目的探究何首乌醇提物对3D肝细胞球的毒性。方法设置不同的条件以及细胞比例,测量细胞球直径、致密度、圆度,探索最佳培养条件。3D肝细胞球分为对照组和低、中、高剂量实验组,低、中、高剂量实验组给予0.25、1.00和2.50 mg·mL^(-1)何首乌醇提物,对照组给予等量的培养基。用CellTiter-Glo试剂检测细胞球活性,用荧光定量聚合酶链反应(RT-qRCR)检测肝功能相关基因表达水平,用活死细胞双荧光染色试剂检测细胞球的毒性作用。结果细胞球理想接种条件为每微孔500个细胞,细胞比例为HepG2-Huvec-LX-2=8∶1∶1,圆度为0.91±0.07,坚实度为0.91±0.02,长径比为1.12±0.14,直径为(170.97±14.79)μm;第3、7、10和14天细胞球白蛋白(ALB)mRNA相对表达水平分别为1.00±0.02、0.96±0.02、0.54±0.07和0.52±0.07,细胞色素P4501A2(CYP1A2)mRNA相对表达水平分别为1.00±0.10、2.15±0.16、2.45±0.33和1.30±0.03。对照组和低、中、高剂量实验组多药耐药蛋白2(MRP2)mRNA相对表达水平分别为1.00±0.31、1.38±0.24、1.48±0.06和1.90±0.08,细胞球存活率分别为(98.19±0.49)%、(88.53±0.90)%、(71.60±2.91)%和(56.65±5.41)%。低、中、高剂量实验组上述指标与对照组比较,在统计学上差异均有统计学意义(均P<0.05)。结论建立的肝细胞球共培养模型中Ⅰ/Ⅱ相药物代谢酶、转运体、及肝细胞特异标志性分子白蛋白均有不同程度的表达,可用于中药何首乌的毒性评价,为何首乌的临床应用提供了参考。

BACKGROUND Primary intracranial alveolar soft-part sarcoma(PIASPS)is a rare malignancy.We aimed to investigate the clinical profiles and outcomes for PIASPS.CASE SUMMARY We firstly reported five consecutive cases from our institute.Then,the cases from previous studies were pooled and analyzed to delineate the characteristics of this disease.Our cohort included two males and three females.The median age was 21-years-old(range:8-54-years-old).All the patients received surgical treatment.Gross total resection(GTR),radiotherapy,and chemotherapy were administered in 3 patients,4 patients,and 1 patient,respectively.After a median follow-up of 36 mo,tumor progression was noticed in 4 patients;and 3 patients died of the disease.Pooled data(n=14)contained 5 males and 9 females with a median age of 19 years.The log-rank tests showed that GTR(P=0.011)could prolong progression-free survival,and radiotherapy(P<0.001)resulted in longer overall survival.CONCLUSION Patients with PIASPS suffer from poor outcomes.Surgical treatment is the first choice,and GTR should be achieved when the tumor is feasible.Patients with PIASPS benefit from radiotherapy,which should be considered as a part of treatment therapies.

AIM:To evaluate the effect of bone morphogenetic protein-6(BMP-6)on transforming growth factor(TGF)-β_(2)-induced epithelial-mesenchymal transition(EMT)in retinal pigment epithelium(RPE).METHODS:Adult retinal pigment epithelial cell line(ARPE-19)were randomly divided into control,TGF-β_(2)(5μg/L),and BMP-6 small interfering RNA(siRNA)group.The cell morphology was observed by microscopy,and the cell migration ability were detected by Transwell chamber.The EMT-related indexes and BMP-6 protein levels were detected by Western blotting.Furthermore,a BMP-6 overexpression plasmid was constructed and RPE cells were divided into the control group,TGF-β_(2)+empty plasmid group,BMP-6 overexpression group,and TGF-β_(2)+BMP-6 overexpression group.The EMT-related indexes and extracellular regulated protein kinases(ERK)protein levels were detected.RESULTS:Compared with the control group,the migration of RPE cells in the TGF-β_(2) group was significantly enhanced.TGF-β_(2) increased the protein expression levels ofα-smooth muscle actin(α-SMA),fibronectin and vimentin but significantly decreased the protein levels of E-cadherin and BMP-6(P<0.05)in RPE.Similarly,the migration of RPE cells in the BMP-6 siRNA group was also significantly enhanced.BMP-6 siRNA increased the protein expression levels ofα-SMA,fibronectin and vimentin but significantly decreased the protein expression levels of E-cadherin(P<0.05).Overexpression of BMP-6 inhibited the migration of RPE cells induced by TGF-β_(2) and prevented TGF-β_(2) from affecting EMT-related biomarkers(P<0.05).CONCLUSION:BMP-6 prevents the EMT in RPE cells induced by TGF-β_(2),which may provide a theoretical basis for the prevention and treatment of proliferative vitreoretinopathy.

