Targeted Genotyping of a Whole-Gene Repertoire by an Ultrahigh-Multiplex and Flexible HD-marker Approach

Targeted genotyping is an extremely powerful approach for the detection of known genetic variations that are biologically or clinically ***,for non-model organisms,large-scale target geno-typing in a cost-effective manner remains a major *** address this issue,we present an ultrahigh-multiplex,in-solution probe array-based high-throughput diverse marker genotyping(HD-marker)approach that is capable of targeted genotyping of up to 86000 loci,with coverage of the whole gene repertoire,in what is a 27-fold and six-fold multiplex increase in comparison with the conventional Illumina GoldenGate and original HD-marker assays,*** perform extensive analyses of var-ious ultrahigh-multiplex levels of HD-marker(30 k-plex,56 k-plex,and 86 k-plex)and show the power and excellent performance of the proposed method with an extremely high capture rate(about 96%)and genotyping accuracy(about 96%).With great advantages in terms of cost(as low as 0.0006 USD per geno-type)and high technical flexibility,HD-marker is a highly efficient and powerful tool with broad appli-cation potential for genetic,ecological,and evolutionary studies of non-model organisms.

Next-generation advanced high/pulsed power capacitors urgently require dielectric materials with outstanding energy storage ***_(0.5)Na_(0.5)TiO_(3)-based lead-free materials exhibit high polarization,but the high remanent polarization and large polarization hysteresis limit their applications in dielectric ***,high-entropy perovskite relaxor ferroelectrics(Na_(0.2)Bi_(0.2)Ba_(0.2)Sr_(0.2)Ca_(0.2))(Ti1-x%Zrx%)O_(3)are designed by adding multiple ions in the A-site and replacing the B-site Ti^(4+)with a certain amount of Zr^(4+).The newly designed system showed high relaxor feature and slim polarization-electric(P-E)***,improved relaxor feature and obviously delayed polarization saturation were found with the increasing of Zr^(4+).Of particular importance is that both high recoverable energy storage density of 6.6 J/cm^(3) and energy efficiency of 93.5%were achieved under 550 kV/cm for the ceramics of x=6,accompanying with excellent frequency stability,appreciable thermal stability,and prosperous discharge *** work not only provides potential dielectric materials for energy storage applications,but also offers an effective strategy to obtain dielectric ceramics with ultrahigh comprehensive energy storage performance to meet the demanding requirements of advanced energy storage applications.

AIM:To evaluate the effect of bone morphogenetic protein-6(BMP-6)on transforming growth factor(TGF)-β_(2)-induced epithelial-mesenchymal transition(EMT)in retinal pigment epithelium(RPE).METHODS:Adult retinal pigment epithelial cell line(ARPE-19)were randomly divided into control,TGF-β_(2)(5μg/L),and BMP-6 small interfering RNA(siRNA)*** cell morphology was observed by microscopy,and the cell migration ability were detected by Transwell *** EMT-related indexes and BMP-6 protein levels were detected by Western ***,a BMP-6 overexpression plasmid was constructed and RPE cells were divided into the control group,TGF-β_(2)+empty plasmid group,BMP-6 overexpression group,and TGF-β_(2)+BMP-6 overexpression *** EMT-related indexes and extracellular regulated protein kinases(ERK)protein levels were ***:Compared with the control group,the migration of RPE cells in the TGF-β_(2) group was significantly ***-β_(2) increased the protein expression levels ofα-smooth muscle actin(α-Sma),fibronectin and vimentin but significantly decreased the protein levels of E-cadherin and BMP-6(Pma,fibronectin and vimentin but significantly decreased the protein expression levels of E-cadherin(Pmarkers(Pmay provide a theoretical basis for the prevention and treatment of proliferative vitreoretinopathy.

