A new cyclometalated iridium（Ⅲ） complex [Ir（2-pq）2（HPIP）]C1 （lrl, 2-pq=3-phenylisoquinoline, HPIP= 2-（2-hydroxyphenyl）imidazo[4,5-f] 1,10-phenanthroline） was synthesized and applied to image mitochondria in living cells. Irl displayed excellent ability to selectively accumulate in mitochondria of live cells with no requirement of replacement of the culture medium. Due to Irl exhibiting better photostability than the commercially available mitochondrial trackers, this complex could be applicable for the imaging and tracking of the mitochondrial mor- phological changes over long periods of time. In addition, in vitro cytotoxicity investigation revealed that it! showed negligible cytotoxicity at the concentrations employed. Based on these, Irl is suitable for organelle-selective imaging in living cells.

Light springs(LSs) have played essential roles in particle rotation and manipulation, optical super-resolution imaging, and optical information coding. In related research areas, it is important to accurately measure spatiotemporal information on LSs to understand and analyze their applications. However, there is no experimental method that can accurately detect the drastic spatial evolution of ultrafast LSs to date. Therefore, in this study, we propose a compressed ultrafast photography(CUP) technique to observe LSs in spatial and temporal dimensions with a snapshot. Using our home-built CUP system, we successfully capture spatiotemporal information on picosecond LSs with two and four petals, involving spatial structure and rotation velocity;furthermore, the experimental measurements are in good agreement with theoretical simulations. This study provides a novel visualization method for specifically measuring the spatial structure and temporal evolution of LSs, thus establishing a new idea for accurately characterizing spatiotemporal information on complex ultrafast laser fields.

DNA molecules were stretched on silanized mica surface with the molecular combing technique, and detected with fluorescence microscopy and atomic force microscopy. Meantime, DNA molecules were stretched with a modified dynamic molecular combing technique and studied with atomic force microscopy. The results indicate that, compared with the dynamic molecular combing technique, the modified dynamic molecular combing technique has advantages of less-sample demand and less contamination to sample; as compared with the molecular combing technique, it has better aligning effect and reproducibility. Combination of this kind of DNA molecular manipulating technique with the single DNA molecule detecting technique by atomic force microscopy and fluorescence microscopy will play an important role in the basic research of molecular dynamics and the application of gene research.

Structured illumination microscopy (SIM) has been widely applied to investigate intricate biological dynamics due to its outstanding super-resolution imaging speed. Incorporating compressive sensing into SIM brings the possibility to further improve the super-resolution imaging speed. Nevertheless, the recovery of the superresolution information from the compressed measurement remains challenging in experiments. Here, we report structured illumination microscopy with complementary encoding-based compressive imaging (CECI-SIM) to realize faster super-resolution imaging. Compared to the nine measurements to obtain a super-resolution image in a conventional SIM, CECI-SIM can achieve a super-resolution image by three measurements;therefore, a threefold improvement in the imaging speed can be achieved. This faster imaging ability in CECI-SIM is experimentally verified by observing tubulin and actin in mouse embryonic fibroblast cells. This work provides a feasible solution for high-speed super-resolution imaging, which would bring significant applications in biomedical research.

松辽盆地北部中央坳陷区大庆长垣、齐家一古龙凹陷和三肇凹陷扶余油层和高台子油层发育致密砂岩油,资源潜力超过10×108t.勘探实践表明,这种致密油层呈现纵向不集中、横向不连续、甜点单体规模小、物性变化大、含油性差异大等非均质特征.针对这些非均质性,按照“逆向思维、反向设计、正向实施”理念,创建了“2＋3”模式,即通过工程地质“一体化”设计引领,依托低成本高效率“工厂化”实施,带来勘探开发“配套工程技术”、水平井“单井高产量”和致密油“整体有效益”.经过2011年以来的探索实践,形成了以甜点精细刻画及水平井轨迹精细控制技术和增产改造设计优化技术为核心的工程地质“一体化设计”技术,建立了差异型和灵活型2种低成本高效率个性化“工厂化”实施模式,完善了“预测甜点目标、钻成优质井眼、评价甜点层段、提供工程参数和提高单井产量”5项致密油勘探开发“配套工程技术”,实现了致密油水平井压裂后产油是周围直井17倍以上的“单井高产量”,展现了致密油“整体有效益”初步成效,建成3个试验区,正建10个试验区,新增致密油储量超过1×108t.“2＋3”模式的建立,不仅为大庆油田原油稳产增加了资源,而且为其后规模生产储备了配套工程技术。

Euglena gracilis 277, a unicellular green alga, demonstrated remarkable ability to transport Nd3+ to the cell compartments. For a given amount of Nd3+ and cells, the results of ICP-AES indicated that the cellular uptake of Nd3+ was independent of Nd3+ concentration in thebulk solution. The average uptake of Nd3+ per celi (mMd) is proportional to a parameter ξ-theratio of neodymium content to the celi counts of the system. A novel approach for probing cellular neodymium by tetraiodotetra chlorofluorescein (I4TCF) has been devised. Data derived from the cryosections of !4TCF-Nd3+ stained cells and EDAX of the fast freezing ultrathin cryosections indicate that Nd3+ is distributed over the celi compartments. Chloroplasts are the major compartments as the residence of Nd3+ in the alga. The transport should be against a concentration gradient of Nd3+ on the order of five, even higher. it is proposed that a calcium ion channel would play an important role in the Nd3+ transportation.

Various super-resolution microscopy techniques have been presented to explore fine structures of biological ***,the super-resolution capability is often achieved at the expense of reducing imaging speed by either point scanning or multiframe *** contradiction between spatial resolution and imaging speed seriously hampers the observation of high-speed dynamics of fine *** overcome this contradiction,here we propose and demonstrate a temporal compressive super-resolution microscopy(TCSRM)*** technique is to merge an enhanced temporal compressive microscopy and a deep-learning-based super-resolution image reconstruction,where the enhanced temporal compressive microscopy is utilized to improve the imaging speed,and the deep-learning-based super-resolution image reconstruction is used to realize the resolution *** high-speed super-resolution imaging ability of TCSRM with a frame rate of 1200 frames per second(fps)and spatial resolution of 100 nm is experimentally demonstrated by capturing the flowing fluorescent beads in microfluidic *** the outstanding imaging performance with high-speed super-resolution,TCSRM provides a desired tool for the studies of high-speed dynamical behaviors in fine structures,especially in the biomedical field.
