STAT1 signaling is required for optimal Th1 cell differentiation in mice

Although IFN-γ alone does not prime type I T helper cell (Th1) differentiation, the loss of IFN-γ signaling leads to impaired Th1 phenotype: IFN-γ receptor-deficient (Ifngr-/-) Th1 cells fail to permanently repress IL-4 expression. They can differentiate into IL-4-producing cells under Th2-inducing conditions. These observations suggest that IFN-γ signaling plays a critical role in si- lencing Il4 gene in Th1 cells and stabilizing Th1 phenotype. IFN-γ signaling has been further shown to inhibit IL-4 expression by inhibiting STAT6 phosphorylation. This work aims to study the mechanism by which STAT1, the downstream molecule that transduces IFN-γ signaling, may mediate suppression of IL-4 expression in Th1 cells. The results show that STAT1-deficient naive CD4+ T cells express a reduced level of IFN-γ as well as an elevated level of IL-4. These cells exhibit bias to differentiate into Th2 cells under unpolarized conditions. Under Th1-inducing conditions, STAT1-deficient naive CD4+ T cells show impaired Th1 differentiation: Stat1-/- Th1 cells express reduced levels of IFN-γ and T-bet. These cells also fail to repress the expression of IL-4 and GATA-3 and retain STAT6 signaling. More importantly, Stat1-/- Th1 cells can be effectively induced to differentiate into IL-4-producing cells under Th2-inducing conditions. Ectopic expression of T-bet in Stat1-/- Th1 cells dramatically represses their ability to do so and drastically restores IFN-γ expression, suggesting that STAT1 may inhibit IL-4 expression through T-bet. Finally, histone H3 acetylation (H3AC) and histone H3 K4 dimethylation (H3K4dim) were observed in two enhancer regions of Il4 gene locus in Stat1-/- Th1 cells, suggesting a permissive status of Il4 gene locus in these cells. AZD1775 synergizes with SLC7A11 inhibition to promote ferroptosis

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies, including WEE1 inhibition. Considering that p53 activation can also lead to multiple types of cell death, the role of this tumor suppressor in WEE1 inhibitor-based therapies remains disputed. In this study, we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer growth. Our findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake, an action similar to that of Erastin. Meanwhile, inhibition of WEE1 by the WEE1 inhibitors or si RNAs induced compensatory upregulation of SLC7A11, which conferred resistance to ferroptosis. Mechanistically, AZD1775 prevented the enrichment of H3K9me3, a histone marker of transcriptional repression, on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1, thereby enhancing NRF2-mediated SLC7A11 transcription. This finding was also validated using the H3K9me3 inhibitor BRD4770. Remarkably, we found that the nucleolar stress-inducing agent Actinomycin D(Act. D) inhibited SLC7A11 expression by activating p53, thus augmenting AZD1775-induced ferroptosis. Moreover, the combination of AZD1775 and Act. D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in vivo. Altogether, our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription, and inhibition of SLC7A11 by Act. D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

Solar flare forecasting based on a Fusion Model

Solar flare prediction is an important subject in the field of space *** learning technology has greatly promoted the development of this *** this study,we propose a novel solar flare forecasting model integrating Deep Residual Network(ResNet)and Support Vector Machine(SVM)for both≥C-class(C,M,and X classes)and≥M-class(M and X classes)*** collected samples of magnetograms from May 1,2010 to September 13,2018 from Space-weather Helioseismic and Magnetic Imager(HMI)Active Region Patches and then used a cross-validation method to obtain seven independent data *** then utilized five metrics to evaluate our fusion model,based on intermediate-output extracted by ResNet and SVM using the Gaussian kernel *** results show that the primary metric true skill statistics(TSS)achieves a value of 0.708±0.027 for≥C-class prediction,and of 0.758±0.042 for≥M-class prediction;these values indicate that our approach performs significantly better than those of previous *** metrics of our fusion model’s performance on the seven datasets indicate that the model is quite stable and robust,suggesting that fusion models that integrate an excellent baseline network with SVM can achieve improved performance in solar flare ***,we also discuss the performance impact of architectural innovation in our fusion model.

