Camouflaging attenuated Salmonella by cryo-shocked macrophages for tumor-targeted therapy

live bacteria-mediated antitumor therapies mark a pivotal point in cancer immunotherapy.However,the difficulty in reconciling the safety and efficacy of bacterial therapies has limited their application.Improving bacterial tumor-targeted delivery while maintaining biosafety is a critical hurdle for the clinical translation of live microbial therapy for cancer.Here,we developed“dead”yet“functional”Salmonella-loaded macrophages using liquid nitrogen cold shock of an attenuated Salmonella typhimurium VNP20009-contained macrophage cell line.The obtained“dead”macrophages achieve an average loading of approximately 257 live bacteria per 100 cells.The engineered cells maintain an intact cellular structure but lose their original pathogenicity,while intracellular bacteria retain their original biological activity and are delay freed,followed by proliferation.This“Trojan horse”-like bacterial camouflage strategy avoids bacterial immunogenicity-induced neutrophil recruitment and activation in peripheral blood,reduces the clearance of bacteria by neutrophils and enhances bacterial tumor enrichment efficiently after systemic administration.Furthermore,this strategy also strongly activated the tumor microenvironment,including increasing antitumor effector cells(including M1-like macrophages and CD8+Teffs)and decreasing protumor effector cells(including M2-like macrophages and CD4+Tregs),and ultimately improved antitumor efficacy in a subcutaneous H22 tumor-bearing mouse model.The cryo-shocked macrophage-mediated bacterial delivery strategy holds promise for expanding the therapeutic applications of living bacteria for cancer.

Suppression of uncontrollable dendrite growth and water-induced side reactions of Zn metal anodes is crucial for achieving long-lasting cycling stability and facilitating the practical implementations of aqueous Zn-metal batteries.To address these challenges,we report in this study a functional nitro-cellulose interfacial layer(NCIL)on the surface of Zn anodes enlightened by a nitro-coordination chemistry strategy.The NCIL exhibits strong zincophilicity and superior coordination capability with Zn^(2+)due to the highly electronegative and highly nucleophilic nature of the nitro functional group.This characteristic facilitates a rapid Zn-ion desolvation process and homogeneous Zn plating,effectively preventing H_(2) evolution and dendrite formation.Additionally,the negatively charged surface of NCIL acts as a shield,repelling SO_(4)^(2-)anions and inhibiting corrosive reactions on the Zn surface.Remarkably,reversible and stable Zn plating/stripping is achieved for over 5100 h at a current density of 1 mA cm^(-2),which is nearly 30 times longer than that of bare Zn anodes.Furthermore,the Zn/V_(2)O_(5) full cells with the functional interface layer deliver a high-capacity retention of 80.3%for over 10,000 cycles at 5 A g^(-1).This research offers valuable insights for the rational development of advanced protective interface layers in order to achieve ultra-long-lifeZnmetal batteries.

Development of EPDM composites reinforced by CNTs@SiO_(2)for thermal protection systems of aerospace propulsion:Significant improvement in oxidation and ablation resistance properties

Carbon Nanotubes(CNTs)reinforced Polymer-Matrix Composites(PMCs)is widely used as insulation materials in thermal protection system of aerospace propulsion.However,CNTs are prone to oxidation and have high thermal conductivities,which makes it difficult to improve the ablation resistance of insulation materials that contain CNTs.SiO_(2)was encapsulated onto the surface of CNTs(CNTs@SiO_(2)),which were then added to Ethylene Propylene Diene Monomer(EPDM)rubber to prepare the insulation materials.Thermogravimetric analysis and ablation test were used to evaluate the resistance of the insulation materials to thermal oxidation and ablation.Additionally,scanning electron microscopy was performed to analyze their microstructures.Results revealed that the addition of CNTs@SiO_(2)could visibly reduce the effects of hot corrosion and ablation on insulation materials.The C-CNTs@SiO_(2)-1 formulation had the best ablative resistance.Further,compared with the unencapsulated formulation(C-CNTs-10),the C-CNTs@SiO_(2)-1 formulation reduced the line ablation rate by 51%to 0.0130 mm/s after oxygen-acetylene experiments.Lastly,the ablation mechanism was investigated based on the effects of the CNTs@SiO_(2)additive on their properties.Thus,the improvement in ablation performance may be attributed to CNTs@SiO_(2)-induced decreases in thermal conductivity,improvement in the hot corrosion resistance in the char layer,and changes in the microstructure.

