Patterns and mechanisms of belowground carbon responses to changes in precipitation

It is well known that aboveground productivity usually increases with ***,how belowground carbon(C)processes respond to changes in precipitation remains elusive,although belowground net primary productivity(BNPP)represents more than one-half of NPP and soil stores the largest terrestrial C in the *** paper reviews the patterns of belowground C processes(BNPP and soil C)in response to changes in precipitation from transect studies,manipulative experiments,modeling and data integration and *** results suggest the possible existence of nonlinear patterns of BNPP and soil C in response to changes in precipitation,which is largely different from linear response for aboveground productivity.C allocation,root turnover time and species composition may be three key processes underlying mechanisms of the nonlinear responses to changes in precipitation for belowground C *** addition,microbial community structure and long-term ecosystem processes(*** assemblage,soil texture,aggregate stability)may also affect patterns of belowground C processes in response to changes in *** last,we discuss implications and future perspectives for potential nonlinear responses of belowground C processes to changes in precipitation.

Background:EBV-miR-BARTs exhibit significant relevance in epithelial tumors,particularly in EBVassociated gastric and nasopharyngeal ***,their specific mechanisms in the initiation and progression of gastric cancer remain insufficiently *** and Methods:Initially,EBV-miRNA-BART6-5p and its target gene SMAD4 expression were assessed in EBV-associated gastric cancer tissues and cell *** transfection induced overexpression of EBV-miRNA-BART6-5p in AGS and MKN-45,and downregulation in EBVpositive cells(SUN-719).The subsequent evaluation aimed to observe their impact on gastric cancer cell proliferation,migration,and glycolytic processes,with the TGF-β/SMAD4 signaling pathway value clarified using a TGF-β***:EBV-miRNA-BART6-5p exhibits pronounced upregulation in EBV-associated gastric cancer tissues and EBV-positive cells,while its target gene SMAD4 demonstrates downregulated *** of it can promote the proliferation and migration of gastric cancer ***,We found EBV-miRNA-BART6-5p promotes glycolysis of gastric cancer *** of the TGF-β/SMAD4 signaling pathway resulted in suppressed proliferation and migration of gastric cancer cells,concomitant with a diminished glycolytic ***:In this study,we found that EBV-miRNA-BART6-5p can target SMAD4,effectively increasing glycolysis in gastric cancer cells by regulating the TGF-β/SMAD4 signaling pathway,thereby enhancing the proliferation and metastasis of gastric cancer *** findings may offer new insights into the metabolic aspects of gastric cancer.

In order to study structure-function details of TGF-β1, the recombinant mature form of rat TGF-β1 was expressed in bacteria. Synthesis of the 112 amino-acid carboxyl-terminal part of TGF-β1 (amino acid 279-390) was controlled by an inducible gene expression system based on bacteriophage T7 RNA polymerase. This system allowed an active and selective synthesis of recombinant TGF-β1. The molecular weight of expressed TGF-β1 monomer determined on SDS-polyacrylamide gel under reducing conditions was about 13 kD. Serial detergent washes combined with a single gel-filtration purification step were sufficient to purify the expression product to homogeneity. Amino-terminal sequencing revealed that the N-terminal of the recombinant protein was identical to the published data. In Western blot analysis the recombinant polypeptide showed excellent antigenicity against polyclonal TGF-β1 antibody. The mature recombinant rat TGF-β1 expressed in this study provides a useful tool for future detailed structural and functional studies.

