Arabidopsis ABF3 and ABF4 Transcription Factors Act with the NF-YC Complex to Regulate SOC1 Expression and Mediate Drought-Accelerated Flowering

The drought-escape response accelerates flowering in response to drought stress, allowing plants to adaptively shorten their life cycles. Abscisic acid (ABA) mediates plant responses to drought, but the role of ABA-responsive element (ABRE)-binding factors (ABFs) in the drought-escape response is poorly understood. Here, we show that Arabidopsis thaliana ABF3 and ABF4 regulate flowering in response to drought through transcriptional regulation of the floral integrator SUPPRESSOR OF OVEREXPRESSION OF CONSTANS1 (SOC1). The abf3 abf4 mutant displayed ABA-insensitive late flowering under long-day conditions. Ectopic expression of ABF3 or ABF4 in the vasculature, but not in the shoot apex, induced early flowering, whereas expression of ABF3 fused with the SRDX transcript!onal repressor domain delayed flowering. We identified SOC1 as a direct downstream target of ABF3/4, and found that SOC1 mRNA levels were lower in abf3 abf4 than in wild-type plants. Moreover, induction of SOC1 by ABA was hampered in abf3 abf4 mutants. ABF3 and ABF4 were enriched at the -1028- to -657-bp region of the SOC1 promoter, which does not contain canonical ABF-ABRE-binding motifs but has the NF-Y binding element. We found that ABF3 and ABF4 interact with nuclear factor Y subunit C (NF-YC) 3/4/9 in vitro and in planta, and induction of SOC1 by ABA was hampered in nf-yc3 yc4 yc9 mutants. Interestingly, the abf3 abf4, nf-yc3 yc4 yc9, and sod mutants displayed a reduced drought-escape response. Taken together, these results suggest that ABF3 and ABF4 act with NF-YCs to promote flowering by inducing SOC1 transcription under drought conditions . This mechanism might contribute to adaptation by enabling plants to complete their life cycles under drought stress.

Acupuncture: a potent therapeutic tool for inducing adult neurogenesis

Acupuncture has been broadly used as a major therapeutic tool for at least 2,100 years in East Asia. Acupuncture is used for improving the pathological condition of remote organs, such as stomach, liver, brain, etc. by stimulating skin and muscle with fine metal needles through meridians which have been regarded as pathways of Qi within the body （Ernst, 2006）. Physicians in East Asia have tried to treat many kinds of diseases by acupuncture and it was considerably success- ful. Various diseases for which acupuncture is known to be effective, are neurological diseases, including sequelae of stroke, Alzheimer＇s disease （AD）, Parkinson＇s disease （PD）, and so on （Nam et al., 2013）.

Survival and maintenance of regulatory T cells require the kinase TAK1

Regulatory T （Treg） cells play a central role in regulating peripheral immune tolerance and preventing autoimmunity. Despite the extensive studies on the development of Treg cells, the molecular mechanisms that maintain the population of committed Treg cells remain poorly understood. We show here that Treg-conditional ablation of the kinase TAK1 reduced the number of Treg cells in the peripheral lymphoid organs, causing abnormal activation of conventional T cells and autoimmune symptoms. Using an inducible gene knockout approach, we further demonstrate that TAK1 is crucial for the survival of Treg cells. Expression of a constitutively active IKB kinase partially restored the level of Treg cells in the TAKITreg-KO mice. These results suggest a crucial role for TAK1 for maintaining the survival of committed Treg cells under physiololzical conditions.

Soil salinity is one of the most serious agricultural problems that significantly reduce crop yields in the aridand semi-arid regions. It influences various phases of plant growth and developmental processes, such as seed germina-tion, leaf and stem growth, and reproductive propagation. Salt stress delays the onset of flowering in many plant spe-cies. We have previously reported that the Arabidopsis BROTHER OF FT AND TFL1 （BFT） acts as a floral repressor undersalt stress. However, the molecular mechanisms underlying the BFT function in the salt regulation of flowering inductionis unknown. In this work, we found that BFT delays flowering under high salinity by competing with FLOWERING LOCUST （FT） for binding to the FD transcription factor. The flowering time of FD-deficient fd-2 mutant was insensitive to highsalinity. BFT interacts with FD in the nucleus via the C-terminal domain of FD, which is also required for the interactionof FD with FT, and interferes with the FT-FD interaction. These observations indicate that BFT constitutes a distinct saltstress signaling pathway that modulates the function of the FT-FD module and possibly provides an adaptation strategythat fine-tunes photoperiodic flowering under high salinity.

Piezoelectric strain sensor with high sensitivity and high stretchability based on kirigami design cutting

Wearable technology requires high-performance sensors with properties such as small size,flexibility,and wireless ***,sensitivity,and tunability are crucial sensor properties;stretchability and sensitivity ensure user comfort and accurate sensing performance,while tunability is essential for implementing sensors in diverse applications with different ranges of *** this study,we developed a high performance kirigami piezoelectric strain *** finite element analysis,the sensing performance was evaluated,and the kirigami patterns were *** electromechanical properties of sensors with four different kirigami patterns were analyzed.A sensor voltage measurement circuit was also designed,amplifying the output voltage 86.5 times by improving measurement accuracy.A piezoelectric kirigami sensor was constructed with a sensitivity of 9.86 V/cm^(2) and a stretchability of 320.8%,higher than those of previously reported kirigami piezoelectric strain ***,the fabricated sensor was successfully applied in a haptic glove for playing musical instruments.

