Extracellular vesicles have been identified as pivotal mediators of intercellular communication with critical roles in physiological and pathological *** this route,several molecules(e.g.,nucleic acids,proteins,metabolites) can be transferred to proximal and distant targets to convey specific *** vesicle-associated cargo molecules have been proposed as markers of several disease conditions for their potential of tracking down the generating ***,circulating extracellular vesicles may represent biomarkers of dysfunctional cellular quality control systems especially in conditions characterized by the accrual of intracellular misfolded ***,the identification of extracellular vesicles as tools for the delivery of nucleic acids or other cargo molecules to diseased tissues makes these circulating shuttles possible targets for therapeutic *** increasing interest in the study of extracellular vesicles as biomarkers resides mainly in the fact that the identification of peripheral levels of extracellular vesicle-associated proteins might reflect molecular events occurring in hardly accessible tissues,such as the brain,thereby serving as a "brain liquid biopsy".The exploitation of extracellular vesicles for diagnostic and therapeutic purposed might offer unprecedented opportunities to develop personalized ***,we discuss the bright and dark sides of extracellular vesicles in the setting of two main neurodegenerative diseases(i.e.,Parkinson's and Alzheimer's diseases).A special focus will be placed on the possibility of using extracellular vesicles as biomarkers for the two conditions to enable disease tracking and treatment monitoring.

Quenching autofluorescence in the alimentary canal tissues of Bactericera cockerelli(hemiptera:Triozidae)for immunofluorescence labeling

Immunofluorescence has been widely used to localize microbes or specific molecules in insect tissues or ***,significant autofluorescence is frequently observed in tissues which can interfere with the fluorescent identification of target antigens,leading to inaccurate or even false positive fluorescent *** alimentary canal of the potato psyllid,Bactericera cockerelliŠulc,exhibits intense autofluorescence,hindering the application of immunolocalization for the detection and localization of the economically important pathogen transmitted by this insect,“Candidatus Liberibacter solanacearum”(Lso).In the present study,we tested the use of irradiation,hydrogen peroxide(h2O2)and Sudan black B(SBB)treatments to reduce the autofluorescence in the *** alimentary canal ***,we assessed the compatibility of the above‐mentioned treatments with Lso immunolocalization and actin staining using *** results showed that the autofluorescence in the alimentary canal was reduced by irradiation,h2O2,or SBB *** compatibility assays indicated that irradiation and h2O2 treatment both greatly reduced the fluorescent signal associated with Lso and ***,the SBB incubation preserved those target signals,while efficiently eliminating autofluorescence in the psyllid alimentary ***,herein we propose a robust method for reducing the autofluorescence in the *** alimentary canal with SBB treatment,which may improve the use of immunofluorescence labeling in this *** method may also have a wide range of uses by reducing the autofluorescence in other arthropod species.

A comparison of conventional and computer-assisted semen analysis.（CRISMAS software） using samples from 166 young Danish men

The aim of the present study was to compare assessments of sperm concentration and sperm motility analysed by conventional semen analysis with those obtained by computer-assisted semen analysis （CASA）（Copenhagen Rigshospitalet Image house Sperm Motility Analysis System （CRISMAS） 4.6 software） using semen samples from 166 young Danish men. The CRISMAS software identifies sperm concentration and classifies spermatozoa into three motility categories. To enable comparison of the two methods, the four motility stages obtained by conventional semen analysis were, based on their velocity classifications, divided into three stages, comparable to the three CRISMAS motility categories： rapidly progressive （A）, slowly progressive （B） and non-progressive （C＋ D）. Differences between the two methods were large for all investigated parameters （P〈0.001）. CRISMAS overestimated sperm concentration and the proportion of rapidly progressive spermatozoa and, consequently, underestimated the percentages of slowly progressive and non-progressive spermatozoa, compared to the conventional method. To investigate whether results drifted according to time of semen analysis, results were pooled into quarters according to date of semen analysis. CRISMAS motility results appeared more stable over time compared to the conventional analysis; however, neither method showed any trends. Apparently, CRISMAS CASA results and results from the conventional method were not comparable with respect to sperm concentration and motility analysis. This needs to be accounted for in clinics using this software and in studies of determinants of these semen characteristics.

In citrus,lignin overaccumulation in the juice sac results in granulation and an unpleasant fruit texture and *** integrating metabolic phenotyping and transcriptomic analyses,we found 702 differentially expressed genes(DEGs),including 24 transcription factors(TFs),to be significantly correlated with lignin ***58 was further identified as a critical R2R3 MYB TF involved in lignin overaccumulation owing to its high transcript levels in huanong Red-fleshed pummelo(hR,Citrus grandis)*** expression of CgMYB58 led to an increase in the lignin content in the pummelo fruit mesocarp,whereas its stable overexpression significantly promoted lignin accumulation and upregulated 19 lignin biosynthetic *** these genes,CgPAL1,CgPAL2,Cg4CL1,and CgC3h were directly modulated by CgMYB58 through interaction with their promoter ***,we showed that juice sac granulation in pummelo fruits could be affected by indole-3-acetic acid(IAA)and abscisic acid(ABA)*** hR pummelo,ABA significantly accelerated this granulation,whereas IAA effectively inhibited this *** together,these results provide novel insight into the lignin accumulation mechanism in citrus *** also revealed the theoretical basis via exogenous IAA application,which repressed the expression of CgMYB58 and its target genes,thus alleviating juice sac granulation in orchards.

