Impact of Drug-Drug Interaction between CDK4/6 Inhibitors and Proton Pump Inhibitors on Survival Outcomes in the Treatment of Metastatic Breast Cancer—Real World Data from a Portuguese Center

Introduction: Proton pump inhibitors (PPi) are widely prescribed, including in patients undergoing treatment for advanced breast cancer (ABC). Due to the pharmacokinetic characteristics of the CDK4/6 inhibitor (Ci) palbociclib a drug interaction with PPi was hypothesized. It was shown in a retrospective study that this association was an independent predictive factor for worse progression-free survival (PFS). Objective: To verify the impact of concomitant administration of PPi with Ci on overall survival (OS) and PFS. Material and Methods: This is a retrospective cohort study of patients treated with Ci for HR+HER2-ABC in the period from Feb/2017 to Aug/2020. SPSS software was used for data processing. Univariate analysis was done by the Kaplan-Meier method and log-rank test, and multivariate analysis by COX regression. P-value < 0.05 was considered significant. Results: 80 patients were included. The median age at diagnosis of ABC was 56 years (25 - 75). Treatment with Ci was 1st line for ABC in 68.8%. Choice of Ci was palbociclib in 73.8% (n = 59) and ribociclib in 26.3% (n = 21). The hormone partner was a nonsteroidal aromatase inhibitor in 45.0%, and fulvestrant in 55.0% of cases. 37.5% of patients were on PPi, and 70.0% of them were during the entire treatment (23.3% omeprazole, 73.4% pantoprazole, 3.3% others). Patients taking concomitant PPi and Ci had lower OS (OS-3 years 42.6% vs. 63.4%, p = 0.254) and PFS (PFS med 15 m. vs. 21 m., p = 0.733), although with no statistically significant difference. Discussion: In the sample, there was a numerical difference, without the statistical significance in the use of PPi in the survival of patients under Ci. This difference could be more evident with a longer follow-up and a larger sample size. This study intends to alert to the growing importance of checking for drug interactions. Hematologic malignancies are the most common type of cancer affecting children and young adults,and encompass diseases,such as leukemia,lymphoma,and myeloma,all of which impact blood associated tissues such as the bone marrow,lymphatic system,and blood *** diagnostics of these malignancies relies heavily on the use of bone marrow samples,which is painful,debilitating,and not free from risks for leukemia *** biopsies are based on minimally invasive assessment of markers in the blood(and other fluids)and have the potential to improve the efficacy of diagnostic/therapeutic strategies in leukemia patients,providing a useful tool for the real time molecular profiling of *** most promising noninvasive biomarkers are circulating tumor cells,circulating tumor DNA,microRNAs,and ***,we discuss the role of assessing these circulating biomarkers for the understanding of tumor progression and metastasis,tumor progression dynamics through treatment and for follow-up.

Background Congenital heart diseases(CHD)are the most common type of birth defects,affecting millions of newborns every year;no prevalence data are available in S?o Paulo State,Brazil's most populous *** objective is to identify trends in prevalence and risk factors for CHD in Sao Paulo State,*** We developed a population-based study to estimate the CHD trend of prevalence in recent years,stratified by maternal age and geographical clusters,using all cases of CHD identified by the Live Births Information System(SINASC-Sistema de Informa??o Sobre Nascidos Vivos)from January 1,2005,to December 31,*** calculated the trends of prevalence,the regression coefficient(β),the annual percent change(APC),and 95%confidence interval using the Prais-Winsten regression model,with the Durbin-Watson *** We found 10,594 cases of CHD among 8,536,101 live births(LB),a prevalence of 12.4/10,000 *** was no difference in the sex distribution;they are primarily Caucasian(60.2%),75.2%born at term,and 74.4%weight>2500 g,66.9%of births were by cesarean section.S?o Paulo State presented an increasing CHD trend of prevalence(APC=18.9%).The highest CHD prevalence rate was in mothers aged≥35 years(22.2/10,000 LB).There were 12,271 specific congenital heart defects among 10,594 patients(1.16 CHD/patient).Atrial septal defect has the highest number of cases(3835),with a prevalence of 4.49/10,000 LB,corresponding to 31.3%of all *** CHD had an increasing prevalence trend in recent years,being highest in S?o Paulo City and≥35-year mothers.