Dynamic risk assessment of gas pipeline operation process by fusing visual and olfactory monitoring

With the rapid increase in urban gas consumption,the frequency of maintenance and repair of gas pipelines has escalated,leading to a rise in safety accidents during these *** traditional manual supervision model presents challenges such as inaccurate monitoring results,incomplete risk factor analysis,and a lack of quantitative risk *** research focuses on developing a dynamic risk assessment technology for gas emergency repair operations by integrating the monitoring outcomes of artificial olfactory for gas leakage information and video object recognition for visual safety factor monitoring *** quantitatively evaluate the risk of the operation process,a three-dimensional risk assessment model combining gas leakage with riskcorrelated sensitivity was established as well as a separate three-dimensional risk assessment model integrating visual risk factors with predictable risk ***,a visual risk quantification expression mode based on the risk matrix-radar map method was ***,a risk quantification model based on the fusion of visual and olfactory results was *** verification results of simulation scenarios based on field data indicate that the visual-olfactory fusion risk assessment method can more accurately reflect the dynamic risk level of the operation process compared to simple visual safety factor *** outcomes of this research can contribute to the identification of safety status and early warning of risks related to personnel,equipment,and environmental factors in emergency repair ***,these results can be extended to other operational scenarios,such as oil and gas production stations and long-distance pipeline operations.

Type I interferon(IFN)inhibits a wide spectrum of viruses through stimulating the expression of antiviral *** an IFN-induced protein,myxovirus resistance B(MXB)protein was reported to inhibit multiple highly pathogenic human *** remains to be determined whether MXB employs a common mechanism to restrict different ***,we find that IFN alters the subcellular localization of hundreds of host proteins,and this IFN effect is partially lost upon MXB *** results of our mechanistic study reveal that MXB recognizes vimentin(VIM)and recruits protein kinase B(AKT)to phosphorylate VIM at amino acid S38,which leads to reorganization of the VIM network and impairment of intracellular trafficking of virus protein complexes,hence causing a restriction of virus *** results highlight a new function of MXB in modulating VIM-mediated trafficking,which may lead towards a novel broad-spectrum antiviral strategy to control a large group of viruses that depend on VIM for successful replication.

Methanol synthesis via CO2hydrogenation stands as a pivotal avenue for CO2conversion and fixation,garnering extensive *** reactor configurations and energy supplies,alongside a spectrum of catalyst formulations,have been developed to enhance reaction metrics including conversion,selectivity,productivity,and *** state-of-the-art Cu-based catalysts from thermal catalysis into a photothermal reactor yielded notable results,achieving an overall CO2conversion of 98%and methanol selectivity of 86%.Utilizing the liquid out/gas in concept （LOGIC） reactor,in which only the catalyst bed is irradiated to raise its temperature,facilitated the condensation of products （H2O and CH3OH）,due to their low vapor *** lower concentration of products in vapor phases ensured the continuous progress of the *** synergistic integration of reactor design and catalyst fabrication could capitalize on the strengths of both components and lead to enhanced performance in methanol synthesis.

Dear Editor,Bamboos(Bambusoideae,Poaceae)are of significant economic and ecological importance,supporting billions of people and contributing to international trade(Wu et al.,2023).They play a crucial role in carbon fixation and support a multitude of other species including the giant panda(Clark et al.,2015).Since the release of the first draft genome of moso bamboo(Phyllostachys edulis)(Peng et al.,2013),there has been a notable increase in the sequencing,assembly,and annotation of diverse bamboo genomes(Guo et al.,2019;Zheng et al.,2022;ma et al.,2024).An existing resource for bamboo genomics,BambooGDB(Zhao et al.,2014),provides information based on the individual draft reference genome of moso *** is needed,however,is a resource reinforcing multiple genome assemblies in Bambusoideae to enable researchers to unravel the evolutionary history of chromosomes and genes through cladespecific comparative genomic ***,we present BambooBase(https://***/),a newly developed web-based database designed to provide a seamless user experience for bamboo omics and systematics.