Effects of ethanol on insulin-like growth factor-Ⅰ system in primary cultured rat hepatocytes: Implications of JNK1/2 and alcoholdehydrogenase

AIM: To evaluate the effects of ethanol on the insulin- like growth factor-Ⅰ (IGF-Ⅰ) system involved in c-Jun N-terminal kinase (JNK1/2) and alcoholdehydrogenase (ADH) activity in primary cultured rat hepatocytes. METHODS: Hepatocytes isolated from male Sprague-Dawley rats were incubated with various concentrations of ethanol for different durations of time. The cells were pretreated with SP600125 (10 μmol/L) and 4-MP (200 μmol/L), and then treated with ethanol (200 mmol/L). We then measured IGF-Ⅰ secretion, IGF-Ⅰ mRNA expression, cell viability and JNK1/2 activity by radioimmunoassay, RT-PCR, MTT assay and Western blot, respectively (n = 6). RESULTS: Ethanol induced the activity of phospho (p)-JNK1/2, reaching a maximum at 60 min and then decreasing at 180 min. The effects of ethanol on the IGF-Ⅰ system were increased at 60 min (secretion: 7.11 ± 0.59 ng/mg protein vs 4.91 ± 0.51 ng/mg, mRNA expression: 150.2% ± 10.2% vs 101.5% ± 11.3%, P = 0.045) and then decreased at 180 min (secretion: 3.89 ± 0.25 ng/mg vs 5.4 ± 0.54 ng/mg protein; mRNA expression: 41.5% ± 10.4% vs 84.7% ± 12.1%, P = 0.04), however cell viability was decreased in a dose- and time-dependent manner. SP600125 blocked the ethanol-induced changes (at 60 min). Additionally, 4-methylpyrazole prevented the ethanol-induced decreases in the IGF-Ⅰ system, cell viability and p-JNK1/2 activity (at 180 min). CONCLUSION: This study suggests that ethanol- induced p-JNK1/2 activation is associated with the IGF-Ⅰ system and cell viability in hepatocytes. Furthermore, alcohol dehydrogenase is involved in the relationship between ethanol-induced inactivation of p-JNK1/2 and the changes of the IGF-Ⅰ system and cell viability.

Bidirectional transfer of quantum information for unknown photons via cross-Kerr nonlinearity and photon-number-resolving measurement

We propose an arbitrary controlled-unitary （CU） gate and a bidirectional transfer scheme of quantum information （BTQI） for unknown photons. The proposed CU gate utilizes quantum non-demolition photon-number-resolving measure- ment based on the weak cross-Kerr nonlinearities （XKNLs） and two quantum bus beams; the proposed CU gate consists of consecutive operations of a controlled-path gate and a gathering-path gate. It is almost deterministic and is feasible with current technology when a strong amplitude of the coherent state and weak XKNLs are employed. Compared with the existing optical multi-qubit or controlled gates, which utilize XKNLs and homodyne detectors, the proposed CU gate can increase experimental realization feasibility and enhance robustness against decoherence. According to the CU gate, we present a BTQI scheme in which the two unknown states of photons between two parties （Alice and Bob） are mutually swapped by transferring only a single photon. Consequently, by using the proposed CU gate, it is possible to experimentally implement the BTQI scheme with a certain probability of success.

FLOWERING LOCUS M isoforms differentially affect the subcellular localization and stability of SHORT VEGETATIVE PHASE to regulate temperature-responsive flowering in Arabidopsis

Temperature is an important environmental cue that affects flowering time in *** MADS-box transcription factor FLOWERING LOCUS M(FLM)forms a heterodimeric complex with SHORT VEGETATIVE PHASE(SVP)and controls ambient temperature-responsive flowering in ***-βand FLM-δ,two major splice variants produced from the FLM locus,exert opposite effects on flowering,but the molecular mechanism by which the interaction between FLM isoforms and SVP affects temperature-responsive flowering remains poorly ***,we show that FLM-βand FLM-δplay important roles in modulating the temperature-dependent behavior,conformation,and stability of *** localization of SVP decreases as temperature ***-βis required for SVP nuclear translocation at low temperature,whereas SVP interacts with FLM-δmainly in the cytoplasm at high *** preferentially binds to FLM-βat low temperature in tobacco leaf *** shows high binding affinity to FLM-βat low temperature and to FLM-δat high *** undergoes similar structural changes in the interactions with FLM-βand FLM-δ;however,FLM-δlikely causes more pronounced conformational changes in the SVP ***-δcauses rapid degradation of SVP at high temperature,compared with FLM-β,possibly via *** of lysine 53 or lysine 165 in SVP causes increased abundance of SVP due to reduced ubiquitination of SVP and thus delays flowering at high *** findings suggest that temperature-dependent differential interactions between SVP and FLM isoforms modulate the temperature-responsive induction of flowering in Arabidopsis.
