the phytoavailability of.copper f.om garden(G)and vineyard(V)soils using the model plant *** and artif.cially contaminated CuSO_(4) soils,unamended and ZIL-amended soil modalities were *** copper/ZIL molar ratio(1/4)introduced was rationally established based on molecular modeling and on the maximal copper concentration in artif.cially contaminated *** accumulation of.copper in the shoots was detected f.r the uncontaminated and copper contaminated ZIL amended V soils(18.9 and 23.3 mg/kg,respectively)contrary to G soils together with a ZIL concentration of.around 3%(W/W)detected by LC-MS *** data evidenced a Cu-accumulation improvement of.38%and 66%compared to non-amended V soils(13.6 and 13.9 mg/kg respectively).ZIL would be mainly present under Cu(II)-ZIL_(4) complexes in the *** impact on the chemical composition of.shoot was also *** results show that depending on the soils modalitity,the presence of.f.ee copper and/or ZIL led to dif.erent chemical compositions in lignin and monomeric sugar *** the bioref.nery context,perf.rmances of.enzymatic hydrolysis of.shoots were also related to the presence of.both ZIL and copper under f.ee or complex *** assessment of.the vineyard soil samples indicated that the quantity of.copper and ZIL remaining in the soils had no signif.cant *** amendment in a copper-contaminated soil was demonstrated as being a promising way to promote the valorization of.phytoremediation plants.
