AIm: To identify signaling pathways and genes that initiate and commit hepatic stellate cells (HSCs) to transdifferentiation. mETHODS: Primary HSCs were isolated from male Sprague-Dawley rats and cultured on plastic for 0-10 d. Gene expression was assessed daily (quiescent to day 10 culture-activation) by real time polymerase chain reaction and data clustered using AmADA software. The significance of JAK/STAT signaling to HSC transdifferentiation was determined by treating cells with a JAK2 inhibitor. RESULTS: Genetic cluster analyses, based on expression of these 21 genes, showed similar expression profiles on days 1-3, days 5 and 6, and days 7-10, while freshly isolated cells (day Q) and day 4 cells were genotypically distinct from any of the other days. Additionally, gene expression clustering revealed strong upregulation of interleukin-6, JAK2 and STAT3 mRNA in the early stages of activation. Inhibition of the JAK/STAT signaling pathway impeded the morphological transdifferentiation of HSCs which correlated with decreased mRNA expression of several profibrotic genes including collagens, α-SmA, PDGFR and TGFβR. CONCLUSION: These data demonstrate unique clustered genetic profiles during the daily progression of HSC transdifferentiation and that JAK/STAT signaling may be critical in the early stages of transdifferentiation.

Inflammatory bowel disease(IBD)is an inflammatory disorder of the gastrointestinal tract that affects millions of patients *** has a complex and multifactorial etiology leading to excessive exposure of intestinal epithelium to microbial antigens,inappropriate activation of the immune system and ultimately to the damage of intestinal *** numerous efforts have been made to improve the disease management,IBD remains persistently recurring and beyond *** is due largely to the gaps in our understanding of the pathogenesis of IBD that hamper the development of timely diagnoses and effective ***,some recent discoveries,including the beneficial effects of interleukin-22(IL-22)on the inflamed intestine,have shed light on a self-protective mechanism in *** islet-derived(REG/Reg)proteins are small secretory proteins which function as IL-22’s downstream *** studies have demonstrated that IBD patients have significantly increased REG expressions in the injured intestine,but with undefined mechanisms and *** reported functions of REG/Reg proteins in intestinal homeostasis,such as those of antibacterial,anti-inflammatory and tissue repair,lead us to discuss their potential mechanisms and clinical relevance in IBD in order to advance IBD research and management.

The length and time scales of atomistic simulations are limited by the computational cost of the methods used to predict material *** recent years there has been great progress in the use of machine-learning algorithms to develop fast and accurate interatomic potential models,but it remains a challenge to develop models that generalize well and are fast enough to be used at extreme time and length *** address this challenge,we have developed a machine-learning algorithm based on symbolic regression in the form of genetic programming that is capable of discovering accurate,computationally efficient many-body potential *** key to our approach is to explore a hypothesis space of models based on fundamental physical principles and select models within this hypothesis space based on their accuracy,speed,and *** focus on simplicity reduces the risk of overfitting the training data and increases the chances of discovering a model that generalizes *** algorithm was validated by rediscovering an exact Lennard-Jones potential and a Sutton-Chen embedded-atom method potential from training data generated using these *** using training data generated from density functional theory calculations,we found potential models for elemental copper that are simple,as fast as embedded-atom models,and capable of accurately predicting properties outside of their training *** approach requires relatively small sets of training data,making it possible to generate training data using highly accurate methods at a reasonable computational *** present our approach,the forms of the discovered models,and assessments of their transferability,accuracy and speed.

Poly(1-phenyl-1-octyne)s containing different stereogenic and chromophoric pendants {-[(C6H13)C= C(C6H4-p-CO2-R)] n -R = [(1S)-endo]-(?)-borneyl (P3), (1R,2S,5R)-(?)-menthyl (P4), -C6H4-p-(1R,2S,5R)-(?)-menthyl (P5), 2-napthyl (P6), 4-biphenylyl (P7)} have been designed and synthesized. The polymers are prepared in moderate yields by WCl6-Ph4Sn and possess high molecular weights (m w up to 64000). The structures and properties of the polymers are characterized and evaluated by NmR, TGA, UV, CD, PL, and EL analyses. All the polymers are thermally stable and their temperatures for 5% weight loss locate in the range of 300 to 416°C under nitrogen. The energy band gaps of all the polymers are ～3.0 eV. Polymers P4 and P5 show CD absorptions associated with the helicity of the polymer segments. Excitation of the THF solutions of P3–P7 by UV irradiation gives strong blue lights of ～485 nm with quantum yields higher than 20%. The thin films of the polymers also emit in the same spectral region, indicative of little aggregation-caused quenching of light emission. multilayer EL devices with a configuration of ITO/Polymer:PVK/BCP/Alq3/LiF/Al are constructed, which emit blue lights of ～487 nm. The maximum luminance and external quantum efficiency vary with the pendant groups, with P6 exhibiting the highest external quantum efficiency of 0.16%. The spectra stability of the EL devices is outstanding and the EL peak maximum experiences little change with the applied voltage.

The control of gene expression by transcriptional regulators and other types of functional y relevant DNA transactions such as chromatin remodeling and replication underlie a vast spectrum of biological processes in al organisms. DNA transactions require the control ed interaction of proteins with DNA sequence motifs which are often located in nucleosome-depleted regions （NDRs） of the chromatin. Formaldehyde-assisted isolation of regulatory elements （FAIRE） has been established as an easy-to-implement method for the isolation of NDRs from a number of eukaryotic organisms, and it has been successful y employed for the discovery of new regulatory segments in genomic DNA from, for example, yeast, Drosophila, and humans. Until today, however, FAIRE has only rarely been employed in plant research and currently no detailed FAIRE protocol for plants has been published. Here, we provide a step-by-step FAIRE protocol for NDR discovery in Arabidopsis thaliana. We demonstrate that NDRs isolated from plant chromatin are readily amenable to quantitative polymerase chain reaction and next-generation sequencing. Only minor modification of the FAIRE protocol wil be needed to adapt it to other plants, thus facilitating the global inventory of regulatory regions across species.

Grain quality is an important agricultural trait that is mainly determined by grain size and ***, we characterize the role of the rice transcription factor （TF） SALT-RESPONSIVE ERF1 （SERF1） during grain develop-ment. Through genome-wide expression profiling and chromatin immunoprecipitation, we found that SERF1 directlyregulates RICE PROLAmIN-BOX BINDING FACTOR （RPBF）, a TF that functions as a positive regulator of grain filling. Lossof SERF1 enhances RPBF expression resulting in larger grains with increased starch content, while SERF1 overexpressionrepresses RPBF resulting in smaller grains. Consistently, during grain filling, starch biosynthesis genes such as GRANULE-BOUND STARCH SYNTHASEI （GBSSI）, STARCH SYNTHASEI （SSI）, SSIIla, and ADP-GLUCOSE PYROPHOSPHORYLASELARGE SUBUNIT2 （AGPL2） are up-regulated in SERF1 knockout grains. moreover, SERF1 is a direct upstream regulator ofGBSSI. In addition, SERF1 negatively regulates germination by controlling RPBF expression, which mediates the gibberel-lic acid （GA）-induced expression of RICE AmYLASEIA （RAmylA）. Loss of SERF1 results in more rapid seedling establish-ment, while SERF1 overexpression has the opposite effect. Our study reveals that SERF1 represents a negative regulatorof grain filling and seedling establishment by timing the expression of RPBF.
