Limosilactobacillus reuteri QS01 is potential probiotic isolated from the intestinal microbiota of healthy women in early pregnancy. In the current study， we examined whether QS01 can prevent gestational diabetes mellitus (GDM) in an enhanced mouse model of the condition. Female C57BL/6J mouse offspring (F1 generation) born to dams fed a control (CON) or low-protein diet (GDM group) during gestation and lactation were used. Pregnant F1 mice fed a standard diet were randomly assigned to 5 groups: CON， GDM， and GDM mice treated with metformin， QS01 or Lacticaseibacillus rhamnosus HN001. An oral glucose tolerance test was performed before sacrifice at gestational day 17. Glucose tolerance was significantly ameliorated by all three treatments. QS01 supplementation fortified the intestinal mucosal barrier and inhibited the escalation of plasma inflammatory cytokines. QS01 treatment altered the cecal microbiota composition and function. Plasma and cecal metabolite profiles were modulated by QS01， prominently demonstrating significant upregulation of indole lactate and L-tryptophan in plasma and 5-hydroxy-tryptophan in the cecum， positively correlated with gut Lactobacillus abundance. In summary， QS01 plays a role in preventing GDM by remodeling the intestinal microbiota， reinforcing the intestinal mucosal barrier and alleviating chronic inflammation.

Dear editor,University laboratory-related events include fire,explosion,chemical gas leakage,poisoning related to the loss of toxic reagents,specific pathogen infection,leakage of radioactive material,etc.[1]Chloroform is an important material used frequently in the laboratory,for organic synthesis or used as an organic glass binder and organic matter extractor.[2]As a widely used anesthetic in clinics in the 1950s and the 1960s,[3]chloroform was abandoned because of hepatorenal toxicity.We report a case of a 24-year-old suicidal female student with chloroform poisoning.

Human-specific insertions play important roles in human phenotypes and diseases.Here we reported a 446-bp insertion(Insert-446)in intron 11 of the TBC1D8B gene,located on chromosome X,and traced its origin to a portion of intron 6 of the EBF1 gene on chromosome 5.Interestingly,Insert-446 was present in the human Neanderthal and Denisovans genomes,and was fixed in humans after human-chimpanzee divergence.We have demonstrated that Insert-446 acts as an enhancer through binding transcript factors that promotes a higher expression of human TBC1D8B gene as compared with orthologs in macaques.In addition,over-expression TBC1D8B promoted cell proliferation and migration through“a dual finger”catalytic mechanism(Arg538 and Gln573)in the TBC domain in vitro and knockdown of TBC1D8B attenuated tumorigenesis in vivo.Knockout of Insert-446 prevented cell proliferation and migration in cancer and normal cells.Our results reveal that the human-specific Insert-446 promotes cell proliferation and migration by upregulating the expression of TBC1D8B gene.These findings provide a significant insight into the effects of human-specific insertions on evolution.

为获取位于雅鲁藏布江断裂带东段加查县新建水电站工程场区的地应力状态,探讨雅鲁藏布江断裂带东段活动性,采用自主研发的新一代水压致裂地应力测试技术,实测获取了工程场区的地应力状态,并在收集雅鲁藏布江断裂带沿线地应力数据基础上,结合摩尔-库仑破裂准则,对其断层活动性进行了探讨。结果表明:(1)在工程厂区内122.75~418.75 m深度,实测最大水平主应力SH介于6.07~37.62MPa之间,最小水平主应力Sh介于3.13~20.33MPa之间,最大水平主应力优势方向为北北东向;(2)主应力关系总体表现为SH>Sh>Sv(Sv为垂向主应力),应力结构有利于逆断层的孕育和活动;(3)实测及收集的大部分数据应力莫尔圆与摩擦系数为0.6的破坏临界线相交,表明整体上雅鲁藏布江断裂带东段发生滑动失稳的风险较高,其中西侧贡嘎至朗县一带风险高于东侧林芝地区。

环渤海河流营养盐的向海输送及环境效应

对2017年枯水期、2019年丰水期、2020年丰水期和2021年丰水期环渤海24条主要入海河流溶解态无机营养盐向海输送的浓度、比值和通量进行了分析.结果表明:环渤海河流溶解无机氮(DIN)浓度在世界河流中处于较高水平,硝酸盐(NO_(3)^(-))为DIN的主要赋存形态,溶解硅(DSi)和溶解无机磷(DIP)浓度较低.环渤海河流每年向渤海输送约0.61Gmol氨盐(NH_(4)^(+))、0.24Gmol亚硝酸盐(NO_(2)^(-))、4.85Gmol NO_(3)^(-)、0.02Gmol DIP和3.27Gmol DSi.环渤海小河流在环渤海河流营养盐的向海输送中扮演着重要作用,尤其是DIP的输送可以占入海河流总通量的80%以上;小河流单位流域面积DIN、DIP和DSi输送通量均高于黄河.与世界河流相比,环渤海河流向海输送的营养盐比例失衡,具有高氮磷比、高硅磷比和低硅氮比的特点,磷限制显著.环渤海河流营养盐向海输送和结构失衡可能会进一步影响到渤海的初级生产和生态环境演变,值得深入研究.