Corrosion behavior of thermal-sprayed WC cermet coatings in SO_4^(2-) environment

Turpentine oil,known as a forest product from pine trees,is a renewable and abundant raw material in the world.Its dominant constituent is α-pinene.Due to the broad bioactivities of α-pinene,synthesis of α-pinene-based bioactive compounds has become an important aspect in the development and utilization of turpentine oil.[1-6] In this work,a series of novel myrtenal-based 1,2,4-triazole-thioether compounds were designed and synthesized by a multiple-step procedure using α-pinene as starting material.The preliminary bioassay showed that some of the target compounds exhibited prominent growth inhibition activity of greater than 80.0%（A-class activity level） against root of rape（Brassica campestris）,which was better than that of the commercial herbicide flumioxazin used as the positive control.Meanwhile,some of the target compounds showed a certain antifungal activity against the tested fungi.

A series of the electrochemical and long-term corrosion tests was carried out in a 3.5 wt% Na2SO4 solution on thermal-sprayed WC-17Co and WC-10Co-4Cr cermet coatings in order to examine the effect of composition of binder materials on the corrosion behavior. The results reveal that the overall corrosion resistance of the WC-17Co coating is inferior to that of the WC-Co-Cr coatings due to the corrosion of binder materials which induce WC particles to fall off. CoO and WO3 oxide films form on the surface of WC-17Co coating in Na2SO4 solution electrochemical corrosion process, which will protect the coating in the process of corrosion. Cr2O3 oxide film formed on the WC-10Co-4Cr coating surface has a strong hindered role to corrosion. The corrosion mechanism of WC-17Co coating in Na2SO4 solution is entire corrosion of Co matrix, while it is film-hole corrosion mechanism for WC-10Co-4Cr coating.

Understanding the driving mechanism of the divergence of hoarding strategies of animals(i.e.,scatter-hoarding vs.larder-hoarding)is crucial to understand the ecological and evolutionary implications of plant-animal interactions.Zhang et al.(2022)compiled a global-scale dataset from the published literature including 183 species of seed-hoarding rodents,and concluded that phylogenetic conservatism,functional traits,and environmental factors counted for the hoarding behavior divergence of rodents.A more thought-provoking question may be asked:Do these variables contribute equally or not to explain the divergence of hoarding strategies?Here,we re-analyzed Zhang et al's(2022)dataset using partial Ralk calculation(Ives 2019)to parse out the relative contri-lik butions of these variables.

To compensate for their sessile nature,plants have evolved sophisticated mechanisms enabling them to adapt to ever-changing environments.One such prominent feature is the evolution of diverse life history strategies,particularly such that annuals reproduce once followed by seasonal death,while perennials live longer by cycling growth seasonally.This intrinsic phenology is primarily genetic and can be altered by environmental factors.Although evolutionary transitions between annual and perennial life history strategies are common,perennials account for most species in nature because they survive well under year-round stresses.This proportion,however,is reversed in agriculture.Hence,perennial crops promise to likewise protect and enhance the resilience of agricultural ecosystems in response to climate change.Despite significant endeavors that have been made to generate perennial crops,progress is slow because of barriers in studying perennials,and many developed species await further improvement.Recent findings in model species have illustrated that simply rewiring existing genetic networks can lead to lifestyle variation.This implies that engineering plant life history strategy can be achieved by manipulating only a few key genes.In this review,we summarize our current understanding of genetic basis of perenniality and discuss major questions and challenges that remain to be addressed.

In this editorial,we comment on the article by Wang et al published in the recent issue of the World Journal of Gastroenterology in 2023.We focused on identifying risk factors for lymph node metastasis(LNM)in superficial esophageal squamous cell carcinoma(SESCC)patients and how to construct a simple and reliable clinical prediction model to assess the risk of LNM in SESCC patients,thereby helping to guide the selection of an appropriate treatment plan.The current standard treatment for SESCC is radical esophagectomy with lymph node dissection.However,esophagectomy is associated with considerable morbidity and mortality.Endoscopic resection(ER)offers a safer and less invasive alternative to surgical resection and can enable the patient's quality of life to be maintained while providing a satisfactory outcome.However,since ER is a localized treatment that does not allow for lymph node dissection,the risk of LNM in SESCC limits the effectiveness of ER.Understanding LNM status can aid in determining whether patients with SESCC can be cured by ER without the need for additional esophagectomy.Previous studies have shown that tumor size,macroscopic type of tumor,degree of differentiation,depth of tumor invasion,and lymphovascular invasion are factors associated with LNM in patients with SESCC.In addition,tumor budding is commonly associated with LNM,recurrence,and distant metastasis,but this topic has been less covered in previous studies.By comprehensively evaluating the above risk factors for LNM,useful evidence can be obtained for doctors to select appropriate treatments for SESCC patients.
