Effect of JIANPI HUOXUE decoction on inflammatory cytokine secretion pathway in rat liver with lipopolysaccharide challenge

AIM： To evaluate the effect of Chinese traditional medicinal prescription, JIANPI HUOXUE decoction （JHD） on cytokine secretion pathway in rat liver induced by lipopolysaccharide （LPS）. METHODS： Twenty-four male SD rats were divided into normal group （n = 4）, model group （n = 10） and JHD group （n = 10） randomly. Rats in model group and JHD group were administrated with normal saline or JHD via gastrogavage respectively twice a day for 3 d. One hour after the last administration, rats were injected with LPS via tail vein, 50 μg/kg. Simultaneously, rats in normal group were injected with equivalent normal saline. After LPS stimulation for 1.5 h, serum and liver tissue were collected. Pathological change of liver tissues was observed through hematoxylineosin （H.E.） staining. Tumor necrosis factor alpha （TNF-α） in serum were assayed by enzyme linked immunosorbent assay （ELISA）. The protein expression of TNF-α, phosphorylated inhibit-κB （p-κB） and CD68 in liver were assayed by Western blot. The distribution of CD68 protein in liver was observed through immunohistochemical staining. The mRNA expression of TNF-α, interleukin-6 （il-6）, CD14, toll-like receptor 2 （TLR2） and TLR4 in liver were assayed by real-time ***： Predominant microvesicular change, hepatocyte tumefaction and cytoplasm dilution were observed in liver tissues after LPS administration as well as obvious CD68 positive staining in hepatic sinusoidal. After LPS stimulation, serum TNF-α （31.35 ± 6.06 vs 12225.40 ± 9007.03, P 〈 0.05）, protein expression of CD68 （1.13 ± 0.49 vs 3.36 ±1.69, P 〈 0.05）, p-IκB （0.01 ±0.01 vs 2.07 ＋ 0.83, P 〈 0.01） and TNF-α （0.27 ± 0.13 vs 1.29 ± 0.37, P 〈 0.01） in liver and mRNA expression of TNF-α （1.96 ± 2.23 vs 21.45 ±6.00, P 〈 0.01）, il-6 （4.80 ± 6.42 vs 193.50 ± 36.36, P 〈 0.01） and TLR2 （1.44 ± 0.62 vs 4.16 ± 0.08, P 〈 0.01） in liver were also increased significantly. Sodium butyrate attenuates high-fat diet-induced steatohepatitis in mice by improving gut microbiota and gastrointestinal barrier

AIM To investigate whether gut microbiota metabolite sodium butyrate (NaB) is an effective substance for attenuating non-alcoholic fatty liver disease (NAFLD) and the internal mechanisms. METHODS Male C57BL/6J mice were divided into three groups, normal control were fed standard chow and model group were fed a high-fat diet (HFD) for 16 wk, the intervention group were fed HFD for 16 wk and treated with NaB for 8 wk. Gut microbiota from each group were detected at baseline and at 16 wk, liver histology were evaluated and gastrointestinal barrier indicator such as zonula occluden-1 (ZO-1) were detected by immunohistochemistry and realtime-PCR, further serum or liver endotoxin were determined by ELISA and inflammation-or metabolism-associated genes were quantified by real-time PCR. RESULTS NaB corrected the HFD-induced gut microbiota imbalance in mice, while it considerably elevated the abundances of the beneficial bacteria Christensenellaceae, Blautia and Lactobacillus. These bacteria can produce butyric acid in what seems like a virtuous circle. And butyrate restored HFD induced intestinal mucosa damage, increased the expression of ZO-1 in small intestine, further decreased the levels of gut endotoxin in serum and liver compared with HF group. Endotoxin-associated genes such as TLR4 and Myd88, pro-inflammation genes such as MCP-1, TNF-alpha, il-1, il-2, il-6 and IFN-gamma in liver or epididymal fat were obviously downregulated after NaB intervention. Liver inflammation and fat accumulation were ameliorated, the levels of TG and cholesterol in liver were decreased after NaB intervention, NAS score was significantly decreased, metabolic indices such as FBG and HOMA-IR and liver function indicators ALT and AST were improved compared with HF group. CONCLUSION NaB may restore the dysbiosis of gut microbiota to attenuate steatohepatitis, which is suggested to be a potential gut microbiota modulator and therapeutic substance for NAFLD.

