Microwave induces apoptosis in A549 human lung carcinoma cell line

Background Microwaves have other biological effects on cancer as well besides killing tumor cells by coagulation.Some studies showed that microwaves may induce apoptosis in some tumor cells. The apoptotic effect of microwaves may help in clinic to remove residual malignant cells nearby the primary lesion and avoid relapse subsequently. However,there is little evidence on this subject from lung cancer. We studied the effect of microwaves on inducing apoptosis in the human lung carcinoma cell line A549 cells, aiming to identify its effect on apoptosis.Methods A549 cells were radiated by various intensities and durations of microwaves. Apoptosis induction in A549 cells was analyzed by morphological observations, tetrazolium blue color method （MTT） assays, flow cytometery,immunohistochemistry, and image analyses.Results Morphological changes in A549 cells, including cell shrinking and nuclear pycnosis, were observed after microwave radiation. Microwaves significantly inhibited metabolic activities and induced apoptosis in A549 cells. The results of the MTT assay showed a significant decrease of cell activities in all the radiation groups compared with the normal control （P ＜0.01）. The low point of cell activities often appeared at 6-12 hours after radiation. Apoptosis was also confirmed by flow cytometery. The early stage apoptotic rate reached 6.10%-17.98% and the advanced stage apoptotic rate ＋ necrosis rate reached 8.04%-44.06% at 6 hours after microwave irradiation, in contrast to 2.32% and 4.10% in the respective control groups. Down-regulation of Bcl-2 expression and up-regulation of p53 expression were observed by immunohistochemistry after radiation. In most treated groups, the down-regulation of Bcl-2 expression reached its lowest level at 3-6 hours after radiation （integrated optical density （IOD）-6 hours： 2.13±0.08-5.14±0.13 vs. control： 5.79±0.10, P＜0.01 ） and the up-regulation of P53 expression peaked at about 3 hours （IOD-3 hours： 2.61±0.13-8.07±0.11 vs. control：1.29±0.07, P ＜0.01）. Towards the controlled CVD growth of graphitic B-C-N atomic layer films： The key role of B-C delivery molecular precursor

Graphene-like, ternary system B-C-N atomic layer materials promise highly tunable electronic properties and a plethora of potential applications. However, thus far, experimental synthesis of the B-C-N atomic layers normally yields a microscopic phase-segregated structure consisting of pure C and BN domains. Further, growing the truly ternary B-C-N phase layers with homogenous atomic arrangements has proven to be very challenging. Here, in designing a better- controlled process for the chemical vapor deposition （CVD） growth of B-C-N atomic layer films with the minimized C and BN phase segregation, we selected trimethyl borane （TMB）, a gaseous organoboron compound with pre-existing B--C bonds, as the molecular precursor to react with ammonia （NH3） gas that serves as the nitrification agent. The use of this unique B-C delivery precursor allows for the successful synthesis of high-quality and large-area B-C-N atomic layer films. Moreover, the TMB/NH3 reactant combination can offer a high level of tunability and control of the overall chemical composition of B-C-N atomic layers by regulating the relative partial pressure of two gaseous reactants. Electrical transport measurements show that a finite energy gap can be opened in the as-grown B-C-N atomic layers and its tunability is essentially dependent on the relative C to BN atomic compositions. On the basis of carefully controlled experiments, we show that the pre-existing B-C bonds in the TMB molecular precursor have played a crucial role in effectively reducing the C and BN phase segregation problem, thereby facilitating the formation of truly ternary B-C-N phase atomic layers.

Constructing charge transfer channel between dopants and oxygen vacancies for enhanced visible-light-driven water oxidation

Photocatalytic water oxidation is a crucial step in water splitting,but is generally restricted by the slow kinetics.Therefore,it is necessary to develop high-performance water oxidation photocatalysts.Herein,the Fe-doped Bi2WO6 nanosheets with oxygen vacancies(OVs)were synthesized for enhanced photocatalytic water oxidation efficiency,showing a synergistic effect between Fe dopants and OVs.When a molar fraction of 2%Fe was doped into the Bi2WO6 nanosheets,the visible-light-driven photocatalytic oxygen evolution rate was increased up to 131.3μmol·h^(-1)·g_(cat)^(-1)under ambient conditions,which was more than 3 times that of pure Bi2WO6 nanosheets.The proper doping concentration of Fe could promote the formation of OVs and at the same time modulate the band structure of catalysts,especially the position of the valence band maximum(VBM),leading to effective visible-light absorption and enhanced oxidizing ability of photogenerated holes.With ameliorated localized electron distribution,fast charge transfer channel emerged between the OVs and adjacent metal atoms,which accelerated the charge carrier transfer and promoted the separation of photoexcited electrons and holes.This work provides feasible approaches for designing efficient two-dimensional semiconductor water oxidation photocatalysts that could utilize visible-light,which will make more use of solar energy.

