MicroRNA aberrations:An emerging field for gallbladdercancer management

Gallbladder cancer(GBC) is infrequent but most lethal biliary tract malignancy characterized by an advanced stage diagnosis and poor survival rates attributed to absence of specific symptoms and effective treatment options. These necessitate development of early prognostic/predictive markers and novel therapeutic interventions. Micro RNAs(mi RNAs) are small, noncoding RNA molecules that play a key role in tumor biology by functioning like tumor suppressor- or oncogenes and their aberrant expression are associated with the pathogenesis of several neoplasms with overwhelming clinical implications. Since mi RNA signature is tissue specific, here, we focused on current data concerning the mi RNAs abberations in GBC pathogenesis. In GBC, mi RNAs with tumor suppressor activity(mi R-135-5p, mi R-335, mi R-34 a, mi R-26 a, mi R-146b-5p, Mir-218-5p, mi R-1, mi R-145, mir-130a) were found downregulated, while those with oncogenic property(mi R-20 a, mi R-182, mir-155) were upregulated. The expression profile of mi RNAs was significantly associated with GBC prognosis and prediction, and forced over-expression/ inhibition of these mi RNAs was shown to affect tumor growth and development. Further, differential expression of mi RNAs in the blood samples of GBC patients suggest mi RNAs as promising noninvasive biomarker. Thus, mi RNAs represent potential candidate for GBC management, though many hurdles need to be overcome before mi RNAs therapy can be clinically applied to GBC prevention and treatment.

Objective:To investigate the interaction of p53 with docetaxel and berberine and their anticancer activities against oral squamous cell ***:The interaction between p53 with docetaxel and berberine was investigated and their mechanisms of action against oral squamous cell carcinoma were *** studies were performed to determine any toxic impact of the drugs on the vital organs of tested ***:In silico results revealed the molecular interaction of docetaxel and berberine with p53 and the molecules were found to be potential p53 *** and berberine inhibited the proliferation of cancer cells in a concentration-dependent *** cytometry analysis revealed that docetaxel and berberine at IC50 concentrations upregulated the expression of p53 in oral squamous cell carcinoma cells,thus triggering apoptotic cell *** addition,no toxicity was observed in the liver and kidney tissues of mice after docetaxel and berberine ***:Docetaxel and berberine significantly suppressed the proliferation of oral cancer cells by activating p53 expression and causing apoptotic cell *** compounds can be potential agents for the treatment of oral cancer,with little to no toxicity at the tissue level.

Effect of exogenous progesterone on cumulus characteristics of buffalo oocytes by allowing passage of more number of sperm through cumulus but not essentially fertilization

Objective:To understand the level of progesterone (p4) in different quality of buffalo cumulus oocyte complexes (COCs) and further to evaluate the effect of exogenous p4 supplementation on maturation and subsequent developmental ability of poor quality brilliant cresyl blue (BCB-)***: progesterone secreted by different quality of buffalo oocytes was estimated by enzyme linked immunosorbent assay and the concentration differences were translated into p4 doses to be incorporated in the maturation medium of BCB-ve COCs followed by expression analysis of genes involved in the cumulus expansion, extracellular matrix disintegration and progesterone receptor signalling. In addition, the study also evaluated the effect of exogenous p4 on sperm-cumulus interaction. Results: More than 10-fold up-regulated expression of progesterone receptor in p4 supplemented oocytes signified that p4 might be acting predominantly through this receptor. Also, exogenous p4 supplementation had significant effect on transcatheter arterial chemoembolization protease regulated by p4-progesterone receptor pathway which in turn had an important role in extracellular matrix disintegration. On the contrary, cumulus expansion genesHAS2,TNFAIp6,AREG were not altered upon p4 supplementation. Also, it was observed that p4 addition did facilitate passage of significantly more number of spermatozoa through p4 treated cumulus cells. Further, incorporation of different doses of p4 did not improve significantly the cleavage and blastocyst rates of BCB-ve ***: Different qualities of buffalo COCs secrete substantially diverse levels of p4, and its supplementation has a role in oocyte maturation via modulation of cumulus characteristics but perhaps not fertilization.

Effect of Chromium on Magnetic Characteristics of powder processed Fe-0.35wt%p Alloy

The present investigation deals with hot powder preform forging technique for the development of high density iron-phosphorus based alloys. These alloys are known for hot-shortness and are therefore not considered suitable for high temperature working. To remove this problem proper soaking at high temperature to eliminate iron- phosphide eutectic and bring entire phosphorus into solution in iron was aimed. Attempting hot forging thereafter completely eliminates hot as well as cold shortness and thereby helps to form these alloys into very thin sheets. It has also been possible to eliminate the use of costly hydrogen atmosphere during sintering by use of addition of carbon as a reducing agent to form CO gas within the compact by reacting with oxygen of iron powder particles. The glassy ceramic coating applied over the compact serves as a protective coating to avoid atmospheric oxygen attack over the compact held at high temperature. Combined application of carbon and glassy ceramic coating has lead to economy in p/M processing for soft magnetic applications. Fe- 0.07C- 0.3O- 0.35wt% p- 0.35wt% Cr alloy so formed yielded coercivity as low as 0.42 Oe, resistivity as high as 21.8 μ cm and total loss as low as 0.170 W/Kg. Such a combination of properties may suit their application in magnetic relays and transformer cores.

Effect of Nutrient Management and planting Geometry on productivity of Hybrid Rice (<i>Oryza sativa</i>L.) Cultivars

Field experiments were conducted during the wet seasons of 2006 and 2007 at the Agricultural Experimental Farm of the Indian Statistical Institute, Giridih, a part of eastern plateau region of India. The study was designed to investigate the effect of planting geometry and nutrient management practices on productivity of two hybrid rice cultivars. Split-plot design with three replications was adopted to carry out the experiment by allocating combinations of treatments of planting geometry and rice cultivar in main-plots and nutrient management treatments in sub-plots. “CNRH-3” rice proved its efficiency in terms of grain yield that was also reflected in yield attributing characters such as number of productive tillers, number of grains per panicle, length of panicle, panicle weight, test weight and harvest index. Higher rice grain yield was registered when the cultivars grown in 20 cm × 20 cm planting geometry. Rice cultivars grown with the application of inorganic fertilizers alone produced maximum grain yield and also recorded higher values of ancillary characters. The maximum amount of N, p and K was taken up by the “CNRH-3” rice, whereas maximum residual soil fertility was recorded in “pro Agro 6201” rice. Maximum N, p and K uptake values were recorded in 20 cm × 20 cm crop geometry and inorganic fertilizers treatment.