Association among plasma interleukin-18 levels, carotid intimamedia thickness and severity of obstructive sleep apnea

Background Epidermic studies have suggested a pathophysiological link between obstructive sleep apnea hypopnea syndrome （OSAHS） and atherosclerosis （AS）; for which carotid intima-media thickness （IMT） has been considered as an early marker. The pathogenesis by which OSAHS can induce AS has not been elucidated. This study was conducted to investigate the association among plasma interleukin-18 （il-18） levels, carotid IMT and the severity of OSAHS. Methods Based on the apnea hypopnea index （AHI） during sleep monitored by polysomnography, 52 male patients with OSAHS were recruited as the OSAHS group which was further divided into mild OSAHS （n=16）, moderate OSAHS （n=18）, and severe OSAHS （n=18） subgroups. Eighteen healthy subjects were selected as the control group. Of all OSAHS patients, 20 with moderate-to-severe OSAHS underwent continuous positive airway pressure （CPAP） treatment for 90 days. HDL5000 color Doppler ultrasonography was used to measure carotid IMT. Plasma il-18 levels were measured bv ELISA.Results Compared with the plasma il-18 levels in the control group （（250.27±76.48） pg/ml）, there was a significant increase in the mild OSAHS subgroup （（352.08±76.32） pg/ml）, the moderate subgroup （（600.17±83.91） pg/ml）, and the severe OSAHS subgroup （（9797.64 ± 109.83） pg/ml）（all P〈0.01）. Moreover, there was a significant difference in plasma il-18 levels among the three OSAHS subgroups （P〈0.01）. Carotid IMT was significantly greater in the severe OSAHS subgroup than in the mild OSAHS subgroup （P〈0.01）. Before CPAP treatment, plasma il-18 levels were positively correlated with carotid IMT （r=0.486, P 〈0.001） and with AHI （r=0.865, P〈0.001）. On day 90 of CPAP treatment, plasma il-18 levels were significantly declined but carotid IMT was not changed significantly. Epidemiological studies of nuclear industry workers have important role to examine the health effect of low dose and low dose-rate radiation *** this study we analyzed the cohort of Korean nuclear power plants *** cohort consists of 7,459 male workers exposed to ionizing radiation and 6,508 controls without exposure *** paper presents the results of analysis on their cancer incidence between 1993 and 2002.A total of 58 cancers were ascertained for radiation workers and 50 for non-radiation ***-standardized rate ratio for two groups was 1.09（95% CI:0.74,1.62）.Trend test showed that cancer incidence rate in individual dose group was not significantly increased with increasing cumulative dose（p=0.2506）.Excess relative risk for all cancers combined was 6.77（95% CI:-4.02,27.54）per sievert,suggesting no statistically significant association between radiation dose and cancer *** of this study are not conclusive because of considerable *** size of the cohort,small cases of cancer incidence,and young ages of subjects limit the power of the *** we need further follow-up and the expansion of cohort to increase the power and to obtain stable risk estimate.

Aim:To investigate the immunoregulatory functions of water extracts of Hericiumerinaceum （WEHE） focusing on natural killer （NK） cell-based anticancer ***:Mouse splenocytes or purely isolated NK ceils were stimulated with1-100 mg/L WEHE for 24 h followed by co-culture with51Cr-labled Yac-1 cells for4 h,then NK cell-derived cytolytic activity was measured using a *** antibodies against mouse interleukin-12 （il-12） were addedinto the WEHE-stimulated splenocytes,thereafter,cytotoxicity was measured toexamine the involvement of ***-PCR and ELISA analyses were performed toconfirm the induction of transcription and the translation of il-12 and interferon-gamma （IFN-gamma） in the WEHE-treated ***:WEHE enhancedthe cytolytic activity of total splenocytes towards Yac-1 ceils in a ***,this activation was not observed when the NK cells isolatedfrom the splenocytes were treated with ***,the treatment withantibodies against il-12 abolished the effect of WEHE on splenocyte-derivedcytolytic ***-PCR and ELISA analyses showed the induction of il-12 andIFN-gamma in the WEHE-treated ***:WEHE indirectlyactivates the cytolytic ability of NK cells via the induction of il-12 in totalsplenocytes,and possibly via other immuno-mediators or cellular components.
