Preparation of true solutions of monomeric amyloidogenic protein/peptide:A critical prerequisite for aggregation kinetic study

Our dynamic laser light scattering(LLS) study shows that the current widely used protocols of dissolving amyloidogenic protein/peptide do not really result in a true solution;namely,there always exist a trace amount of interchain aggregates,which greatly affect the association kinetics,partially explaining why different kinetics were reported even for a solution with identical protein and ***,using a combination of the conventional dissolution procedure and our newly developed ultra-filtration method,we have developed a novel protocol to prepare a true solution of amyloidogenic protein/peptide without any interchain *** resultant solutions remain in their monomeric state for at least one week,which is vitally important for further study of the very initial stage of the interchain association under the physiological conditions because more and more evidence suggests that it is those small oligomers rather than large fabric aggregates that are *** addition,this study shows that combining static and dynamic LLS can lead to more physical and microscopic information about the protein association instead of only the size distribution.

Federated learning has emerged as a distributed learning paradigm by training at each client and aggregat-ing at a parameter *** heterogeneity hinders stragglers from responding to the server in time with huge com-munication *** client grouping in federated learning can solve the straggler problem,the stochastic selection strategy in client grouping neglects the impact of data distribution within each ***,current client grouping ap-proaches make clients suffer unfair participation,leading to biased performances for different *** order to guaran-tee the fairness of client participation and mitigate biased local performances,we propose a federated dynamic client selec-tion method based on data representativity(FedSDR).FedSDR clusters clients into groups correlated with their own lo-cal computational *** estimate the significance of client datasets,we design a novel data representativity evalua-tion scheme based on local data ***,the two most representative clients in each group are selected to optimize the global ***,the DYNAMIC-SELECT algorithm updates local computational efficiency and data representativity states to regroup clients after periodic average *** on real datasets show that FedS-DR improves client participation by 27.4%,37.9%,and 23.3%compared with FedAvg,TiFL,and FedSS,respectively,tak-ing fairness into account in federated *** addition,FedSDR surpasses FedAvg,FedGS,and FedMS by 21.32%,20.4%,and 6.90%,respectively,in local test accuracy variance,balancing the performance bias of the global model across clients.

Meningeal malignant melanosis is a meninges tumor that can produce melanin. Primary intracranial neurocutaneous melanosis is rare. It grows fast with a high degree of malignancy and is associated with earlier intracranial hypertension and meningeal irritation. Most of them can not be diagnosed unless by biopsy or autopsy1. lntracranial malignant melanosis is divided into two kinds： primary and secondary which transfers through the lymph or blood. We report one case of meningeal malignant melanosis that was confirmed by meningeal pathology.

The problem of free surface is one of main difficulties in the numerical calculation of turbulent flow. This paper presents the concept of 'elastic-lid' for the first time, which is used to calculate the steady turbulent flow domains with complex free surfaces. This method can be combined with the existing steady turbulence models. Compared with MAC and other unsteady methods, it is simpler and can give us the parameters related to turbulent flow. The method is supported by the techniques of 'block-off' and 'elastic-grid', which can be selected for different problems. Finally, three examples are given to show the application of the 'elastic-lid' method. The calculated results are in good agreement with the measured.

Composite biomaterials made of biodegradable polylactic acid （PLA） and bioactive magnesium （Mg） salt are developed for orthopaedic implants or metal implant coatings. The releasing of Mg salt into the biological environment benefits the bone growth, while with the releasing of Mg salt and degradation of PLA there forms a porous scaffold for tissue engineering. The size and morphology of the salt and voids are adjustable with such preparation conditions as salt content, pH of casting solution, and the solidification rate, so that we can control the salt releasing and degradation rate of PLA. Dielectric spectroscopy is used to investigate the dispersive structures of Mg salt and voids in the polymer matrix and to monitor the in situ releasing of Mg salts in the simulated body fluid （SBF）. The current study provides us with an orthopedic biomaterial with controllable multi-phase structures, and a tool to investigate the in vivo behaviors of biomaterials.

