Xuebijing improves intestinal microcirculation dysfunction in septic rats by regulating the VEGF-A/PI3K/Akt signaling pathway

BACKGROUND:This study aims to explore whether Xuebijing(XBJ) can improve intestinal microcirculation dysfunction in sepsis and its mechanism.METHODS:A rat model of sepsis was established by cecal ligation and puncture(CLP).A total of 30 male SD rats were divided into four groups:sham group,CLP group,XBJ + axitinib group,and XBJ group.XBJ was intraperitoneally injected 2 h before CLP.Hemodynamic data(blood pressure and heart rate) were recorded.The intestinal microcirculation data of the rats were analyzed via microcirculation imaging.Enzyme-linked immunosorbent assay(ELISA) kits were used to detect the serum levels of interleukin-6(IL-6),C-reactive protein(CRP),and tumor necrosis factor-α(TNF-α) in the rats.Histological analysis and transmission electron microscopy were used to analyze the injury of small intestinal microvascular endothelial cells and small intestinal mucosa in rats.The expression of vascular endothelial growth factor A(VEGF-A),phosphoinositide 3-kinase(PI3K),phosphorylated PI3K(p-PI3K),protein kinase B(Akt),and phosphorylated Akt(p-Akt) in the small intestine was analyzed via Western blotting.RESULTS:XBJ improved intestinal microcirculation dysfunction in septic rats,alleviated the injury of small intestinal microvascular endothelial cells and small intestinal mucosa,and reduced the systemic inflammatory response.Moreover,XBJ upregulated the expression of VEGF-A,p-PI3K/total PI3K,and p-Akt/total Akt in the rat small intestine.CONCLUSION:XBJ may improve intestinal microcirculation dysfunction in septic rats possibly through the VEGF-A/PI3K/Akt signaling pathway.

A visible-light-promoted formal[2+2+1]cyclization of N-aryl glycines with quinoxalin-2(1H)-ones to synthesize tetrahydroimidazo[1,5-a]quinoxalin-4(5H)-ones have been developed.The protocol features operational simplicity,mild reaction conditions with blue LED light employing Ru(bpy)3Cl2.6H2O as a photoredox catalyst,a combination of O2 from air and Cu(OAc)2 as the oxidant and broad substrate applicability.

Semiannual oscillation,annual oscillation,quasibiennial oscillation,and solar cycle variation of the OH airglow emission in the mesopause region

The vertically integrated emission rate,centroid altitude,peak emission rate,and peak height of the hydroxyl(OH)airglow were calculated from Thermosphere Ionosphere Mesosphere Energetics and Dynamics(TIMED)/Sounding of the Atmosphere using Broadband Emission Radiometry(SABER)observations to study the seasonal and interannual variations in the intensity and location of the OH emission.The emission rate is inversely proportional to the height of the emission,with the semiannual oscillation dominating at low latitudes and the annual oscillation dominating at higher latitudes.The OH emission is modulated by the quasibiennial oscillation at the equator,and the quasibiennial oscillation signal is weak at other latitudes.We represented the vertical transport of atomic oxygen by using atomic oxygen concentrations obtained from a global atmospheric model,the Specified Dynamics Whole Atmosphere Community Climate Model with thermosphere and ionosphere eXtension simulations.Compared with the amplitudes of the migrating diurnal tide(DW1)calculated from temperature data observed by TIMED/SABER,we found that both the vertical transport of atomic oxygen and DW1 amplitudes in the equatorial region exhibit semiannual oscillation and quasibiennial oscillation,which have a strong correlation with the variations in the amplitude and phase of semiannual oscillation and quasibiennial oscillation in OH emission.It is likely that the DW1 affects the vertical transport of atomic oxygen that is involved in the reaction to produce O3,thus affecting the OH emission.We analyzed the relationship between OH emission and solar activity by using the solar radio flux at 10.7 cm as a proxy for solar activity.The results showed that the OH emission is well correlated with solar activity,and the modulation of OH emission by solar activity has a significant latitudinal variation.The small correlation between emission height and solar activity indicates that solar activity modulates OH emission mainly through chemical rather than dynamic processes.

