Seasonal Variations of Mesospheric Densities Observed by Rayleigh Lidar at Golmud,Qinghai

From Aug.2013 to Oct.2015,a Rayleigh lidar has been used to study the middle atmosphere at Golmud(36.25°N,94.54°E),Qinghai,located in the northeastern part of the Tibetan Plateau.Mesospheric density profiles from 50 to 90 km were retrieved based on 205 nights of lidar observation,with a total of 1616hours of operation.We compared our lidar density measurements with SABER observations onboard TIMED satellite and MSIS-00 model data.The results showed that the annual mean density measured by lidar agreed well with SABER data,but both were lower than that of MSIS-00.All datasets exhibited dominant annual oscillation in the mesosphere.From 63 to 85 km,the annual amplitude of lidar density is larger than those of SABER and MSIS-00.PDD(Percentage of Density Difference)was calculated to investigate the mesospheric density climatology.The largest density variations of lidar,MSIS-00,and SABER occurred at around 72 km.Both lidar and SABER PDD reached their maximum in May,about one month earlier than the MSIS-00;while the minimum PDD appeared in late December for all datasets.

Proteins and peptides have become a significant therapeutic modality for various diseases because of their high potency and specificity.However,the inherent properties of these drugs,such as large molecular weight,poor stability,and conformational flexibility,make them difficult to be formulated and delivered.Injection is the primary route for clinical administration of protein and peptide drugs,which usually leads to poor patient’s compliance.As a portable,minimally invasive device,microneedles(MNs)can overcome the skin barrier and generate reversible microchannels for effective macromolecule permeation.In this review,we highlighted the recent advances in MNs-mediated transdermal delivery of protein and peptide drugs.Emphasis was given to the latest development in representative MNs design and fabrication.We also summarize the current application status of MNs-mediated transdermal protein and peptide delivery,especially in the field of infectious disease,diabetes,cancer,and other disease therapy.Finally,the current status of clinical translation and a perspective on future development are also provided.

Methylation profile of bovine Oct4 gene coding region in relation to three germ layers

Previous studies have shown that octamer-binding transcription factor 4（Oct4） plays a significant role in early embryonic development of mammalian animals, and different Oct4 expression levels induce multi-lineage differentiation which are regulated by DNA methylation. To explore the relationship between the methylation pattern of Oct4 gene exon 1 and embryonic development, in this work, five different tissues（heart, liver, lung, cerebrum and cerebellum） from three germ layers were chosen from low age（50–60 d） and advanced age（60–70 d） of fetal cattle and the differences between tissues or ages were analyzed, respectively. The result showed that the DNA methylation level of Oct4 gene exon 1 was significant different（P〈0.01） between any two of three germ layers in low age（〈60 d）, but kept steady of advanced age（P〉0.05）（〉60 d）, suggesting that 60-d post coital was an important boundary for embryonic development. In addition, in ectoderm（cerebrum and cerebellum）, there was no significant methylation difference of Oct4 gene exon 1 between low age and advanced age（P〉0.05）, but the result of endoderm（liver and lung） and mesoderm（heart） were on the contrary（P〈0.01）, which indicated the development of ectoderm was earlier than endoderm and mesoderm. The methylation differences from the 3rd, 5th and 9th Cp G-dinucleotide loci of Oct4 gene exon 1 were significantly different between each two of three germ layers（P〈0.05）, indicating that these three loci may have important influence on bovine embryonic development. This study showed that bovine germ layers differentiation was significantly related to the DNA methylation status of Oct4 gene exon 1. This work firstly identified the DNA methylation profile of bovine Oct4 gene exon 1 and its association with germ layers development in fetus and adult of cattle. Moreover, the work also provided epigenetic information for further studying bovine embryonic development and cellular reprogramming.

Extensive usage of highly conductive carbon materials with large specific surface area(e.g.,carbon nanotubes,CNTs)in lithium ion batteries(LIBs),especially as current collector of anodes,suffers from low initial coulombic efficiency(ICE),large interfacial resistance,and severe embrittlement,as the large specific surface area often results in severe interfacial decomposition of the electrolyte and the formation of thick and fluffy solid electrolyte interphase(SEI)during cycling of LIBs.Herein,we demonstrate that when the CNT-based current collector and Na foil(which are being stacked intimately upon each other)are being placed in Na+-based organic electrolyte,local redox reaction between the Na foil and the electrolyte would occur spontaneously,generating a thin and homogeneous NaF-based passivating layer on the CNTs.More importantly,we found that owing to the weak solvation behaviors of Na+in the organic electrolyte,the resulting passivation layer,which is rich in NaF,is thin and dense;when used as the anode current collector in LIBs,the pre-existing passivating layer can function effectively in isolating the anode from the solvated Li+,thus suppressing the formation of bulky SEI and the destructive intercalation of solvated Li+.The relevant half-cell(graphite as anode)exhibits a high ICE of 92.1%;the relevant pouch cell with thus passivated CNT film as current collectors for both electrodes(LiCoO_(2)as cathode,graphite as anode)displays a high energy density of 255 Wh kg^(-1),spelling an increase of 50%compared with that using the conventional metal current collectors.