A waveguide coupled surface plasmon sensor for detection of liquid with high refractive index(RI)is designed based on polymer *** effects of variation of the thickness of the Au film,polymethyl methacrylate(PMMA)buffer,and waveguide layer on the sensing performance of the waveguide are comprehensively investigated by using the finite difference *** simulations show that a thinner gold film gives rise to a more sensitive structure,while the variation of the thickness of the PMMA buffer and waveguide layer has a little effect on the *** liquid with high RI,the sensitivity of the sensor increases *** RI of liquid to be measured increases from 1.45 to 1.52,the sensitivity is as high as 4518.14 nm/RIU,and a high figure of merit of 114.07 is *** waveguide coupled surface plasmon RI sensor shows potential applications in the fields of environment,industry,and agriculture sensing with the merits of compact size,low cost,and high integration density.

This study aimed to report the outcomes of active surveillance(AS)in the management of low-risk prostate cancer(PCa).It recruited87 men who were prospectively followed up according to the Prostate Cancer Research International Active Surveillance(PRIAS)protocol with local adaptation at SH Ho Urology Centre,Prince of Wales Hospital,Hong Kong,*** investigated the predictorsof disease progression and found that baseline prostate-specific antigen density(PSAD)and the presence of the highest ProstateImaging-Reporting and Data System(PI-RADS)score 5 lesion on magnetic resonance imaging(MRI)are significantly correlatedwith disease ***,men with PSAD>0.2 ng ml^(−2)or PI-RADS 4 or 5 lesions had significantly worse upgradingfree survival compared to those with PSAD≤0.2 ng ml−2 and PI-RADS 2 or 3 *** study concludes that AS is a safe andeffective management strategy for selected patients to defer radical treatment and that most disease progression can be detectedafter the first repeated *** combination of PSAD>0.2 ng ml^(−2)and PI-RADS 4 or 5 lesions may serve as a useful predictorof early disease progression and provide a guide to optimize follow-up protocols for men in different risk groups.

Breast cancer,a predominant global health issue,requires ongoing exploration of new therapeutic ***(PAL),a well-known cyclin-dependent kinase(CDK)inhibitor,plays a critical role in breast cancer *** its efﬁcacy is recognized,the interplay between PAL and cellular autophagy,particularly in the context of the RAF/MEK/ERK signaling pathway,remains insufﬁciently *** study investigates PAL’s inhibitory effects on breast cancer using both in vitro(MCF7 and MDA-MB-468 cells)and in vivo(tumor-bearing nude mice)*** at elucidating the impact of PAL on autophagic processes and exploring the potential of combining it with trametinib(TRA),an MEK inhibitor,our research seeks to address the challenge of PAL-induced drug ***ﬁndings reveal that PAL signiﬁcantly decreases the viability of MCF7 and MDA-MB-468 cells and reduces tumor size in mice while showing minimal cytotoxicity in MCF10A ***,PAL also induces protective autophagy,potentially leading to drug resistance via the RAF/MEK/ERK pathway *** TRA effectively neutralized this autophagy,enhancing PAL’s anti-tumor efﬁcacy.A combination of PAL and TRA synergistically reduced cell viability and proliferation,and in vivo studies showed notable tumor size *** conclusion,the PAL and TRA combination emerges as a promising strategy for overcoming PAL-induced resistance,offering a new horizon in breast cancer treatment.

Objective:Cytogenetic abnormalities have been proven to be the most valuable parameter for risk stratification of childhood acute lymphoblastic leukemia(ALL).However,studies on the prevalence of cytogenetic abnormalities and their correlation to clinical features in chinese pediatric patients are limited,especially large-scale ***:We collected the cytogenetics and clinical data of 1541 children newly diagnosed with ALL between 2001 and 2014 in four chinese hospitals,and retrospectively analyzed their clinical features,prognosis and risk factors associated with pediatric ALL.
