Effects of purposeful soccer heading on circulating small extracellular vesicle concentration and cargo

Background:Considering the potential cumulative effects of repetitive head impact(HI)exposure,we need sensitive biomarkers to track shortand long-term *** small extracellular vesicles(sEVs)(traffic biological molecules throughout the body and may have diagnostic value as biomarkers for *** purpose of this study was to identify the microRNA(miRNA)profile in circulating sEVs derived from human plasma following repetitive HI ***:Healthy adult(aged 18-35 years)soccer players were randomly assigned to one of 3 groups:the HI group performed 10 standing headers,the leg impact group performed 10 soccer ball trapping maneuvers over 10 min,and the control group did not participate in any soccer *** was collected before testing and 24 h afterward,and sEVs were isolated and characterized via nanoparticle tracking ***-generation sequencing was utilized to identify candidate miRNAs isolated from sEVs,and candidate microRNAs were analyzed via quantitative polymerase chain *** silico target prediction was performed using targetScan(Version 7.0;***)and miRWalk(http://***/)programs,and target validation was performed using luciferase reporter vectors with a miR-7844-5p mimic in human embryonic kidney(HEK)293t/17 ***:Plasma sEV concentration and size were not affected across time and group following repetitive HI *** 24 h,the HI read count from next-generation sequencing showed a 4-fold or greater increase in miR-92b-5p,miR-423-5p,and miR-24-3p and a 3-fold or greater decrease in miR-7844-5p,miR-144-5p,miR-221-5p,and *** of quantitative polymerase chain reaction revealed that leg impact did not alter the candidate miRNA *** our knowledge,miR-7844-5p is a previously unknown *** identified 8 miR-7844-5p mRNA targets:protein phosphatase 1 regulatory inhibitor subunit 1B(PPP1R1B),LIM and senescent cell antigen-like domains 1(LIMS1),au

Scavenging of reactive oxygen and nitrogen species with nanomaterials

Reactive oxygen and nitrogen species （RONS） are essential for normal physiological processes and play important roles in cell signaling, immunity, and tissue homeostasis. However, excess radical species are implicated in the development and augmented pathogenesis of various diseases. Several antioxidants may restore the chemical balance, but their use is limited by disappointing results of clinical trials. Nanoparticles are an attractive therapeutic alternative because they can change the biodistribution profile of antioxidants, and possess intrinsic ability to scavenge RONS. Herein, we review the types of RONS, how they are implicated in several diseases, and the types of nanoparticles with inherent antioxidant capability, their mechanisms of action, and their biological applications.

Knowing what native trees can recr uit on degraded areas allows selecting the best species to restore these ***,as this information is not often available,experimentation is required before large-scale *** study used ex situ experiments to make these decisions on *** with r-strategist plants,excessive solar radiation and water shortage commonly impair tree recruitment in open *** experiments focused on the interactions among these factors and were conducted with three pioneer species from seasonally dry forests of northwest Argentina,Anadenanthera colubrina,Ceiba chodatii and Jacaranda *** of each species were sown at two light levels(sunlight/shade),two rainfall levels(full/reduced)and two levels of interspecific competition(with/without competitor)in a tree *** emergence and survival were monitored over a year and the results indicate that species differentially respond to varying levels of light and *** of *** tolerated water shortages under elevated solar radiation,which are desirable features for forest *** of *** tolerated shade and drought,suggesting that they require shading for establishing in open ***,*** seedlings neither tolerated full sunlight nor water shortages,suggesting that this species requires shading and regular watering if used in *** of the effects of light and water,the survival of all species was reduced by interspecific *** results highlight the importance of experimentation for selecting the best species for forest restoration and can enhance the cost/benefit ratios of these actions.