Understanding the rapid spread of antimicrobial resistance genes mediated by Is26

Insertion sequences(ISs)promote the transmission of antimicrobial resistance genes(ARGs)across bacterial populations.However,their contributions and dynamics during the transmission of resistance remain unclear.In this study,we selected is26 as a representative transposable element to decipher the relationship between ISs and ARGs and to investigate their transfer features and transmission trends.We retrieved 2656 translocatable IS26-bounded units with ARGs(tiS26-bUs-ARGs)in complete bacterial genomes from the NCBI RefSeq database.In total,124 ARGs spanning 12 classes of antibiotics were detected,and the average contribution rate of IS26 to these genes was 41.2%.We found that IS26-bounded units(IS26-bUs)mediated extensive ARG dissemination within the bacteria of the Gammaproteobacteria class,showing strong transfer po-tential between strains,species,and even phyla.The Is26-bUs expanded in bacterial populations over time,and their temporal expansion trend was significantly correlated with antibiotic usage.This wide dissemination could be due to the nonspecific target site preference of is26.Finally,we experimentally confirmed that the introduction of a single copy of is26 could lead to the formation of a composite transposon mediating the transmission of“passenger”genes.These observations extend our knowledge of the Is26 and provide new insights into the mediating role of ISs in the dissemination of antibiotic resistance.

Females adopt sexual catalepsy to facilitate mating

Theory predicts that males and females of dioecious species typically engage in an evolutionary sexual conflict over the frequency and choice of mating partner.Female sexual cannibalism,a particularly dramatic illustration of this conflict,is widespread in certain animal taxa including spiders.Nevertheless,females of some funnel weaving spiders that are generally aggressive to conspecifics enter a cataleptic state after male courtship,ensuring the males can mate without risk of attack.In this study,we demonstrated that the physical posture and duration,metabo-lites,and central neurotransmitters of females of Aterigena aculeata in sexual catalepsy closely resemble females in thanatosis but are distinct from those in anesthesia,indicating that the courted females feign death to eliminate the risk of potentially aggressive responses and thereby allow preferred males to mate.Unlike the taxonomically widespread thanatosis,which generally represents a deceptive visual signal that acts against the interest of the receivers,sexual catalepsy of females in the funnel weaving spiders may deliver a sexual-receptive signal to the courting males and thereby benefit both the signal senders and receivers.Therefore,sexual catalepsy in A.aculeata may not reflect a conflict butrather a confluence of interest betweenthe sexes.

Influence of pore structure on thermal stress distribution inside coal particles during primary fragmentation

Thermal stress is an important reason of coal particle primary fragmentation,during which the role of pore structure is ambiguous.Thermal stress induced fragmentation experiments were conducted with low volatile coal/char particles,and the results show that the fragmentation severity enhances with increasing porosity.Various porous thermal stress models were developed with finite element method,and the influences of the pore shape,size,position and porosity on the thermal stress were discussed.The maximum thermal stress inside particle increases with pore curvature,the pore position affects the thermal stress more significantly at the particle center and surface.The expectation of the maximum tensile thermal stress linearly increases with porosity,making the particles with higher porosity easier to fragment,contrary to the conclusion deduced from the devolatilization theory.The obtained results are valuable for the analysis of different thermal processes concerning the thermal stresses of the solid feedstocks.

Propagation form of internal cracks induced by continuous casting soft reduction and control strategy for internal quality

The propagation form of internal cracks induced by continuous casting soft reduction and the control strategy for enhancing the internal quality of 45 steel through industrial trials and a three-dimensional flow-heat transfer-solidification coupling model were investigated.The results showed that the internal cracks induced by soft reduction exhibited a characteristic of being"coarse in the middle and fine at both ends",and displayed an elliptical arc distribution on the loose side of the strand cross section.The cracks originated within the brittle temperature range and propagated inward to the liquid impenetrable temperature and outward to the zero ductility temperature or below.The control strategy for enhancing the internal quality of the 45 steel strand through soft reduction is to adjust the casting speed or the reduction zone appropriately,ensuring that the central solid fraction of the reduction zone falls within the range of 0.33-0.99.At this point,a reasonable reduction amount is allocated to eliminate the center shrinkage cavities and center segregation,even if it results in minor reduction-induced cracks.