嗜酸性粒细胞(Eos)是一种多功能免疫细胞,在外周血循环和组织中均有分布,主要受IL-5和IL-33所诱导产生。Eos与肺癌在内的多种肿瘤存在相互作用,能够通过直接杀伤、免疫调节、促进肿瘤血管正常化等方式发挥抗肿瘤作用,其功能受诱导的细胞因子、所处的肿瘤微环境(TME)、肿瘤的病理类型、作用的时间不同而具有较强的可塑性。免疫检测点抑制剂(ICI)作为一种新兴免疫疗法极大改变了肺癌的治疗模式,然而,目前在临床实践中,仍然缺乏能够较为稳定且有效地预测其疗效和免疫相关不良反应的生物标志物。相较于PD-L1表达水平、肿瘤突变负荷等生物标志物,Eos作为一种常见的循环血细胞具有检测成本低、易获取等优点。有多项研究发现,Eos在肺癌免疫治疗中的特殊改变,且与患者预后和不良反应有一定的相关性。进一步深入开展相关的机制和临床研究后,Eos将具有成为肺癌免疫治疗中有效生物标志物的能力。

Salmeterol attenuates the inflammatory response in asthma and decreases the pro-inflammatory cytokine secretion of dendritic cells

Salmeterol is a long-acting β2-agonist that activates adenylate cyclase, causing long-lasting bronchodilation and has been used for many years to control asthma. However, little information is available about the immunoregulatory effects of salmeterol. We found that salmeterol decreases the production of pro-inflammatory cytokines in a model of allergen-challenged mice that expressed tumor-necrosis factor-alpha, interleukin-1 and interleukin-6. Dendritic cells （DCs） are antigen-presenting cells and act as sentinels in the airway. We found that salmeterol （10-s mol/I） reduced the inflammation caused by lipopolysaccharide （0.1 pg/ml） in activated murine bone marrow-derived DCs. Moreover, western blots demonstrated that this protective effect was mediated partially by inhibiting signaling through the nuclear factor-kappa B （NF-κB）, mitogen-activated protein kinase （MAPK） pathways and dramatically decreased levels of p-ERK. We suggest that salmeterol regulates the inflammation of allergen-induced asthma by modulating DCs. In conclusion, we provide evidence that DCs are the target immune cells responsible for the action of salmeterol against asthma.

Impact of primary tumor volume and location on the prognosis of patients with locally recurrent nasopharyngeal carcinoma

Introduction:The properties of a tumor itself were considered the main factors determining the survival of patients with locally recurrent nasopharyngeal carcinoma(NPC)treated with intensity-modulated radiotherapy(IMRT).However,recurrent tumors were mainly evaluated by using the American Joint Committee on Cancer staging system,which was modeled on primary tumors and did not incorporate the tumor volume.This study aimed to investigate the prognostic values of the primary tumor location and tumor volume,and to determine whether evaluating these parameters could improve the current staging system.Methods:Magnetic resonance(MR)images for 229 patients with locally recurrent NPC who underwent IMRT were analyzed retrospectively.Results:The skull base,parapharyngeal space,and intracranial cavity were the most common sites of tumors.There was a difference in the survival between patients with T1 and T2 diseases(77.6%vs.50.0%,P38 cm3(48.7%vs.15.2%,P<0.01).Conclusions:A new staging system has been proposed,with T3 tumors being down-staged to T2 and with the tumor volume being incorporated into the staging,which may lead to an improved evaluation of these tumors.This new system can be used to guide the treatment strategy for different risk groups of recurrent NPC.

Time-Modulated Transmissive Programmable Metasurface for Low Sidelobe Beam Scanning

Programmable metasurfaces have great potential for the implementation of low-complexity and low-cost phased arrays.Due to the difficulty of multiple-bit phase control,conventional programmable metasurfaces suffer a relatively high sidelobe level(SLL).In this manuscript,a time modulation strategy is introduced in the 1-bit transmissive programmable metasurface for reducing the SLLs of the generated patterns.After the periodic time modulation,harmonics are generated in each reconfigurable unit and the phase of the first-order harmonic can be dynamically controlled by applying different modulation sequences onto the corresponding unit.Through the high-speed modulation of the real-time periodic coding sequences on the metasurface by the programmable bias circuit,the equivalent phase shift accuracy to each metasurface unit can be improved to 6-bit and thus the SLLs of the metasurface could be reduced remarkably.The proposed time-modulated strategy is verified both numerically and experimentally with a transmissive programmable metasurface,which obtains an aperture efficiency over 34%and reduced SLLs of about-2o dB.The proposed design could offer a novel approach of a programmable metasurface framework for radar detection and secure communication applications.