OBJECTIVE: To investigate the efficacy of Yuzhizi(Fructus Akebiae Quinatae) seed extract(FAQSE) on inhibiting the proliferation of hepatocellular carcinoma(HCC) cells in vitro and to explore the anti-HCC action mechanism of FAQSE. METHODS: Human HCC Hep G2 and Huh7 cells were used to investigate the anti-HCC effect of FAQSE in vitro. 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) method was used to measure cell viability. Affymetrix microarray was adopted to detect the expression of transcriptome. The differentially expressed genes(DEGs) of each cell line were identified. For coDEGs of both cell lines, the gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway were enriched using the Database for Annotation, Visualization and Integrated Discovery(DAVID), and the network analysis of protein-protein interaction(PPI) was mapped using the Retrieval of Interacting Genes/Proteins(STRING) and Cytoscape software. Some important genes in the PPI network of coDEGs were selected to verify by quantitative real-time reverse transcription-polymerase chain reaction, Western blot and enzyme-linked immunosorbent assay. RESULTS: FAQSE decreased the viability of Hep G2 and Huh7 cells. There were 211 co-upregulated and 86 codownregualted genes in both cell lines after FAQSE treatment. The enriched GO terms of co-upregulated DEGs were primarily involved cell-cell adhesion, viral process, transcription initiation from RNA polymerase II promoter, positive regulation of transcription from RNA polymerase II promoter and actin cytoskeleton organization. The GO terms of co-downregulated DEGs were mainly enriched in the processes of SRP-dependent cotranslational protein targeting to membrane, viral transcription, nuclear-transcribed m RNA catabolic process, nonsense-mediated decay, translational initiation and r RNA processing. Gastric cancer is the fifth most common cancer and the third leading cause of cancer death worldwide,posing a severe threat to human health.Surgical resection remains the most preferred option for gastric cancer treatment.However,for advanced gastric cancer,the curative effect of surgical resection is usually limited by the local recurrence,peritoneal carcinomatosis,or distal metastasis.Intraoperative chemotherapy is an attractive in situ adjuvant treatment strategy to reduce the recurrence and metastasis after surgical resection.Here,we designed a 5-fluorouracil(5-FU)and cis-platinum(DDP)co-delivery system based on a biodegradable temperature-sensitive hydrogel(PDLLA-PEG-PDLLA,PLEL)for intraoperative adjuvant combination chemotherapy of gastric cancer.This 5-FU+DDP/PLEL hydrogel system characterized by a special sol-gel phase transition in response to physiological temperature and presented sustained drug release in vitro and in vivo.A strong synergistic cell proliferation inhibition and apoptosis promotion of 5-FU+DDP/PLEL were observed against gastric cancer MKN45-luc cells.After intraperitoneal injection,the dual-drug loaded hydrogel formulation showed superior anti-tumor effects than the single-drug carrying hydrogels and combination of free 5-FU and DDP on the gastric cancer peritoneal carcinomatosis model.The use of hydrogel for dual-drug delivery had benefited to fewer side effects as well.What’s more,we established a mouse model for postsurgical residual tumors and peritoneal carcinomatosis of gastric cancer,in which the intraoperative administration of 5-FU+DDP/PLEL also remarkably inhibited the local recurrence of the orthotopic tumors and the growth of the abdominal metastatic tumors,resulting in an extended lifetime.Hence,this developed dual-drug loaded hydrogel system has great potential in the intraoperative chemotherapy of gastric cancer,that suggests a clinically-relevant and valuable option for postsurgical management of gastric cancer.

A review on application of dielectric barrier discharge plasma technology on the abatement of volatile organic compounds

Volatile organic compounds(VOCs)released from the waste treatment facilities have become a significant issue because they are not only causing odor nuisance but may also hazard to human health.Non-thermal plasma(NTP)technologies are newly developed methods and became a research trend in recent years regarding the removal of VOCs from the air stream.Due to its unique characteristics,such as rapid response at room temperature,bulk homogenized volume,high reaction efficiency,dielectric barrier discharge(DBD)plasma technology is considered one of the most promising techniques of NTP.This paper reviews recent progress of DBD plasma technology for abatement of VOCs.The principle of plasma generation in DBD and its configurations(electrode,discharge gap,dielectric barrier material,etc.)are discussed in details.Based on previously published literature,attention has been paid on the effect of DBD configuration on the removal of VOCs.Effect of various process parameters such as initial concentration,gas feeding rate,oxygen content and input power on VOCs removal are also considered.Moreover,the role of catalysis and inhibitors in VOCs removal by DBD system are presented.Finally,a modified configuration of the DBD reactor,i.***.double dielectric barrier discharge(DDBD)for the abatement of VOCs is discussed.It was suggested that the DDBD plasma reactor could be used for higher conversion efficiency as well as for avoiding solid residue deposition on the electrode.These depositions can interfere with the performance of the reactor.