AIM: To investigate the effect of N-desulfated heparin onhepatic/renal ischemia and reperfusion injury in ***: Using rat models of 60 minutes hepatic or renalischemia followed by 1 h,3 h,6 h and 24 h reperfusion,animalswere randomly divided into following groups,the shamoperated controls,ischemic group receiving only normalsaline,and treated group receiving N-desulfated heparin ata dose of 12 mg/kg at 5 minutes before reperfusion. P-selectin expression was detected in bepatic/renal tissueswith immunohistochemistry methodRESULTS: P-selectin expression, serum ALT, AST, BUN andCr levels were significantly increased during 60 minuteischemia and 1 h, 3 h, 6 h and 24 h reperfusion,while theincrement was significantly inhibited,and hepatic/renalpathology observed by light microscopy was remarkablyimproved by treatment with the N-desulfated ***,the heparin was found no effects on PT and ***: P-selectin might mediate neutrophilinfiltration and contribute to hepatic/renal ischemia andreperfusion. The N-desulfated heparin might preventhepatic/renal damage induced by ischemia and reperfusioninjury without significant anticoagulant activity.

The second envelope glycoprotein （E2） of hepatitis C virus has been shown to bind human target cells and has become a major target for the development of anti-HCV vaccines. Anti-E2 antibodies have been suggested to be of clinical significance in diagnosis, treatment and prognosis of hepatitis C. However, large-scale expression and purification of E2 proteins in mammalian cells is difficult. As an alternative, E2 fragment （aa 385-730） with a four-amino-acid mutation （aa 568-571 PCNI to RVTS） was expressed as hexa-histidine-tagged full length protein [E2N730（m）] in E. coli and purified to over 85% purity. Purified E2N730（m） was specifically recognized by homologous hepatitis C patient serum in Western blot, suggesting that it displayed E2specific antigenicity. Rabbit antiserum raised against E2N730（m） recognized E2 glyco-proteins expressed in mammalian cells in Western blot. AIM:To elucidate the underlying mechanisms of metastasis and to identify the metabolomic markers of gastric cancer ***:Gastric tumors from metastatic and nonmetastatic groups were used in this *** and different metabolic patterns were analyzed by gas chromatography,mass spectrometry and principal components analysis (PCA),*** performance was validated by the area under the curve (AUC) of receiver operating characteristic ***:Twenty-nine metabolites were differentially expressed in animal models of human gastric *** the 29 metabolites,20 were up-regulated and 9 were down-regulated in metastasis group compared to non-metastasis *** models from the metabolite profiles could differentiate the metastatic from the nonmetastatic specimens with an AUC value of *** metabolites were mainly involved in several metabolic pathways,including glycolysis (lactic acid,alaline),serine metabolism (serine,phosphoserine),proline metabolism (proline),glutamic acid metabolism,tricarboxylic acid cycle (succinate,malic acid),nucleotide metabolism (pyrimidine),fatty acid metabolism (docosanoic acid,and octadecanoic acid),and methylation(glycine).The serine and proline metabolisms were highlighted during the progression of ***:Proline and serine metabolisms play an important role in *** metabolic profiling of tumor tissue can provide new biomarkers for the treatment of gastric cancer metastasis.

We propose an efficient scheme to produce ultrahigh-brightness tens of MeV electron beams by designing a density-tailored plasma to induce a wakefield in the weakly nonlinear regime with a moderate laser energy of 120 *** this scheme,the second bucket of the wakefield can have a much lower phase velocity at the steep plasma density down-ramp than the first bucket and can be exploited to implement longitudinal electron injection at a lower laser intensity,leading to the generation of bright electron beams with ultralow emittance together with low energy ***-dimensional particle-in-cell simulations are carried out and demonstrate that high-quality electron beams with a peak energy of 50 MeV,ultralow emittance of28 nm rad,energy spread of 1%,charge of 4.4 pC,and short duration less than 5 fs can be obtained within a 1-mm-long tailored plasma density,resulting in an ultrahigh six-dimensional brightness B6D,n of2×1017 A/m2/0.1%.By changing the density parameters,tunable bright electron beams with peak energies ranging from 5 to 70 MeV,a small emittance of B0.1 mm mrad,and a low energy spread at a few-percent level can be *** bright MeV-class electron beams have a variety of potential applications,for example,as ultrafast electron probes for diffraction and imaging,in laboratory astrophysics,in coherent radiation source generation,and as injectors for GeV particle accelerators.
