In Vivo Phosphorylation Site.Mapping and Functional Characte.ization of Arabidopsis Phototropin 1

Phototropins （phot1 and phot2） are.blue.light re.e.tor kinase. controlling a range.of re.ponse. that optimize.the.photosynthe.ic e.ficie.cy of plants. Light se.sing is me.iate. by two flavin-binding motifs, known as LOVl and LOV2, locate. within the.N-te.minal re.ion of the.prote.n. Photoe.citation via LOV2 le.ds to activation of the.C-te.minal kinase.domain and conse.ue.tly re.e.tor autophosphorylation. Howe.e., knowle.ge.of the.in-vivo phosphorylation site. for Arabidopsis phototropins is lacking and has impe.e. progre.s in e.ucidating the.functional significance.of re.e.tor phos- phorylation. We.have.purifie. photl from Arabidopsis and ide.tifie. the.in-vivo site. of re.e.tor phosphorylation by liquid chromatography tande. mass spe.trome.ry. Arabidopsis-de.ive. photl binds flavin mononucle.tide.as chromophore.and is phosphorylate. at four major site. locate. upstre.m of LOV2 （Se.^58, Se.^85, Se.^350, and Se.^410）, thre. of which are.induce. by blue.light. Ne.e.the.e.s, structure.function analysis indicate. that the.biological activity of photl can be.attribute. to a modular unit comprising the.LOV2-kinase.re.ion of the.prote.n. Thus, pe.tide.re.ions upstre.m of LOV2, including the.site. of re.e.tor phosphorylation ide.tifie. he.e. do not appe.r to be.important for re.e.tor signaling. By contrast, the.e.re.ions may be.ne.e.sary for maximizing stomatal pe.formance.and possibly light-induce. re.ocalization of photl.

Although pre.e.m birth, hypoxic-ische.ic e.e.ts formula fe.ding, and abnormal bacte.ia colonization are.e.tablishe. risk factors, the.role.of ge.e.ics and vasoactive.inflammatory me.iators is uncle.r Conse.ue.tly, tre.tme.ts do not targe. the.spe.ific unde.lying dise.se.proce.se. and are.symptomatic and surgically invasive. Bre.st-fe.ding is the.most e.fe.tive.pre.e.tative.me.sure. Re.e.t advance. in the.pre.e.tion of ne.rotizing e.te.ocolitis have.focuse. on bioactive.nutrie.ts and trophic factors in human milk. Me.HODS A prospe.tive.institutional database.of 461 patie.ts with pancre.tic injurie. tre.te. from 1990 to 2015 was re.ie.e.. One.hundre. and thirty patie.ts with AAST grade.3, 4 or 5 pancre.tic injurie. unde.we.t re.e.tion(pancre.toduode.e.tomy, n = 20, distal pancre.te.tomy, n = 110), including 30 who had an initial damage.controllaparotomy(DCL) and late. de.initive.surge.y. AAST injury grade., type.of pancre.tic re.e.tion, ne.d for DCL and incide.ce.and ASGS se.e.ity of complications we.e.asse.se.. Uni-and multivariate.logistic re.re.sion analysis was applie.. Re.ULTS Ove.all 238 complications occurre. in 95(73%) patie.ts of which 73% we.e.ASGS grade. 3-6. Nine.e.n patie.ts(14.6%) die..
