Studies of plants have been instrumental for revealing how new species *** several decades,botanical research has complemented and,in some cases,challenged concepts on speciation developed via the study of other organisms while also revealing additional ways in which species can ***,the ability to sequence genomes at an unprecedented pace and scale has allowed biologists to settle decades-long debates and tackle other emerging challenges in speciation ***,we review these recent genome-enabled developments in plant *** discuss complications related to identification of reproductive isolation(RI)loci using analyses of the landscape of genomic divergence and highlight the important role that structural variants have in speciation,as increasingly revealed by new sequencing ***,we review how genomics has advanced what we know of some routes to new species formation,like hybridization or whole-genome duplication,while casting doubt on others,like population bottlenecks and genetic *** genomics can fast-track identification of genes and mutations that confer RI,we emphasize that follow-up molecular and field experiments remain ***,genomics has clarified the outsized role of ancient variants rather than new mutations,particularly early during *** conclude by highlighting promising avenues of future *** include expanding what we know so far about the role of epigenetic and structural changes during speciation,broadening the scope and taxonomic breadth of plant speciation genomics studies,and synthesizing information from extensive genomic data that have already been generated by the plant speciation community.

Quantification and discovery of PCR inhibitors found in food matrices commonly associated with foodborne viruses

Human norovirus is the leading cause of foodborne illness *** and quantification of norovirus commonly involves the use of reverse transcriptase quantitative polymerase chain reaction(RT-qPCR);however,the presence of inhibitory compounds in foods limit detection and accurate *** some studies have been done on PCR inhibitors from foods,many of them are over a decade old and do not investigate inhibition in contemporary one-step RT-qPCR-based detection *** purpose of this work was to quantify the degree of inhibition that occurs from inhibitory compounds found in produce(pectin)and mollusks(hemocyanin,glycogen)—foods commonly associated with norovirus ***-qPCR reactions containing different amounts of genomic bacteriophage MS2 RNA,a norovirus surrogate,were spiked with different concentrations of pectin(0.0625%–0.25%w/V),glycogen(1.25%–10%),and hemocyanin(0.0625%–0.25%).Past research has implicated glycogen as an inhibitory compound in oysters;however,even high levels of glycogen(10%)had no significant effect(P>0.05)on ***,both pectin and hemocyanin caused complete inhibition at 0.25%,with no significant inhibition observed at 0.0625%(P<0.05).Hemocyanin is abundant in the hemolymph of mollusks and previously untested as a PCR *** work demonstrates that pectin and hemocyanin should be considered when testing produce and mollusk samples with PCR-based methods.

Forest composition and red oak (Quercus sp.) response to elevation gradients across greentree reservoirs

Elevation gradients within forested wetlands have long been recognized for their role in defining species composition through factors such as hydrology and soil *** reservoirs(GTRs)are leveeimpounded tracts of bottomland hardwood forest flooded throughout the winter months to provide habitat for overwintering *** flooding of GTRs alters the forest composition due to flood frequency,depth,and duration in combination with slight changes in *** evaluate the effect of elevation gradients,soil properties,and management techniques in the overstory species composition and red oak(Quercus spp.)species abundance,we inventoried 662 plots across 12 independent GTRs in eastern *** the lower elevations ranging from 50.98 to 54.99 m above sea level,the importance value index(IVI)was highest for nuttall oak(Quercus texana)and overcup oak(Quercus lyrata),whereas IVI shifted to cherrybark oak(Quercus pagoda)in the higher elevations ranging from 54.99 to 58.00 *** diversity did not differ by elevation gradient,soil property,or management technique within *** diversity,using non-metric multi-dimensional scaling(NMDS)analysis,indicated site-specific variability significantly correlated with the environmental predictors,including elevation(R^(2)=0.57),easting(R^(2)=0.47),soil texture(R^(2)=0.21),and pH(R^(2)=0.12).Red oak species-specific mixed-effects modeling of abundance response using Poisson distribution suggested an inverse correlation of nuttall oak and a direct correlation of cherrybark oak abundance with ***,willow oak(Quercus phellos)abundance was not significantly affected by elevation but was by silt loam soil texture and restoration management *** findings will aid management efforts to reduce the dominance of less desirable species that are prominent under specific environmental conditions and promote the dominance of more desirable *** GTR sustainability is increas

Interrelationship of soil moisture and temperature to sylvatic plague cycle among prairie dogs in the Western United States

Plague,caused by Yersinia pestis,is a flea-borne disease that is endemic in areas throughout the world due to its successful maintenance in a sylvatic cycle,mainly in areas with temperate *** rodents are thought to play a key role in the enzootic maintenance as well as epizootic outbreaks of *** the United States,prairie dogs(Cynomys),rodents(Muridae),and ground squirrels(Spermophilus)are susceptible to infection and are parasitized by fleas that transmit *** particular,prairie dogs can experience outbreaks that rapidly spread,which can lead to extirpation of colonies.A number of ecological parameters,including climate,are associated with these *** this study,we asked whether soil parameters,primarily moisture and temperature,are associated with outbreaks of plague in black-tailed prairie dogs and Gunnison’s prairie dogs in the Western United States,and at what depth these associations were *** collected publicly available county-level information on the occurrence of population declines or colony extirpation,while historical soil data was collected from SCAN and USCRN stations in counties and states where prairie dogs have been *** analysis suggests that soil moisture at lower depths correlates with colony die-offs,in addition to temperature near the surface,with key differences within the landscape ecology that impact the occurrence of ***,the model suggests that the burrow environment may play a significant role in the epizootic spread of disease amongst black-tailed and Gunnison’s prairie dogs.

The fly factor phenomenon is mediated by interkingdom signaling between bacterial symbionts and their blow fly hosts

We tested the recent hypothesis that the"fly factor"phenomenon(food cur-rently or previously fed on by flies attracts more flies than the same type of food kept inccessible to flies)is mediated by bacterial symbionts deposited with feees or regur-gitated by feeding *** allowed laboratory-reared black blow flies,Phormia regina(Meigen),to feed and de fecate on bacterial Luria-Bertani medium solidified with agar,and isolated seven morphologically distinct bacterial *** identified these us-ing matrix-assisted laser desorption/ionization mass spectrometry and sequencing of the 165 rRNA *** two-choice laboratory experiments,traps baited with cultures of Pro-teus mirabilis Hauser,Morganella morganii *** Jensen,or Serratia marcescens Bizio,captured significantly more flies than corresponding control jars baited with tryptic soy agar only.A mixture of seven bacterial strains as a trap bait was more attractive to flies than a single bacterial isolate(***).In a field experiment,traps baited with agar cultures of P:mirabilis and M.m siboni in combination captured significantly more flies than lraps baited with either bacterial isolate alone or the agar *** evident by gas chromatography-mass spectrometry,the odor profiles of bacterial isolates differ,which may explain the additive effect of bacteria to the attractiveness of bacterial trap ***"generalist bacteria,"P mirabilis and *** growing on animal protein(beef liver)or plant protein(tofu)are similarly effective in attracting ***-derived airborne semiochemicals appear to mediate foraging by flies and to inform their feeding and oviposition decisions.
