重症急性呼吸综合征 (severeacuterespiratorysyndrome ,SARS) ,临床表现为非典型肺炎。2 0 0 2年底我国广东发现此种当时不明病因的疾病 ,此后在越南、中国香港、加拿大、美国等三十多个国家和地区也相继发现类似病例。由于SARS具有很高的传染性 ,死亡率也高 ,各国和世界卫生组织 (WHO)对此病高度重视。WHO迅速建立起由全球 10个国家的 13个实验室组成的协作研究和监测网络。一种新型的冠状病毒 (coronavirus)被认为极可能是SARS的病原体 ,因为它满足了 6个科赫要点 (Koch’spostulates)。2 0 0 3年 4月 12日 ,加拿大科学家首次公布了SARS病毒Tor2的基因组序列。4月 16日 ,WHO正式确认SARS病毒是SARS的病原体。目前我国SARS的病情仍然十分严峻 ,现就SARS病毒及相关冠状病毒的生物学特征和可能的药物防治靶点作一综述。

芋螺毒素是近年来国际上的一个研究热点 .它属于一类海洋生物活性多肽 ,多数由 12～ 4 6个氨基酸残基所组成 ,能特异性地作用于体内各种离子通道和细胞膜上神经递质和激肽的受体 .具有分子质量小、结构多样、作用靶点广泛、功能专一、组织特异性强等优点 ,因而较之其他生物来源的活性多肽有更多的优越性 .芋螺毒素是一待挖掘的“富矿区” ,可作为体内一些具有重要生理功能靶点的探针和“解密器” ,亦可作为新药的先导化合物或直接开发成新药。

Aim:The aim of the present study was to further improve the therapeutic effectsfor human hepatocellular carcinoma (HCC) and reduce the damage in normal cellsusing a novel chemo-gene-virotherapeutic ***:An oncolyticadenoviral vector (ZD55) similar to the typical oncolytic adenovirus ONYX-015,with a deletion of ElB-55K gene,was employed to express the second mitochon-dria-derived activator of caspases (Smac) protein by constructing a *** *** enhanced cytotoxicity of the combined treatment of ZD55-Smac with cisplatin or 5-fluorouracil (5-FU) was evaluated in several HCC ***,the negative effects on normal cells have been tested in humannormal liver cell lines L-02 and QSG-7701 cell lines by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and apoptotic cell ***:According to our observation,ZD55-Smac is superior to ONYX-015 in sensitizingchemotherapy,ZD55-Smac used in conjunction with chemotherapy was found toexhibit obviously enhanced cytotoxicity in HCC cells,yet significantly abolishedthe negative toxicity in normal cells by utilizing the tumor selective replicationvector and reducing the ***:This chemo-gene-virotherapeutic(cisplatin or 5-FU+ZD55-Smac) strategy is superior to the conventional chemo-gene or chemo-viro approach.

Interleukin-33(IL-33)is a crucial nuclear cytokine that induces the type 2 immune response and maintains immune *** fine-tuned regulation of IL-33 in tissue cells is critical to control of the type 2 immune response in airway inflammation,but the mechanism is still ***,we found that healthy individuals had higher phosphate-pyridoxal(PLP,an active form of vitamin B6)concentrations in the serum than asthma *** serum PLP concentrations in asthma patients were strongly associated with worse lung function and *** a mouse model of lung inflammation,we revealed that PLP alleviated the type 2 immune response and that this inhibitory effect relied on the activity of IL-33.A mechanistic study showed that in vivo,pyridoxal(PL)needed to be converted into PLP,which inhibited the type 2 response by regulating IL-33 *** mice heterozygous for pyridoxal kinase(PDXK),the conversion of PL to PLP was limited,and IL-33 levels were increased in the lungs,aggravating type 2 ***,we found that the mouse double minute 2 homolog(MDM2)protein,an E3 ubiquitin-protein ligase,could ubiquitinate the N-terminus of IL-33 and sustain IL-33 stability in epithelial *** reduced MDM2-mediated IL-33 polyubiquitination and decreased the level of IL-33 through the proteasome *** addition,inhalation of PLP alleviated asthma-related effects in mouse *** summary,our data indicate that vitamin B6 regulates MDM2-mediated IL-33 stability to constrain the type 2 response,which might help develop a potential preventive and therapeutic agent for allergy-related diseases.

Interferon-γ （IFN-γ） triggers macrophage for inflammation response by activating the intracellular JAK-STAT1 signaling. Suppressor of cytokine signaling 1 （SOCS1） and protein tyrosine phosphatases can negatively modulate IFN-γ signaling. Here, we identify a novel negative feedback loop mediated by STAT3-SOCS3, which is tightly controlled by SENP1 via de-SUMOylation of protein tyrosine phosphatase 1B （PTPIB）, in IFN-y signaling. SENP1-deficient macrophages show defects in IFN-γ signaling and M1 macrophage activation. PTP1B in SENP1-deficient macrophages is highly SUMOylated, which reduces PTP1B-induced de-phosphorylation of STAT3. Activated STAT3 then suppresses STAT1 activation via SOCS3 induction in SENP1-deficient macro- phages. Accordingly, SENP1-deficient macrophages show reduced ability to resist Listerio rnonocytogenes infection. These results reveal a crucial role of SENP1-controlled STAT1 and STAT3 balance in rnacrophage polarization.

Monkeypox was declared a global health emergency by the World Health Organization,and as of March 2023,86,000 confirmed cases and 111 deaths across 110 countries have been *** causal agent,monkeypox virus(MPV)belongs to a large family of double-stranded DNA viruses,Orthopoxviridae,that also includes vaccinia virus(VACV)and *** produces two distinct forms of viral particles during its replication cycles:the enveloped viron(EV)that is released via exocytosis,and the mature viron(MV)that is discharged through lysis of host *** study was designed to develop multi-valent m RNA vaccines against monkeypox EV and MV surface proteins,and examine their efficacy and mechanism of *** m RNA vaccines were produced with different combinations of surface proteins from EV(A35R and B6R),MV(A29L,E8L,H3L and M1R),or EV and MV,and were administered in Balb/c mice to assess their immunogenicity potentials.A dynamic immune response was observed as soon as seven days after initial immunization,while a strong Ig G response to all immunogens was detected with ELISA after two *** higher number of immunogens contributed to a more robust total Ig G response and correlating neutralizing activity against VACV,indicating the additive potential of each immunogen in generating immune response and nullifying VACV ***,the m RNA vaccines elicited an antigen-specific CD4^(+)T cell response that is biased towards *** m RNA vaccines with different combinations of EVand MV surface antigens protected a mouse model from a lethal dose VACV challenge,with the EV and MV antigens-combined vaccine offering the strongest *** findings provide insight into the protective mechanism of multi-valent m RNAvaccines against MPV,and also the foundation for further development of effective and safe m RNA vaccines for enhanced protection against monkeypox virus outbreak.