The early determination of disease-related biomarkers can significantly improve the survival rate of patients.Thus,a series of explorations for new diagnosis technologies,such as optical and electrochemical methods,have been devoted to life and health monitoring.Organic thin-film transistor(OTFT),as a state-of-the-art nano-sensing technology,has attracted significant attention from construction to application owing to the merits of being label-free,low-cost,facial,and rapid detection with multi-parameter responses.Nevertheless,interference from non-specific adsorption is inevitable in complex biological samples such as body liquid and exhaled gas,so the reliability and accuracy of the biosensor need to be further improved while ensuring sensitivity,selectivity,and stability.Herein,we overviewed the composition,mechanism,and construction strategies of OTFTs for the practical determination of disease-related biomarkers in both body fluids and exhaled gas.The results show that the realization of bio-inspired applications will come true with the rapid development of high-effective OTFTs and related devices.

Metabolomics as a research field and a set of techniques is to study the entire small molecules in biological samples.Metabolomics is emerging as a powerful tool generally for pre-cision medicine.Particularly,integration of microbiome and metabolome has revealed the mechanism and functionality of microbiome in human health and disease.However,metabo-lomics data are very complicated.Preprocessing/pretreating and normalizing procedures on metabolomics data are usually required before statistical analysis.In this review article,we comprehensively review various methods that are used to preprocess and pretreat metabolo-mics data,including MS-based data and NMR-based data preprocessing,dealing with zero and/or missing values and detecting outliers,data normalization,data centering and scaling,data transformation.We discuss the advantages and limitations of each method.The choice for a suitable preprocessing method is determined by the biological hypothesis,the characteristics of the data set,and the selected statistical data analysis method.We then provide the perspective of their applications in the microbiome and metabolome research.

The Linxia Basin is characterized by an abundance of cenozoic sediments,that contain exceptionally rich fossil resources.Equids are abundant in the Linxia Basin,the fossil record of equids in this region including 16 species that represent 10 genera.In comparison to other classic late cenozoic areas in China,the Linxia Basin stands out,because the fauna and chronological data accompanying Linxia equids render them remarkably useful for biostratigraphy.The anchitheriines in the region,such as Anchitherium and Sinohippus,represent early equids that appeared in the late stages of the middle and late Miocene,respectively.Among the equines,most species of Chinese hipparions have been identified in the Linxia Basin and some species of the genera Hipparion and Hippotherium have FAD records for China.Furthermore,Equus eisenmannae is one of the earliest known species of Equus in the Old World and is well-represented at the Longdan locality.Some species with precise geohistorical distributions can serve as standards for high-resolution chronological units within this framework.Located at the eastern margin of the Tibetan Plateau and subject to considerable uplift,the Linxia Basin has served as a biogeographic transition area for equids throughout the late cenozoic.

Diseases like Alzheimer’s and Parkinson’s diseases are defined by inflammation and the damage neurons undergo due to oxidative stress. A primary reactive oxygen species contributor in the central nervous system, NADPH oxidase 4, is viewed as a potential therapeutic touchstone and indicative marker for these ailments. This in-depth review brings to light distinct features of NADPH oxidase 4, responsible for generating superoxide and hydrogen peroxide, emphasizing its pivotal role in activating glial cells, inciting inflammation, and disturbing neuronal functions. Significantly, malfunctioning astrocytes, forming the majority in the central nervous system, play a part in advancing neurodegenerative diseases, due to their reactive oxygen species and inflammatory factor secretion. Our study reveals that aiming at NADPH oxidase 4 within astrocytes could be a viable treatment pathway to reduce oxidative damage and halt neurodegenerative processes. Adjusting NADPH oxidase 4 activity might influence the neuroinflammatory cytokine levels, including myeloperoxidase and osteopontin, offering better prospects for conditions like Alzheimer’s disease and Parkinson’s disease. This review sheds light on the role of NADPH oxidase 4 in neural degeneration, emphasizing its drug target potential, and paving the path for novel treatment approaches to combat these severe conditions.
