PI3K-AKT-mTOR signaling in prostate cancer progression and androgen deprivation therapy resistance

Prostate cancer （PCa） is the second most common malignancy among men in the world. Castration-resistant prostate cancer （CRPC） is the lethal form of the disease, which develops upon resistance to first line androgen deprivation therapy （ADT）. Emerging evidence demonstrates a key role for the PI3K-AKT-mTOR signaling axis in the development and maintenance of CRPC. This pathway, which is deregulated in the majority of advanced PCas, serves as a critical nexus for the integration of growth signals with downstream cellular processes such as protein synthesis, proliferation, survival, metabolism and differentiation, thus providing mechanisms for cancer cells to overcome the stress associated with androgen deprivation. Furthermore, preclinical studies have elucidated a direct connection between the PI3K-AKT-mTOR and androgen receptor （AR） signaling axes, revealing a dynamic interplay between these pathways during the development of ADT resistance. Thus, there is a clear rationale for the continued clinical development of a number of novel inhibitors of the PI3K pathway, which offer the potential of blocking CRPC growth and survival. In this review, we will explore the relevance of the PI3K-AKT-mTOR pathway in PCa progression and castration resistance in order to inform the clinical development of specific pathway inhibitors in advanced PCa. In addition, we will highlight current deficiencies in our clinical knowledge, most notably the need for biomarkers that can accurately predict for response to PI3K pathway inhibitors.

Investigation of a Pressure Sensor with Temperature Compensation Using Two Concentric Wheatstone-Bridge Circuits

＜正＞This paper presents some effective method for improving the performance of a speaker identification system. Based on the multiresolution property of the wavelet transform, the input speech signal is decomposed into various frequency bands in order not to spread noise distortions over the entire feature space. The linear predictive cepstral coefficients （LPCC） of each band are calculated. Furthermore, the cepstral mean normalization technique is applied to all computed features. In order to effectively utilize these multiband speech features, we use feature recombination and likelihood recombination methods to evaluate the task of the textindependent speaker identification. The feature recombination scheme combines the cepstral coefficients of each band to form a single feature vector used to train the Gaussian mixture model （GMM）. The likelihood recombination scheme combines the likelihood scores of independent GMM for each band. Experimental results show that the proposed both methods outperform the GMM model using full-band LPCC and MFCC features in both clean and noisy environments.

This study presents a silicon-based pressure sensor with temperature compensation. The eight piezoresistors were designed on the polycrystalline silicon membrane and constructed by two concentric Wheatstone-bridge circuits to perform two sets of sensors. The sensor in the central circuit measures the membrane deflection caused by the combined effects of pressure and temperature, while the outer one measures only the deflection caused by the working temperature. From this arrangement, it is reliable and accurate to measure the pressure by comparing the output signals from the two concentric Wheatstone-bridge circuits. The optimal positions of the eight piezoresistors were simulated by simulation software ANSYS. The investigated pressure sensor was fabricated by the micro electro-mechanical systems (MEMS) techniques. The measuring performance and an indication of the conventional single Wheatstone-bridge pressure sensor is easily affected under variation of different working temperature and causes a maximum absolute error up to 45.5%, while the double Wheatstone-bridge pressure sensor is able to compensate the error, and reduces it down to 1.13%. The results in this paper demonstrate an effective temperature compensation performance, and have a great performance and stability in the pressure measuring system as well.

Linear and nonlinear analyses of water waves in an elastic vessel are carried out to study the dramatic phenomena of Dragon Wash as well as related controllable experiments. It is proposed that the capillary edge waves are generated by parametric resonance, which is shown to be a possible mechanism for both rectangular an circular vessels. For circular vessel, the normal geometric resonance is also operating, thus greatly. enhance the dramatic effect. The mechanism of nonlinear mode-mode interaction is proposed far the generation of axisymmetric low-frequency gravity waves by the high-frequency external excitation. A simple model system is studied numerically to demonstrate explicitly this interaction mechanism.