Harnessing sub-comb dynamics in a graphene-sensitized microresonator for gas detection

Since their inception, frequency combs generated in microresonators, known as microcombs, have sparked significant scientific interests. Among the various applications leveraging microcombs, soliton microcombs are often preferred due to their inherent mode-locking capability. However, this choice introduces additional system complexity because an initialization process is required. Meanwhile, despite the theoretical understanding of the dynamics of other comb states, their practical potential, particularly in applications like sensing where simplicity is valued, remains largely untapped. Here, we demonstrate controllable generation of sub-combs that bypasses the need for accessing bistable regime. And in a graphene-sensitized microresonator, the sub-comb heterodynes produce stable, accurate microwave signals for high-precision gas detection. By exploring the formation dynamics of sub-combs, we achieved 2 MHz harmonic comb-to-comb beat notes with a signal-to-noise ratio (SNR) greater than 50 dB and phase noise as low as – 82 dBc/Hz at 1 MHz offset. The graphene sensitization on the intracavity probes results in exceptional frequency responsiveness to the adsorption of gas molecules on the graphene of microcavity surface, enabling detect limits down to the parts per billion (ppb) level. This synergy between graphene and sub-comb formation dynamics in a microcavity structure showcases the feasibility of utilizing microcombs in an incoherent state prior to soliton locking. It may mark a significant step toward the development of easy-to-operate, systemically simple, compact, and high-performance photonic sensors.

Arbuscular mycorrhizal fungi inoculation and exogenous indole-3-acetic acid application induce antioxidant defense response to alleviate cadmium toxicity in Broussonetia papyrifera

Cadmium(Cd)contamination in soil poses a huge threat to plants even at low concentrations;Broussonetia papyrifera has great potential in remediation of soil heavy metal contamination.However,whether exogenous indole-3-acetic acid(IAA)application and arbuscular mycorrhizal fungi(AMF)have synergistic effects on Cd tolerance of B.papyrifera remains unclear.To investigate the effects of AMF inoculation and IAA application on the tolerance of B.papyrifera to Cd stress,two experiments were conducted:the first to investigate the effect of AMF(Rhizophagus irregularis)inoculation on the tolerance of B.papyrifera to Cd stress and the second to investigate the combined effects of AMF inoculation and IAA application on the tolerance of B.papyrifera to Cd stress.Parameters including endogenous hormone concentration,antioxidant defense response,malondialdehyde(MDA)content,and gene expression related to antioxidant enzyme system and hormone were measured.The results indicated that AMF alleviated Cd toxicity of B.papyrifera by reducing MDA content and improving antioxidant enzyme activities and Cd absorption capacity.Furthermore,the combination of AMF inoculation and IAA application had a synergetic effect on the tolerance of B.papyrifera to Cd stress through upregulating BpAUX1 and BpAUX2,which might contribute to root growth and root xylem synthesis,and by upregulating BpSOD2 and BpPOD34 to enhance the antioxidant enzyme system.This work provides a new insight into the application of IAA in the remediation of soil Cd pollution by mycorrhizal plants.

The underappreciated role of fugitive VOCs in ozone formation and health risk assessment emitted from seven typical industries in China

Fugitive emission from industrial sources may result in ozone formation and health risk,while the exact contribution of this source remains incompletely understood.In this study,emission characteristics,ozone formation potential(OFP)and health risk of fugitive VOCs in7 representative industries were investigated.Chemical material industry was the dominant contributor to VOCs of fugitive emission in comparison with other industries.The OFP of VOCs from fugitive emission was in the range of 1.45×10^(3)-3.98×10^(5)μg/m^(3),with a higher value than that of organized emission in seven industries except for the coking industry and the chemical material industry,suggesting that fugitive VOCs should be taken into account while developing control strategies.Acetaldehyde,m,p-xylene,n-nonane,ethylene,vinyl chloridethe and other high OFP-contributing species were the major reactive species that should be targeted.Health risk assessment investigated non-cancer and cancer risks of fugitive VOCs in 7 industries were all above safe level(HR>1 and LCR>1×10^(-4)),posing remarkable health threats to human health.OVOCs were the main contributor to non-cancer risk,while halohydrocarbons and aromatics contributed most to cancer risks,posing remarkable health threat on human health.Our findings highlighted the contribution of fugitive VOCs on ozone formation and health risk was underestimated,indicating which should be considered in emission control strategies of industrial sources.
