Deep learning-based virtual staining,segmentation,and classification in label-free photoacoustic histology of human specimens

In pathological diagnostics,histological images highlight the oncological features of excised specimens,but they require laborious and costly staining *** recent innovations in label-free microscopy that simplify complex staining procedures,technical limitations and inadequate histological visualization are still problems in clinical ***,we demonstrate an interconnected deep learning(DL)-based framework for performing automated virtual staining,segmentation,and classification in label-free photoacoustic histology(PAH)of human *** framework comprises three components:(1)an explainable contrastive unpaired translation(E-CUT)method for virtual H&E(VHE)staining,(2)an U-net architecture for feature segmentation,and(3)a DL-based stepwise feature fusion method(StepFF)for *** framework demonstrates promising performance at each step of its application to human liver *** virtual staining,the E-CUT preserves the morphological aspects of the cell nucleus and cytoplasm,making VHE images highly similar to real H&E *** segmentation,various features(e.g.,the cell area,number of cells,and the distance between cell nuclei)have been successfully segmented in VHE ***,by using deep feature vectors from PAH,VHE,and segmented images,StepFF has achieved a 98.00%classification accuracy,compared to the 94.80%accuracy of conventional PAH *** particular,StepFF’s classification reached a sensitivity of 100%based on the evaluation of three pathologists,demonstrating its applicability in real clinical *** series of DL methods for label-free PAH has great potential as a practical clinical strategy for digital pathology.

A potential oncogenic role of the commonly observed E2F5 overexpression in hepatocellular carcinoma

AIM: To explore the expression pattern of E2F5 in primary hepatocellular carcinomas (HCCs) and elucidate the roles of E2F5 in hepatocarcinogenesis. METHODS: E2F5 expression was analyzed in 120 primary HCCs and 29 normal liver tissues by immunohistochemistry analysis. E2F5-small interfering RNA was transfected into HepG2, an E2F5-overexpressed HCC cell line. After E2F5 knockdown, cell growth capacity and migrating potential were examined. RESULTS: E2F5 was significantly overexpressed in primary HCCs compared with normal liver tissues (P = 0.008). The E2F5-silenced cells showed significantly reduced proliferation (P = 0.004). On the colony formation and soft agar assays, the number of colonies was significantly reduced in E2F5-silenced cells (P = 0.004 and P = 0.009, respectively). E2F5 knockdown resulted in the accumulation of G0/G1 phase cells and a reduction of S phase cells. The number of migrating/invading cells was also reduced after E2F5 knockdown (P = 0.021). CONCLUSION: To our knowledge, this is the first evidence that E2F5 is commonly overexpressed in primary HCC and that E2F5 knockdown significantly repressed the growth of HCC cells.

AIM: To assess the role of computed tomography(CT) and magnetic resonance imaging(MRI) and establish imaging criteria of lymph node metastasis in early colorectal ***: One hundred and sixty patients with early colorectal cancer were evaluated for tumor location, clinical history of polypectomy, depth of tumor invasion, and lymph node metastasis. Two radiologists assessed preoperative CT and/or MRI for the primary tumor site detectability, the presence or absence of regional lymph node, and the size of the largest lymph node. Demographic, imaging, and pathologic findings were compared between the two groups of patients based on pathologic lymph node metastasis and optimal size criterion was ***: The locations of tumor were ascending, transverse, descending, sigmoid colon, and rectum. One hundred and sixty early colorectal cancers were classified into 3 groups based on the pathological depth of tumor invasion; mucosa, submucosa, and depth unavailable. A total of 20(12.5%) cancers with submucosal invasion showed lymph node metastasis. Lymph nodes were detected on CT or MRI in 53 patients. The detection rate and size of lymph nodes were significantly higher(P = 0.000, P = 0.044, respectively) in patients with pathologic nodal metastasis than in patients without nodal metastasis. Receiver operating curve analysis showed that a cut-off value of 4.1 mm is optimal with a sensitivity of 78.6% and specificity of 75%.CONCLUSION: The short diameter size criterion of≥ 4.1 mm for metastatic lymph nodes was optimal for nodal staging in early colorectal cancer.