road to stemness in hepatocellular carcinoma

Carcinogenic process has been proposed to relay on the capacity to induce local tissue damage and proliferative repair. Liver has a great regeneration capacity and currently, most studies point towards the dominant role of hepatocytes in regeneration at all levels of liver damage. the most frequent liver cancer is hepatocellular carcinoma(HCC). Historical findings originally led to the idea that the cell of origin of HCC might be a progenitor cell. However, current linage tracing studies put the progenitor hypothesis of HCC origin into question. In agreement with their dominant role in liver regeneration, mature hepatocytes are emerging as the cell of origin of HCC, although, the specific hepatocyte subpopulation of origin is yet to be determined. the relationship between the cancer cell of origin(CCO) and cancer-propagating cells, known as hepatic cancer stem cell(HCSC) is unknown. It has been challenging to identify the definitive phenotypic marker of HCSC, probably due to the existence of different cancer stem cells(CSC) subpopulations with different functions within HCC. there is a dynamic interconversion among different CSCs, and between CSC and non-CSCs. Because of that, CSC-state is currently defined as a description of a highly adaptable and dynamic intrinsic property of tumor cells, instead of a static subpopulation of a tumor. Altered conditions could trigger the gain of stemness, some of them include: EMt-MEt, epigenetics, microenvironment and selective stimulus such as chemotherapy. this CSC heterogeneity and dynamism makes them out reach from therapeutic protocols directed to a single target. A further avenue of research in this line will be to uncover mechanisms that trigger this interconversion of cell populations within tumors and target it.

this work presents the preliminary results of the first field calibration campaign performed in the Atacama Desert,Chile,between the 18 and 22 August 2014,called the Atacama Field Campaign(AtAFIC 2014).In situ measurements were performed in order to spectrally characterize the surface reflectance spectra between 0.3 and 2.5μm,radiometric temperature(8.0–14.0μm)and atmospheric measurements.A soil sample was collected and analyzed using Fourier transform Infrared Spectroscopy and X-Ray Diffraction techniques to characterize the surface reflectance spectra and mineralogical composition,*** land surface emissivity in addition to GOES,MODIS and Landsat-8 land surface temperature(LSt)were also *** showed that the spectral features of the Atacama soil and the characteristics of this geographical zone,which is featured as the most hyper-arid and cloudless place in the world,make this area a potential target for surface reflectance *** and night LSt comparison between field and remote sensing data are lower than 2 K and the Root Mean Square Error for land surface emissivity is close to 2%.this work opens the possibilities to consider the Atacama Desert as a reference target for calibration and validation activities for earth observation missions’purposes.

Beneﬁcial microbes can improve soil health by promoting soil structure,nutrient cycling,and disease *** addition,a wide array of rhizospheric microbes are responsible for producing metabolically active compounds including various types of plant growth ***,microbial biodiversity studies could contribute to the improvement of agricultural practices in deprived areas,such as the Pampean semiarid *** vast majority of studies conducted on endophytic microorganisms have focused on intensive crop legume *** contrast,little attention has been paid to microorganisms of native legumes,whose ecology is not directly affected by human *** this study,endophytic microorganisms isolated from root nodules of a selected native legume of the genus Rhynchosia were *** isolates were studied with a focus on their plant growth-pro-moting rhizobacteria(PGPR)*** the edaphic characteristics of the Pampean semiarid region,the isolates obtained were evaluated for their ability to grow under three salt stress conditions(50,100,and 200 mM NaCl)and four different pH values(6,7,8,and 9).Based on their PGPR activities,the selected strains were phylogenetically grouped using *** results showed great variability among the isolates in terms of the characteristics *** legumes manifested a wide endophytic variability and remarkable perfor-mance in PGPR *** conclude that they could be used as potential bioinoculants for legume cultivation,an excellent alternative to the use of chemical fertilizers that currently pollute the environment.

Autophagy is an essential cellular homeostatic mechanism by which intracellular components are delivered into the lysosomes for degradation and *** has been related with a diversity of pathological or physiological dentary processes such as bone remodeling,skeletal aging,osteoclastogenesis,osteoblastogenesis and different types of oral ***-rich plasma(PRP),isolated from autologous blood,is a plasma preparation containing a higher concentration of platelets which contains numerous different growth factors and cytokines that activate several cellular signaling *** purpose of this study is to investigate the effect of PRP on autophagy stimulation in both osteoblast precursor 3t3-L1 and non-related osteoblastic *** results showed that PRP can increase the number of autophagic structures in 3t3-L1 and HeLa(cervical cancer cells)***,we have determined by Western blot a rise in the lipidated form of the autophagic protein LC3(***3-II)upon PRP *** together,our results suggest that PRP is able to induce a strongly autophagy response in osteoblast precursor and,to a lesser extent,in non-related osteoblastic cells,suggesting that PRP could be a potential therapeutic tool for some autophagy-related diseases associated with bone homeostasis.