Polyphenol Oxidase Activity and Colour Changes of ‘Starking’ Apple Cubes Coated with Alginate and Dehydrated with Air

The objective was to study the effect of alginate coating on polyphenol oxidase (PPO) activity and colour of ‘Starking’ apple cubes during dehydration with hot air. Apple cubes were dehydrated at 20oC, 35oC, or 40oC, with a parallel airflow. Analysis of PPO activity, colour (L*, a*, b*) and dry matter were performed along the dehydration process at each temperature. All samples presented a peak in relative PPO activity in the beginning of the drying. Exponential models fitted well the experimental data after the peak. Cubes without coating presented lower PPO activity, suggesting lower browning, than coated samples throughout the dehydration process, for all temperatures. Better results for coated samples were obtained with a perpendicular airflow drying at 40oC, after dipping the whole apple in water at 60oC for 10 min. In order to prevent coated samples from browning, drying by perpendicular airflow preceded by a thermal treatment of the whole apple is required.

Objective： The present study aimed to evaluate the analgesic and anti-inflammatory effects of far infrared-emitting ceramics （cFIRs） in a model of persistent inflammatory hyperalgesia and to elucidate the possible mechanisms of these effects. Methods： Mice were injected with complete Freund＇s adjuvant （CFA） and treated with cFIRs via place- ment on a pad impregnated with cFIRs on the bottom of the housing unit for different periods of time. Mice underwent mechanical hyperalgesia and edema assessments, and tumor necrosis factor-α （TNF-α）, interleukin-1β （IL-1β） and IL-10 levels were measured. Twenty-four hours after CFA injection and 30 min before cFIR treatment, mice were pretreated with a nonselective adenosinergic antagonist, caffeine, the selective adenosine receptor A antagonist, 1,3-dipropyl-8-cyclopentylxanthine （DPCPX）, the selective cannabinoid receptor type 1 antagonist, AM281, the selective cannabinoid receptor type 2 antagonist, AM630, or the nonselective opioid receptor antagonist, naloxone, and mechanical hyperalge- sia was assessed. Results： cFIRs statistically （P 〈 0.05） decreased CFA-induced mechanical hyperalgesia （82.86 ±5.21）% in control group vs （56.67±9.54）% in cFIR group） and edema （（1699.0 ± 77.8） gm in control group vs （988.7±107.6） gm in cFIR group）, cFIRs statistically （P 〈 0.05） reduced TNF-α （0.478± 0.072） pg/mg of protein in control group vs （0.273 ±0.055） pg/mg of protein in cFIR group） and IL-113 （（95.81 ± 3.95） pg/mg of protein in control group vs （80.61 ±4.71）pg/mg of protein in cFIR group） levels and statistically （P〈 0.05） increased IL-10 （（18.32 ±0.78） pg/mg of protein in control group vs （25.89 ±1.23） pg/mg of protein in cFIR group） levels in post-CFA-injected paws. Organizational Culture and Favorable Environment for the Development of Innovations

The objective of the research dialogues with recent studies on the presence of a favorable environment for innovation while respecting and guided by theoretical perspectives that were built by classic authors in this field of research, such as Geert Hofstede and Edgar Schein. 258 questionnaires were answered and the data obtained were analyzed using structural equation modeling (SEM) after tabulation in statistical analysis software. The research model is quantitative-descriptive and is based on the methodology developed by MIRP and later adapted by other authors. As a result of this research, it was identified that employees perceive the culture of the analyzed organization with a high level of collectivism and high cultural congruence. It was found that employees have a perception that there is a small power distance. At the end of the research, it is possible to affirm that there is a relationship between the organizational culture and the favorable environment for the development of innovations in the analyzed company.

The geographical extent of Brazil exceeds 8.5 million km2 and encompasses a complex mix of biomes and other environmental *** decision support tools are needed to help support management of these diverse Brazilian natural resources including ecohydrological *** use of the Soil and Water Assessment Tool(SWAT)ecohydrological watershed-scale model in Brazil has increased greatly during the past *** over 100 SWAT studies were identified in this review which have been published during 1999 to 2015 in Brazilian and international journals,conference proceedings,and as theses or dissertations,many of which are written in *** majority of these studies(102 total)are reviewed here as part of an extensive survey covering the 1999 to 2013 time *** and spatial distributions,a summary of hydrologic calibration and validation results and a synopsis of the types of applications that were performed are reported for the surveyed studies.A smaller subset of recent Brazilian studies published in English between 2012 and 2015 in scientific journals are also reviewed,with emphasis on hydrologic and sediment transport testing results as well as scenario applications that were *** majority of the surveyed SWAT studies was performed for watersheds located in the South and Southeast regions of Brazil(67%)and was conducted in the context of academic *** 50%of the surveyed studies reported only hydrologic *** trends were found for the subset of more recent English *** studies have been reported that describe applications of SWAT in Brazil by private firms or government agencies;this review indicates that the potential exists for increased numbers of such studies in the ***,there is evidence that a lack of accessibility to adequate quality input data is a possible hindrance to the more general use of SWAT for watershed applications in Brazil.
