Epilepsy,which is characterized by unprovoked seizures and has a prevalence of nearly 1%,is becoming one of the leading causes of disability and death globally[1].The exact mechanisms behind epilepsy are still unknown,which contributes to the inadequate efficacy of anti-seizure medications(ASMs)that mainly target membrane ion channels and neurotransmission[2].

Over 70 million people worldwide suffer from epilepsy-one of the most prevalent chronic neurological disorders[1].Despite the availability of over thirty anti-seizure drugs(ASDs)that mainly act on voltage-gated ion channels or inhibitory GABAergic receptors,the effectiveness of these drugs in achieving seizure freedom remains unsatisfactory due to pharmacoresistance and the inevitable occurrence of side-effects.

Satellite glial cells are unique glial cells that surround the cell body of primary sensory *** increasing body of evidence suggests that in the presence of inflammation and nerve damage,a significant number of satellite glial cells become activated,thus triggering a series of functional *** suggests that satellite glial cells are closely related to the occurrence of chronic *** this review,we first summarize the morphological structure,molecular markers,and physiological functions of satellite glial ***,we clarify the multiple key roles of satellite glial cells in chronic pain,including gap junction hemichannel Cx43,membrane channel Pannexin1,K channel subunit 4.1,ATP,purinergic P2 receptors,and a series of additional factors and their receptors,including tumor necrosis factor,glutamate,endothelin,and ***,we propose that future research should focus on the specific sorting of satellite glial cells,and identify genomic differences between physiological and pathological *** review provides an important perspective for clarifying mechanisms underlying the peripheral regulation of chronic pain and will facilitate the formulation of new treatment plans for chronic pain.

How to deal with severe spinal cord injuries(SCI)?This question remains an everlasting challenge for *** innovative therapies such as neuromodulation,which targets the surviving tissue,have shown the potential to restore cognitive functions in patients with severe SCI after rehabilitation[1,2].These therapies might enable SCI patients to sustain active movements during rehabilitation training to enhance the reorganization of the neuronal pathways,however,the complete recovery of body movement functions was still not reached with these therapies.A characteristic feature of SCI is the impairment of neural information communication between the neurons in the spinal cord and the brain,which are responsible for regulating body ***-computer interfaces(BCIs)have been successfully applied in dealing with movement dysfunction by establishing a direct connection between brain cortical activity and electrical muscle stimulation,thereby restoring limb function in cases of paralysis[2].

OBJECTIVE Epilepsy is considered a circuit-level dysfunction associated with imbalanced excitation-inhibition, it is therapeutically necessary to identify key brain regions and related circuits in epilepsy. The subiculum is an essential participant in epileptic seizures, but the circuit mechanism underlying its role remains largely elusive. METHODS Here we deconstruct the diversity of subicular circuits in mouse models of epilepsy. Fiber photometry was used to detect intrinsic activities of subicular PV, SST-positive interneurons and CaMKⅡα-positive pyramidal neurons. Optogenetics and chemogenetics were used to selectively active or inactive subicular neurons or their projecting terminals. We also used in vivo and in vitro electrophysiology to record membrane characteristics of single neuron in distinct sub-regions of the subiculum. Finally, single pulse test was used to detect synaptic transmission strength between the subiculum and its downstream target. RESULTS First, we found that two majority of subicular interneurons, which innervate local pyramidal neurons to constrain their excitability,PV and SST-positive neurons showcase distinct calcium dynamics during hippocampal seizures. This could be attributed to distinct neural inputs from para-hippocampal regions of these two neuronal types. During epileptogenesis, PV and SST neurons undergo different circuit reorganization patterns, that is, remarkable increase of external input to subicular PV neurons are seen after seizures,while SST cells receive decimated neural input. As their downstream targets, excitatory subicular pyramidal neurons are also intrinsically activated during hippocampal seizures. Moreover, we found that the subiculum heterogeneously controls the generalization of hippocampal seizures by projecting to different downstream regions. Notably, anterior thalamus projecting subicular neurons bidirectionally mediate seizures, while entorhinal cortex-projecting subicular neurons act oppositely in seizu