Primary adenocarcinoma of the appendix is a rare malignancy that constitutes 〈 0.5% of all gastrointestinal neoplasms. Moreover, primary signet ring cell carc noma of the appendix is an exceedingly rare entity. We have encountered 15 cases of primary appendiceal cancer among 3389 patients who underwent appen- dectomy over the past 18 years. In the present report, we describe a rare case of primary signet ring cell carcinoma of the appendix with ovarian metastases and unresectable peritoneal dissemination occurring in a 67-year-old female patient. She underwent ap- pendectomy and bilateral salpingo-oophorectomy with a laparoscopy procedure. She then received palliative systemic chemotherapy with 12 cycles of oxaliplatin, 5-flurorouracil, and leucovorin （FOIFOX-4）. The patient currently is well without progression of disease 12 mo after beginning chemotherapy.

field attraction test of (7R,8S)-cis-7,8-epoxy-2- methyloctadec-17-ene (= 7R,8S-epo-2me-17ene-18Hy), a trace component of the sex pheromone gland of the Asian gypsy moth,(Lymantria dispar), traps that were baited with the trace component attracted more L. dispar than traps baited with (7S,8R)-cis-7,8-epoxy-2-methyloctadec-17-ene (= 7S,8R-epo-2me-17ene-18Hy) and unbaited traps. However, traps baited with 7R,8S-epo-2me-17ene-18Hy were less attractive than traps baited with (+)-disparlure [=(7R,8S)-cis-7,8-epoxy-2-methyloctadecane], the sex pheromone of L. dispar. Combination tests with (?)-disparlure, 7R,8S-epo-2me-17ene-18Hy, and 7S,8R-epo-2me- 17ene-18Hy revealed that 7R,8S-epo-2me-17ene-18Hy acted synergistically with (?)-disparlure.

In this study,we investigated the preventive effects of emamectin benzoate 9.7%SL,which was newly developed to reduce the injection volume and number of injection holes required to protect against pine wood *** of the Pinus thunbergii trees injected with emamectin benzoate 9.7%SL at 0.3 mL/cm diameter at breast height(DBH)died within 2 years of inoculation with pine wood *** benzoate 9.7%SL injected at 0.6 mL/cm DBH resulted in no tree mortality for 3 *** residue of emamectin benzoate 9.7%SL in pine twigs injected with 0.3 mL/cm DBH was 0.490μg/g at 1 year after injection and 0.303μg/g after 2 *** residues values are greater than 0.031μg/g,previously determined IC95 value for emamectin benzoate against the pine wood *** field experiment and residue analysis showed that emamectin benzoate 9.7%SL could be a substitute agent for emamectin benzoate 2.15%EC,which is widely used to prevent pine wood nematode in the field and that injection volume and number of injection holes can be greatly educed using this new formulation,which will reduce injury to the cambium,interruption of water movement,and infection of inoculation wounds by wood-decay or blue stain fungi.

Is transcranial direct current stimulation a potential method for improving response inhibition?

Inhibitory control of movement in motor learning requires the ability to suppress an inappropriate action, a skill needed to stop a planned or ongoing motor response in response to changes in a variety of environments. This study used a stop-signal task to determine whether transcranial direct-current stimulation over the pre-supplementary motor area alters the reaction time in motor inhibition. Forty healthy subjects were recruited for this study and were randomly assigned to either the transcranial direct-current stimulation condition or a sham-transcranial direct-current stimulation condition. All subjects consecutively performed the stop-signal task before, during, and after the delivery of anodal transcranial direct-current stimulation over the pre-supplementary motor area （pre-transcranial direct-current stimulation phase, transcranial direct-current stimulation phase, and post-transcranial direct-current stimulation phase）. Compared to the sham condition, there were significant reductions in the stop-signal processing times during and after transcranial direct-current stimulation, and change times were significantly greater in the transcranial direct-current stimulation condition. There was no significant change in go processing-times during or after transcranial direct-current stimulation in either condition. Anodal transcranial direct-current stimulation was feasibly coupled to an interactive improvement in inhibitory control. This coupling led to a decrease in the stop-signal process time required for the appropriate responses between motor execution and inhibition. However, there was no transcranial direct-current stimulation effect on the no-signal reaction time during the stop-signal task. Transcranial direct-current stimulation can adjust certain behaviors, and it could be a useful clinical intervention for patients who have difficulties with response inhibition.