Objective:To investigate whether major dengue outbreaks in the last two decades in Kaohsiung follow a precise temporal ***:Government daily lab-confirmed dengue case data from three major dengue outbreaks occurring during the last two decades in Kaohsiung in2002,2014 and 2015,is utilized to compute the corresponding weekly cumulative percentage of total case *** divide each of the three time series data into two periods to examine the corresponding weekly cumulative percentages of case numbers for each ***’s correlation coefficient was calculated to compare quantitatively the similarity between the temporal patterns of these three ***:Three cutoff points produce the most interesting comparisons and the most different ***’s correlation coefficient indicates quantitative discrepancies in the similarity between temporal patterns of the three years when using different cutoff ***:Temporal patterns in 2002 and 2014 are comparatively more similar in early *** 2015 outbreak started late in the year,but ended more like the outbreak in 2014,both with record-breaking number of *** retrospective analysis shows that the temporal dynamics of dengue outbreaks in Kaohsiung can strongly vary from one year to another,making it difficult to identify any common predictor.

Based on the principle of “pre-disaster prevention outweighs rescue during disasters”, this study targets areas threatened by natural disasters, and develops an automatic algorithm based on the Prim algorithm to serve as an automatic identification system. In the face of natural disasters that disable key facilities in the region and prevent settlements from contacting the outside world or outsiders from sending rescuers to the settlements, the proposed system helps to identify whether these regions will become isolated areas and conduct disaster mitigation and relief resource allocation before any natural disaster in order to reduce potential disaster losses. An automatic identification system, based on the threshold of channel blocking due to broken roads and bridges, determines through the decision tree model and relevant patterns whether such regions will become isolated areas by identifying areas based on the results of model analysis. The proposed system’s identification results are verified by actual case histories and comparisons;the results can be used to correctly identify isolated areas. Finally, Microsoft Visual Studio C # and Google Map are employed to apply the results and to produce an information mode for the determination and decision support of isolated areas affected by natural disasters.

Aim： To determine if there are different penile hemodynamic patterns between sildenafil non-responders and responders by using color Doppler ultrasonography. Methods： A total of 69 erectile dysfunction （ED） patients aged 22-79 years were enrolled into the present study. Thirty-eight （55.1%） men with ED who did not respond to four attempts of treatment with 100 mg sildenafil after re-education were classified as sildenafil non-responders. A com- bination of three vasodilator drugs, 1.25 mg papaverine, 0.4 mg phentolamine and 5 ug prostaglandin E1, was given by intracavernous injection before penile Doppler ultrasonography was carried out. The erectile response to intracavernous injection and vascular parameters including peak systolic velocity （PSV）, resistance index （RI）, end diastolic velocity （EDV） and cavernosa artery diameter （CD） were measured and the results between sildenafil nonresponders and responders were compared. Results： No statistical difference in vascular parameters measured by Doppler ultrasonography studies between non-responders and responders was noted. Sildenafil non-responders had a poorer penile rigidity response to intracavernous injection than responders （P 〈 0.05）. Among patients with adequate PSV （〉 30 cm/s） and abnormal EDV （〉 5 cm/s）, individuals in the non-responder group had fewer positive responses to intracavernous vasodilator injection than in the responder group （35.3% vs. 72.2%, P 〈 0.05）. Advanced age and comorbidity with diabetes mellitus were significantly associated with sildenafil non-response （P 〈 0.05）. Conclusion： Sildenafil non-responders were characterized by a poorer penile rigidity response to intracavernous injection and had an associated impaired veno-occlusive mechanism. Advanced age and comorbidity with diabetes mellitus were two common factors associated with non-response.

Electroless Ni-Cu-P deposits were deposited on the Si substrate in a basic hypophosphite-type plating bath. The effects of pH value and the metal source composition, Ni and Cu, in the plating bath on the kinetics of the Ni-Cu-P deposition were studied. The electroless Ni-Cu-P deposits were characterized by a scanning electron microscope, a transmission electron microscope, an energy dispersive X-ray spectroscope, and an X-ray diffractometer. The results showed that the pH value of the plating bath had no obvious effect on the morphology and composition of electroless Ni-Cu-P deposits. However, the composition of the metal source, Ni and Cu, in the plating bath had great effect on the kinetics of electroless Ni-Cu-P deposition.