Event-triggered model-free adaptive control for a class of surface vessels with time-delay and external disturbance via state observer

This paper provides an improved model-free adaptive control(IMFAC)strategy for solving the surface vessel trajectory tracking issue with time delay and restricted ***,the original nonlinear time-delay system is transformed into a structure consisting of an unknown residual term and a parameter term with control inputs using a local compact form dynamic linearization(local-CFDL).To take advantage of the resulting structure,use a discrete-time extended state observer(DESO)to estimate the unknown residual ***,according to the study,the inclusion of a time delay has no effect on the linearization structure,and an improved control approach is provided,in which DESO is used to adjust for ***,a DESO-based event-triggered model-free adaptive control(ET-DESO-MFAC)is established by designing event-triggered conditions to assure Lyapunov *** when the system’s indicator fulfills the provided event-triggered condition will the control input signal be updated;otherwise,the control input will stay the same as it is at the last trigger moment.A coordinate compensation approach is developed to reduce the steady-state inaccuracy of trajectory ***,simulation experiments are used to assess the effectiveness of the proposed technique for trajectory tracking.

A method using N,O-bis（trimethylsilyl）acetamide/N-hydroxysuccinimide ester（BSA/NHS） as coupling agents for dipeptide synthesis is descried. The coupling reaction between N-hydroxysuccinimide（NHS）esters and amines could be performed under mild conditions with N,O-bis（trimethylsilyl）acetamide（BSA） as coupling reagent and no additional acid/base is required. All byproducts and excessive reactants are water soluble or hydrolysable and easy to eliminate through water-washing at the purification ***, all the reactants are inexpensive and widely used in conventional drug production.

On learning the right attention point for feature enhancement

We present a novel attention-based mechanism to learn enhanced point features for point cloud processing tasks, e.g., classification and segmentation. Unlike prior studies, which were trained to optimize the weights of a pre-selected set of attention points, our approach learns to locate the best attention points to maximize the performance of a specific task, e.g., point cloud classification. Importantly, we advocate the use of single attention point to facilitate semantic understanding in point feature learning. Specifically,we formulate a new and simple convolution, which combines convolutional features from an input point and its corresponding learned attention point(LAP). Our attention mechanism can be easily incorporated into state-of-the-art point cloud classification and segmentation networks. Extensive experiments on common benchmarks, such as Model Net40, Shape Net Part, and S3DIS, all demonstrate that our LAP-enabled networks consistently outperform the respective original networks, as well as other competitive alternatives, which employ multiple attention points, either pre-selected or learned under our LAP framework.

Comparative evaluation of data mining methods in predicting the water vapor permeability of cement-based materials

Water vapor permeability of building materials is a crucial parameter for analysing and optimizing the hygrothermal performance of building envelopes and built *** measurement is accurate but time-consuming,while data mining methods have the potential to predict water vapor permeability *** this study,six data mining methods—support vector regression(SVR),decision tree regression(DT),random forest regression(RF),K-nearest neighbor(KNN),multi-layer perceptron(MLP),and adaptive boosting regression(AdaBoost)—were compared to predict the water vapor permeability of cement-based materials.A total of 143 datasets of material properties were collected to build prediction models,and five materials were experimentally determined for model *** results show that RF has excellent generalization,stability,and *** has great generalization and precision,only slightly inferior to the former,and its stability is *** has good precision and acceptable generalization,but its stability is *** and KNN have superior stability,but their generalization and precision are *** lacks generalization,and its stability and precision are *** short,RF has the best comprehensive performance,demonstrated by a limited prediction deviation of 26.3%from the experimental results,better than AdaBoost(38.0%)and DT(38.3%)and far better than other remaining *** is also found that data mining methods provide better predictions when cement-based materials’water vapor permeability is high.