The development of tellurium(Te)-based semiconductor nanomaterials for efficient light-to-heat conversion may offer an effective means of harvesting sunlight to address global energy concerns.However,the nanosized Te(nano-Te)materials reported to date suffer from a series of drawbacks,including limited light absorption and a lack of surface structures.Herein,we report the preparation of nano-Te by electrochemical exfoliation using an electrolyzable room-temperature ionic liquid.Anions,cations,and their corresponding electrolytic products acting as chemical scissors can precisely intercalate and functionalize bulk Te.The resulting nano-Te has high morphological entropy,rich surface functional groups,and broad light absorption.We also constructed foam hydrogels based on poly(vinyl alcohol)/nano-Te,which achieved an evaporation rate and energy efficiency of 4.11 kg m^(−2)h^(−1)and 128%,respectively,under 1 sun irradiation.Furthermore,the evaporation rate was maintained in the range 2.5-3.0 kg m^(−2)h^(−1)outdoors under 0.5-1.0 sun,providing highly efficient evaporation under low light conditions.

Engineering single-atom Mn on nitrogen-doped carbon to regulate lithium-peroxide reaction kinetics for rechargeable lithium-oxygen batteries

Precision engineering of catalytic sites to guide more favorable pathways for li_(2)O_(2) nucleation and decom-position represents an enticing kinetic strategy for mitigating overpotential,enhancing discharge capac-ity,and improving recycling stability of li-O_(2) batteries.In this work,we employ metal-organic frameworks(MOFs)derivation and ion substitution strategies to construct atomically dispersed Mn-N_(4) moieties on hierarchical porous nitrogen-doped carbon(Mn SAs-NC)with the aim of reducing the over-potential and improving the cycling stability of li-O_(2) batteries.The porous structure provides more chan-nels for mass transfer and exposes more highly active sites for electrocatalytic reactions,thus promoting the formation and decomposition of li_(2)O_(2).The li-O_(2) batteries with Mn SAs-NC cathode achieve lower overpotential,higher specific capacity(14290 mA h g^(-1) at 100 mAg^(-1)),and superior cycle stability(>100 cycles at 200 mA g^(-1))compared with the Mn NPs-NC and NC.Density functional theory(DFT)cal-culations reveal that the construction of Mn-N_(4) moiety tunes the charge distribution of the pyridinic N-rich vacancy and balances the affinity of the intermediates(liO_(2) and li_(2)O_(2)).The initial nucleation of li_(2)O_(2) on Mn SAs-NC favors the O_(2)-→liO_(2)→li_(2)O_(2) surface-adsorption pathway,which mitigates the overpoten-tials of the oxygen reduction(ORR)and oxygen evolution reaction(OER).As a result,Mn SAs-NC with Mn-N_(4) moiety effectively facilitates the li_(2)O_(2) nucleation and enables its reversible decomposition.This work establishes a methodology for constructing carbon-based electrocatalysts with high activity and selectivity for li-O_(2)batteries.

Turbo-shaft engine adaptive neural network control based on nonlinear state space equation

Intelligent Adaptive Control(AC) has remarkable advantages in the control system design of aero-engine which has strong nonlinearity and uncertainty. Inspired by the Nonlinear Autoregressive Moving Average(NARMA)-L2 adaptive control, a novel Nonlinear State Space Equation(NSSE) based Adaptive neural network Control(NSSE-AC) method is proposed for the turbo-shaft engine control system design. The proposed NSSE model is derived from a special neural network with an extra layer, and the rotor speed of the gas turbine is taken as the main state variable which makes the NSSE model be able to capture the system dynamic better than the NARMA-L2 model. A hybrid Recursive Least-Square and Levenberg-Marquardt(RLS-LM) algorithm is advanced to perform the online learning of the neural network, which further enhances both the accuracy of the NSSE model and the performance of the adaptive controller. The feedback correction is also utilized in the NSSE-AC system to eliminate the steady-state tracking error. Simulation results show that, compared with the NARMA-L2 model, the NSSE model of the turboshaft engine is more accurate. The maximum modeling error is decreased from 5.92% to 0.97%when the LM algorithm is introduced to optimize the neural network parameters. The NSSE-AC method can not only achieve a better main control loop performance than the traditional controller but also limit all the constraint parameters efficiently with quick and accurate switching responses even if component degradation exists. Thus, the effectiveness of the NSSE-AC method is validated.

