Dear Editor,Since the first recognized case of human infection with avian influenza H5NI virus in 1997, the worldwide spread and re-emergence of this highly pathogenic influenza virus have led to 694 human infection cases. Of these cases, 402 died from 2003 to 2015 （http：//***/influenza/ human animal_interface/EN_GIP_20150106Cumulative- ***？ua=l）, with a morbidity rate of 57.9%, which is much higher than that of seasonal influenza H1N1 virus.

Influenza is a persistent threat to human health and there is a continuing requirement for updating antiinfluenza strategies. Initiated by observations of different endoplasmic reticulum(ER) responses of host to seasonal H1N1 and highly pathogenic avian influenza(HPAI) A H5N1 infections, we identified an alternative antiviral role of tauroursodeoxycholic acid(TUDCA), a clinically available ER stress inhibitor, both in vitro and in vivo. Rather than modulating ER stress in host cells, TUDCA abolished the proton conductivity of viral M2 by disrupting its oligomeric states, which induces inefficient viral infection. We also showed that M2 penetrated cells, whose intracellular uptake depended on its proton channel activity,an effect observed in both TUDCA and M2 inhibitor amantadine. The identification and application of TUDCA as an inhibitor of M2 proton channel will expand our understanding of IAV biology and complement current anti-IAV arsenals.

Deep vein thrombosis(DVT)is a common complication following traumatic fracture with a 0.5%–1%annual *** molecular weight heparin(LMWH)is the most commonly used anticoagulation drug for DVT prevention,but treatment with LMWH is *** aim is to compare the antithrombotic effect of dragon’s blood,an oral botanical anticoagulant medicine approved by the Chinese FDA,with LMWH in patients undergoing hip fracture surgery and to explore the molecular mechanisms of anticoagulation *** study recruited patients and divided them into LMWH and dragon’s blood treatment *** index tests,Doppler ultrasound and mRNA sequencing were performed before and after anticoagulation *** was no significant difference in postoperative DVT incidence between the two groups(23.1%versus 15.4%,P=0.694).D-dimer(D-D)and fibrinogen degradation product(FDP)showed significant reductions in both groups after anticoagulation *** identified SLC4A1,PROS1,PRKAR2B and seven other genes as being differentially expressed during anticoagulation therapy in both *** correlated with coagulation indexes were also ***’s blood and LMWH showed similar effects on DVT and produced similar gene expression changes in patients undergoing hip fracture surgery,indicating that dragon’s blood is a more convenient antithrombosis medicine(oral)than LMWH(hypodermic injection).

Influenza A virus H5N1 presents a major threat to human health. The entry of influenza virus into host cells is believed to be mediated by hemagglutinin (HA), a virus surface glycoprotein that can bind terminal sialic acid residues on host cell glycoproteins and glycolipids. In this study, we elucidated the pathways through which H5N1 enters human lung carcinoma cell line A549. We first proved that H5N1 can enter A549 cells via endocytosis, as lysosomotropic agents, such as bafilomycin A1 and chloroquine, can rescue H5N1-induced A549 cell death. By using specific inhibitors, and siRNAs that target the clathrin pathway, we further found that H5N1 could enter A549 cells via clathrin-mediated endocytosis, while inhibitors targeting caveolae-mediated endocytosis could not inhibit H5N1 cell entry. These findings expand our understanding of H5N1 pathogenesis and provide new information for anti-viral drug research.

Avian influenza A virus constitutes a large threat to human health. Recent outbreaks of highly pathogenic avian influenza H5N1 virus in poultry and in humans have raised concerns that an influenza pandemic will occur in the near future. Transmission from avian species to humans remains sporadic, but the mortality associated with human infection is very high (about 62%). To date, there are no effective therapeutic drugs or a prophylactic vaccines available, which means that there is still a long way to go before we can eradicate or cure avian influenza. This review focuses on the molecular pathogenesis of avian influenza H5N1 virus infection. An understanding of the viral pathogenesis may facilitate the development of novel treatments or effective eradication of this fatal disease.
