Peripheral nerve injuries induce a severe motor and sensory deficit. Since the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, including the use of tubular nerve guidance conduits(tnGCs). However, the use of tnGCs results in poor functional recovery and central necrosis of the regenerating tissue, which limits their application to short nerve lesion defects(typically shorter than 3 cm). Given the importance of vascularization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tnGC would help eliminate necrotic processes and lead to improved regeneration. In this study, we reported the application of macroscopic holes into the tubular walls of silk-based tnGCs and compared the various features of these improved silk^(+) tnGCs with the tubes without holes(silk^(–) tnGCs) and autologous nerve transplants in an 8-mm sciatic nerve defect in rats. Using a combination of micro-computed tomography and histological analyses, we were able to prove that the use of silk^(+) tnGCs induced the growth of blood vessels from the adjacent tissue to the intraluminal neovascular formation. A significantly higher number of blood vessels in the silk^(+) group was found compared with autologous nerve transplants and silk^(–), accompanied by improved axon regeneration at the distal coaptation point compared with the silk^(–) tnGCs at 7 weeks postoperatively. In the 15-mm(critical size) sciatic nerve defect model, we again observed a distinct ingrowth of blood vessels through the tubular walls of silk^(+) tnGCs, but without improved functional recovery at 12 weeks postoperatively. Our data proves that macroporous tnGCs increase the vascular supply of regenerating nerves and facilitate improved axonal regeneration in a short-defect model but not in a critical-size defect model. This work aimed to determine the effect of native probiotic administration on body weight gain, nutritional characteristics and fatty acid profile of meat of creole goats. Equally distributed forty postweaned goats (75 days old) divided into control (n=20) and probiotic (n=20) groups were fed ad libitum for 60 days with native pasture and commercial diet. Lactobacillus reuteri DDL19, Lactobacillus alimentarius DDL48, Enterococcus faecium DDE39, and Bifidobacterium bifidum DDBA mixed culture (108 CFU/ml, 1:1:1:1 ratio) were daily orally delivered to the probiotic group. At the end of the study, muscle samples (Longissimus dorsi, LD, and Bicep femoris, BF) were analyzed for their chemical composition (protein, fat, moisture, pH, water-holding capacity, cholesterol) and fatty acid profile. Probiotic supplementation significantly increased body weight gain, reduced the incidence of diarrhea and improved the fatty acid profile of goat meat. Probiotic-fed animals exhibited increased monounsaturated and polyunsaturated fatty acids and a significant decrease in saturated fatty acid content. Conjugated linoleic acid content was significantly higher in probiotics than in the control group (P ndexes and a significant decrease in the n-6/n-3 ratio (~2.50), atherogenic (~0.30) and thrombogenic (~0.75) indexes. In addition, increases in desirable fatty acids (~79.40 g/100 g of FAME) and nutritional value index (~2.86) were observed compared to control groups. The native mix probiotic could be used as growth promoters to reduce the antibiotics, and to improve its derived-foods.

Times Series Applied to Study Vitamin D Seasonality in Argentina

In this study, we analyze how vitamin D (VD) serum levels flow with latitude and throughout seasons of the year within a population sample over three years, taking into account that VD is mainly photosynthesized in the skin from sun exposure. Vitamin D levels have been measured in 80,763 patients during 2013, 2014, and 2015. To accomplish the objectives, we first perform some inference tests like two-way Analysis of Variance (AnOVA) followed by post-hoc tests. Secondly, we develop time series techniques including cross correlation calculations. Least than 10% of the sample had healthy VD levels, which should be a fact of public health major concern. The effect of the interaction between the two factors, zones and seasons, was proved by AnOVA. The mean values which are significantly different were determined by post hoc test. Furthermore, we find that mean serum VD levels, measured as 25-hydroxy-VD, follow a seasonal lag pattern of 9 weeks, a delay for minimum and maximum values after the respective equinoxes and daily sunlight duration. Reliable estimates of the population are provided in the present study, since one of the strengths is its huge sample size. We have quantitatively characterized the seasonality of serum vitamin D levels in the Argentine and the seasonal lag pattern has been determined for the study region.

Preliminary examination of early neuroconnectivity features in the R6/1 mouse model of Huntington's disease by ultra-high field diffusion MRI

During the last decades,advances in the understanding of genetic,cellular,and microstructural alterations associated to Huntington's disease(HD)have improved the understanding of this progressive and fatal ***,events related to early neuropathological events,neuroinflammation,deterioration of neuronal connectivity and compensatory mechanisms still remain vastly ***-high field diffusion MRI(UHFD-MRI)techniques can contribute to a more comprehensive analysis of the early microstructural changes observed in *** addition,it is possible to evaluate if early imaging microstructural parameters might be linked to histological ***,qualitative studies analyzing histological complexity in brain areas susceptible to neurodegeneration could provide information on inflammatory events,compensatory increase of neuroconnectivity and mechanisms of brain repair and *** application of ultra-high field diffusion-MRI technology in animal models,particularly the R6/1 mice(a common preclinical mammalian model of HD),provide the opportunity to analyze alterations in a physiologically intact model of the *** some disparities in volumetric changes across different brain structures between preclinical and clinical models has been documented,further application of different diffusion MRI techniques used in combination like diffusion tensor imaging,and neurite orientation dispersion and density imaging have proved effective in characterizing early parameters associated to alteration in water diffusion exchange within intracellular and extracellular compartments in brain white and grey ***,the combination of diffusion MRI imaging techniques and more complex neuropathological analysis could accelerate the discovery of new imaging biomarkers and the early diagnosis and neuromonitoring of patients affected with HD.

Multiple sclerosis(MS)is a complex chronic disease of the central nervous system(CnS)which includes three main anatomopathological characteristics:inflammation,demyelination,*** pathology induces many clinical symptoms,depending on the site of the lesions,such as motor,sensitive,visual,urinary and cognitive *** addition,MS is a very heterogeneous disease that displays different clinical courses or phenotypes(relapsing/remitting MS,primary progressive MS(PPMS)and secondary progressive MS(SPMS).Both neurodegeneration and inflammation are present along the natural history of the ***,the balance of both phenomena varies according to the course of the *** relapsing/remitting MS form,the acute inflammation,mainly adaptive immunity,predominates over the neurodegeneration,being the main cause of the *** SPMS and PPMS,the neurodegeneration predominates over the inflammation,which appears as chronic inflammation,being innate immunity(microglia and astroglia)the main feature in the neurodegenerative process.

Laparoscopic complete mesocolic excision: West meets East

Complete mesocolic excision is a relatively new concept in western literature. It follows the same concept of total mesorectal excision and units’ routinely performing complete mesocolic excisions have good pathological results as well as good improvements in overall survival, disease free survival and local recurrence. And yet unlike total mesorectal excision, uptake in the West has been relatively slow with many units sceptical of the true benefits gained by taking up a more technically challenging and potentially more morbid procedure when there is a paucity of literature to support these claims. This article reviews complete mesocolic excision for colon cancer, attempting to identify the risks and benefits of the technique and particularly looking at the reasons why its uptake has not been universal. It also discusses the similarities of a complete mesocolic excision to a colon resection with a D3 lymphadenectomy as well as the role of a laparoscopic approach to this technique. Considering a D3 lymphadenectomy has been the standard of care for stage II and III colon cancers in many of our Asian neighbours for over 20 years, combining this data with data on complete mesocolic excision may provide enough evidence to support or refute the need for complete mesocolic excisions. Maybe there might be lessons to be learnt from our colleagues in the east.
