Rational design of oral drugs targeting mucosa delivery with gut organoid platforms

Effective oral drugs and vaccines require high delivery efficiency across the gastrointestinal epithelia and protection of medically effective payloads(i.e.,immunogens)against gastric *** this study,hollowed nanocarriers(NCs:silica nanospheres and gold nanocages)with poly-l-lysine(Pll)coating and mammalian orthoreovirus cell attachment proteinσ1 functionalization(NC-Pll-σ1)were explored as functional oral drug delivery vehicles(ODDVs).The transport of these ODDVs to mucosal lymphoid tissues could be facilitated by microfold cells(M-cells)mediated transcytosis(viaσ1-α2–3-linked sialic acids adherence)across gastrointestinal *** coating provided protection and slow-release of rhodamine 6 G(R6G),a model *** transport effectiveness of these ODDVs was tested on intestinal organoid monolayers in *** compared with other experimental groups,the fully functionalized ODDV system(with Pll-σ1)demonstrated two significant advantages:a significantly higher transport efficiency(198%over blank control at 48 h);and protection of payloads which led to both better transport efficiency and extended-release of payloads(61%over uncoated carriers at 48 h).In addition,it was shown that the M cell presence in intestinal organoid monolayers(modulated by Rank l stimulation)was a determining factor on the transport efficiency of the ODDVs:more M-cells(induced by higher Rank l)in the organoid monolayers led to higher transport efficiency for ODDV-delivered model payload(R6G).The fully functionalized ODDVs showed great potential as effective oral delivery vehicles for drugs and vaccines.

Superconducting Electronics Research at CSIRO Australia——20 Years after Discovery of HTS

CSIRO has had a long-term research effort in superconductivity, in particular, since the discovery of HTS which promised big prospects. Significant progress has been made in research and development of HTS electronic devices and systems for practical applications such as mineral and exploration as well as some niche applications in emerging science and technology areas. This article presents an overview of the CSIRO research activities in HTS supercon- ducting electronics since 1987, outlining the HTS junction and device technology as well as various application systems developed by the group.

Identification of the Mamestra configurata （lepidoptera Noctuidae） peritrophic matrix proteins and enzymes involved in peritrophic matrix chitin metabolism

The peritrophic matrix （PM） is essential for insect digestive system physiol- ogy as it protects the midgut epithelium from damage by food particles, pathogens, and toxins. The PM is also an attractive target for development of new pest control strategies due to its per os accessibility. To understand how the PM performs these functions, the molecular architecture of the PM was examined using genomic and proteomic approaches in Mamestra configurata （lepidoptera： Noctuidae）, a major pest of cruciferous oilseed crops in North America. liquid chromatography-tandem mass spectrometry analyses of the PM identified 82 proteins classified as：（i） peritrophins, including a new class with a CBDIII domain; （ii） enzymes involved in chitin modification （chitin deacetylases）, di- gestion （serine proteases, aminopeptidases, carboxypeptidases, lipases and ^-amylase） or other reactions （/^-l,3-glucanase, alkaline phosphatase, dsRNase, astacin, pantetheinase）; （iii） a heterogenous group consisting of polycalin, REPATs, serpin, C-Type lectin and lsti99/lsti201 and 3 novel proteins without known orthologs. The genes encoding PM proteins were expressed predominantly in the midgut, cDNAs encoding chitin synthase-2 （McCHS-2）, chitinase （McCHI）, and fl-N-acetylglucosaminidase （McNAG） enzymes, in- volved in PM chitin metabolism, were also identified. McCHS-2 expression was specific to the midgut indicating that it is responsible for chitin synthesis in the PM, the only chitinous material in the midgut. In contrast, the genes encoding the chitinolytic enzymes were expressed in multiple tissues. McCHS-2, McCHI, and McNAG were expressed in the midgut of feeding larvae, and NAG activity was present in the PM. This information was used to generate an updated model of the lepidopteran PM architecture.

Use of Synteny Conversion in Identification of Candidate Genes for Somitogenesis in Humans

Understanding the genetic component of scoliosis in humans has relied on the assumption that spine development is conserved across species. Since evolutionary conserved genes tend to lie within synteny blocks (HSBs) and genes which are not conserved lie within evolutionary breakpoint regions (EBRs), HSB analysis may be used to determine if spine development is conserved across species. We hypothesized that vertebral patterning genes are conserved in amniotes and their location is within stable or “syntenic” regions of chromosomes. Seventy seven patterning genes involved in Fgf, Wnt and Notch signaling pathways were analyzed to determine their location within HSBs or EBRs in the genomes of several amniotic species. The human genome was divided into 1 Mbp intervals and a comparison was made to determine whether these genes were preferentially localized within HSBs or EBRs associated with rapid evolution. The results indicate that genes associated with somite development in humans are preferentially located away from the EBRs: 0.014 genes in EBRs on genome average vs. 0.030 on average in other parts of the genome (p-value = 0.01). The concentration of vertebral patterning genes in HSBs, provides evidence that developmental pathways involved in vertebral morphogenesis are likely conserved across amniotes, consistent with their known function. These data support prior observations indicating that gene networks associated with major developmental processes such as neuronal, central nervous system, bone and blood vessel development, some mediated by Wnt and Notch signaling pathways, were less likely to be localized at EBRs.