Background:Among urban stimuli,anthropogenic noise has been identified to be one of the behavioral drivers of species that rely on acoustic signals for *** have shown both species‑specific and assemblage responses to urban noise,ranging from the modulation of their acoustic frequencies and spatiotemporal adjustments to declines in species *** this study,we assessed the citywide relationship between two anthropogenic noise variables(noise levels recorded during bird surveys and daily average noise levels)and vegetation cover with bird species ***:This study was conducted in the city of Xalapa(Mexico)through a 114 citywide point‑count *** recorded bird communities at each sampling *** measured noise levels using a sound level meter while performing point‑***,we generated a map of average daily noise of the city using an array of 61 autonomous recording units distributed across the city of Xalapa and calculated daily noise levels for the 114 *** ran a linear model(LM)to assess potential relationships between both point‑count and daily(24 h)noise values and vegetation cover with bird ***:Results from the LM show:(1)a negative relationship between maximum point‑count noise and avian species richness,(2)no relationship between 24 h noise and bird species richness,and(3)a positive relationship between vegetation cover and bird species ***:Results provide evidence that decreases in urban bird species richness do not necessarily imply the permanent absence of species,suggesting that birds can temporarily fly away from or avoid sites when noisy,become cryptic while noisy events are occurring,or be undetected due to our inability to record them in the field during noisy events.

With the continued development of tracking technology and increasing interest in animal movement,our understanding of migration behavior has become more ***,there are still many species that have not been well studied,particularly sea ***,we present the first year-round Global Positioning System(GPS)tracking data of the Black-tailed Gull(Larus crassirostris)at the population *** used solar-powered GPS-Global System for Mobile communication(GSM)loggers to successfully track 30 individuals breeding at Xingrentuo Islet,Liaoning Province,China,for 1-3 *** for one individual who roamed in the far north of the Yellow Sea during non-breeding period,all others did a directed southward *** routes and wintering sites differed among migrating gulls and between years for the birds tracked for two or more ***,during wintering,the migrating gulls were more likely to travel over a large body of water and shift sites,and some trajectories were quite complex,which was probably closely related to what we observed in the field about their boat-chasing *** to wintering movements,the post-breeding movements ranged over a smaller ***,almost all of them had a long post-breeding period near the breeding islet(≥120days,he breeding islet),and 80%of the gulls who were tracked more than one year had at least one faithful post-breeding *** to the post-breeding period,only approximately half of the migrating gulls had a pre-breeding period that was shorter(3-20 days)and closer to the breeding islet(≤80 km).Migration distance varied among migrating gulls(range 209-2405 km)and the gulls moved least distance during postbreeding ***,we found that the southward movement of the migrating gulls occurred when the temperature near the breeding islet dropped;specifically,the gulls directly migrated southward away from the post-breeding *** results suggest that the B

Implications of turbulent shear on clay floc break-up along the Atlantic estuary(Bouregreg),Morocco

Water depth,salinity,current,and suspended sediment concentration(SSC) were measured along with the grain size distribution of bed sediment along an estuarine longitudinal *** floc size increased with increase in the percentage of clay and silt,while decreased with increase in the percentage of sand content of bed *** turbulent shear,G,had a direct effect on floc size with its value increasing with increase in G up to a G value of 15 s,while an inverse relation existed between floc size and G at higher G(C> 15 s).Further,higher turbulence enabled sand to get resuspended and cause additional shear leading to the break-up of *** attempt was made to modify G to account for the combined effect of water turbulence(C) and shear imparted by sand(C) and the impact of the modification of G on the predictability of floc size was evaluated.A new model was developed which explains floc size in terms of sediment concentration(C),salinity gradient(S),and G for different scenarios based on the value of *** analysis was done for observed floc size(FS) and predicted floc size using four approaches:(Ⅰ) FS β C~χ;(Ⅱ) FS α C~χS;(Ⅲ) FS α C~χSG~z for G 15 s; and(Ⅳ) FS α C~χSGfor G> 15 sand G=G+G,where x,y,and z are determined by *** was observed that the predictability of the floc size improved when the turbulence was modified to account for shear imparted by sand so that the coefficient of determination was increased from 0.78 for model Ⅲ to 0.89 for model Ⅳ.Further,the settling velocity was expressed as a function of suspended sediment concentration,turbulent shear,and salinity *** predictability of settling velocity was improved(R~2 increased from 0.77 to 0.86) when the additional turbulence created by sand was incorporated in the non-dimensional empirical *** study highlights the influence of sand in causing the break-up of flocs and suggests that for turbulence shear values high enough to resuspend

Genotyping-by-sequencing of pear (Pyrus spp.) accessions unravels novel patterns of genetic diversity and selection footprints