Microchannel flow boiling heat transfer has the advantages of strong heat dissipation capacity,good temperature uniformity,and compact structure.It is an excellent way to thermally manage electronic devices,but when the heat flux exceeds CHF(Critical Heat Flux),the heat transfer performance deteriorates as the working fluid dries out.Non-azeotropic mixtures have the potential to effectively delay or avoid dry-out during the boiling process due to their temperature slide characteristics which causes the mass transfer resistance.To understand the influence of non-azeotropic mixtures on microchannel flow boiling,using the phase-change microchannel heat sink as the research object,the experiments on the flow boiling heat transfer performance of R245fa/R134a mixtures under different working conditions were carried out,and the characteristics of flow boiling heat transfer were obtained under the different working conditions,and comparison was developed with those of pure substance R245fa.The results demonstrated that a small amount of low-boiling-point components in the high-boiling-point working fluid inhibited boiling heat transfer to some extent,and lowered the average heat transfer coefficient under the non-dryout condition slightly lower than that of the pure substance;however,it also effectively delayed the onset of local dry-out and prevented significant deterioration in thermal transfer performance under the lower mass flow rate and higher heat flux,which could enhance the heat sink's stability.

A combination of hydrogels and microfluidics allows the construction of biomimetic three-dimensional(3D)tissue models in vitro,which are also known as organ-on-a-chipmodels.The hydrogel patterningwith awell-controlled spatial distribution is typically achieved by embedding sophisticated microstructures to act as a boundary.However,these physical barriers inevitably expose cells/tissues to a less physiologically relevant microenvironment than in vivo conditions.Herein,we present a novel dissolvable temporary barrier(DTB)strategy that allows robust and flexible hydrogel patterning with great freedom of design and desirable flow stimuli for cellular hydrogels.The key aspect of this approach is the patterning of a water-soluble rigid barrier as a guiding path for the hydrogel using stencil printing technology,followed by a barrier-free medium perfusion after the dissolution of the DTB.Single and multiple tissue compartments with different geometries can be established using either straight or curved DTB structures.The effectiveness of this strategy is further validated by generating a 3D vascular network through vasculogenesis and angiogenesis using a vascularized microtumor model.As a new proof-of-concept in vasculature-on-a-chip,DTB enables seamless contact between the hydrogel and the culture medium in closed microdevices,which is an improved protocol for the fabrication ofmultiorgan chips.Therefore,we expect it to serve as a promising paradigm for organ-on-a-chip devices for the development of tumor vascularization and drug evaluation in the future preclinical studies.

CO_(2) hydrogenation to methanol over the copper promoted In_(2)O_(3) catalyst

The metal promoted In_(2)O_(3) catalysts for CO_(2) hydrogenation to methanol have attracted wide attention because of their high activity with high methanol selectivity.However,there was still no experimental confirmation if copper could be a good promoter for In_(2)O_(3).Herein,the Cu promoted In_(2)O_(3) catalyst was prepared using a deposition-precipitation method.Such prepared Cu/In_(2)O_(3) catalyst shows significantly higher CO_(2) conversion and space time yield(STY)of methanol,compared to the un-promoted In_(2)O_(3) catalyst.The loading of Cu facilitates the activation of both H_(2) and CO_(2) with the interface between the Cu cluster and defective In_(2)O_(3) as the active site.The Cu/In_(2)O_(3) catalyst takes the CO hydrogenation pathway for methanol synthesis from CO_(2) hydrogenation.It exhibits a unique size effect on the CO adsorption.At temperatures below 250℃,CO adsorption on Cu/In_(2)O_(3) is stronger than that on In_(2)O_(3),causing higher methanol selectivity.With increasing temperatu res,the Cu catalyst aggregates,which leads to the formation of weak CO adsorption site and causes a decrease in the methanol selectivity.Compared with other metal promoted In_(2)O_(3) catalysts,it can be concluded that the catalyst with stronger CO adsorption possesses higher methanol selectivity.

Dear Editor,The Cretaceous-Paleogene(K-Pg)mass extinction precipitated one of the most profound restructurings of biodiversity in recent geological history.Despite the extinction of many iconic groups,particularly the non-avian dinosaurs,there is emerging evidence that previously overlooked taxa experienced stark morphological and evolutionary stasis.In this study,we report both adult and larva of beetles of the genus Loricera preserved in mid-Cretaceous Kachin amber(Figure 1),which display striking similarity to extant congeners,indicating a stasis of their specialized feeding behavior persisting at least 100 million years.