Yolk-shell-structured （***.s）9S8 solid-solution powders：Synthesis and application as anode materials for Na-ionbatteries

Multicomponent metal sulfide materials with a yolk–shell structure and a single phase were studied for the first time as anode materials for sodium-ion batteries. Yolk–shell-structured Fe–Ni–O powders with a molar ratio of iron and nickel components of 1/1 were prepared via one-pot spray pyrolysis. The prepared Fe–Ni–O powders were transformed into yolk–shell-structured (Fe0.5Ni0.5)9S8 solid-solution powders via a sulfidation process. The initial discharge and charge capacities of the (Fe0.5Ni0.5)9S8 powders at a current density of 1 A·g−1 were 601 and 504 mA·h·g−1, respectively. The discharge capacities of the (Fe0.5Ni0.5)9S8 powders for the 2nd and 100th cycle were 530 and 527 mA·h·g−1, respectively, and their corresponding capacity retention measured from the 2nd cycle was 99%. The (Fe0.5Ni0.5)9S8 powders had high initial discharge and charge capacities at a low current density of 0.1 A·g−1, and the reversible discharge capacity decreased slightly from 568 to 465 mA·h·g−1 as the current density increased from 0.1 to 5.0 A·g−1. The synergetic effect of the novel yolk–shell structure and the multicomponent sulfide composition of the (Fe0.5Ni0.5)9S8 powders resulted in excellent sodium-ion storage performance.

changes in brain activation patterns according to cross-training effect in serial reaction time task An functional MRI study

Cross-training is a phenomenon related to motor learning, where motor performance of the untrained limb shows improvement in strength and skill execution following unilateral training of the homologous contralateral limb. We used functional MRI to investigate whether motor performance of the untrained limb could be improved using a serial reaction time task according to motor sequential learning of the trained limb, and whether these skill acquisitions led to changes in brain activation patterns. We recruited 20 right-handed healthy subjects, who were randomly allocated into training and control groups. The training group was trained in performance of a serial reaction time task using their non-dominant left hand, 40 minutes per day, for 10 days, over a period of 2 weeks. The control group did not receive training. Measurements of response time and percentile of response accuracy were performed twice during pre- and post-training, while brain functional MRI was scanned during performance of the serial reaction time task using the untrained right hand. In the training group, prominent changes in response time and percentile of response accuracy were observed in both the untrained right hand and the trained left hand between pre- and post-training. The control group showed no significant changes in the untrained hand between pre- and post-training. In the training group, the activated volume of the cortical areas related to motor function （i.e., primary motor cortex, premotor area, posterior parietal cortex） showed a gradual decrease, and enhanced cerebellar activation of the vermis and the newly activated ipsilateral dentate nucleus were observed during performance of the serial reaction time task using the untrained right hand, accompanied by the cross-motor learning effect. Naturalization of Landscaping Woody Plant, Magnolia obovata Potentially Invasive Species

Magnolia obovata, a tree species native to Japan, is a traditional landscaping tree that has also been introduced to various countries. M. obovata individuals have recently emerged in non-planting areas in Korea, prompting us to investigate its distribution by establishing Dosolsan in an urban forest of Daejeon City as the target site. In order to determine its naturalization, the study explored the status of population growth by examining the diameter at breast height of the individuals, and diameter diminution quotient was calculated. Cores of the trees were collected, and the age distribution was estimated by regression analysis. Reproduction possibility was analyzed by verifying the flowerblooming and fruit-bearing. Spontaneous colonization was investigated in the other potential sites which have different location and environment, *** diameter distribution showed a reverse J-shaped curve, and the diminution quotient was ≥ 1.8. The population was composed of different generations,thus confirming the growth of the population. The distributed M. obovata grew and flowered normally,and followed a normal life cycle. The same phenomenon was observed in other planting areas,residual forests in urban or rural areas, alluvial islands, and mountain forests. The continuous spread of M. obovata in new ecosystems may be attributable to its long-term distribution by birds, relatively fast growth, and maturity time. Investigations on the naturalization and spread of M. obovata in Korea, as well as in various countries, are warranted.
