Divergence and constraint in the thermal sensitivity of aquatic insect swimming performance

Environmental temperature variation may play a significant role in the adaptive evolutionary divergence of ectotherm thermal performance curves(TPCs).However,divergence in TPCs may also be constrained due to various ***,we measured TPCs for swimming velocity of temperate and tropical mayflies(Family:Baetidae)and their stonefly predators(Family:Perlidae)from different *** predicted that differences in seasonal climatic regimes would drive divergence in TPCs between temperate and tropical *** tropical temperatures should favor the evolution of"specialists"that perform well across a narrow range of ***,variable temperatures in temperate zones,however,should favor"generalists"that perform well across a broad range of *** phylogenetically paired comparisons of mayflies and stoneflies,swimming speed was generally unaffected by experimental temperature and did not differ among populations between latitudes,suggesting a maintenance of performance breadth across elevation and *** exception was found between temperate and tropical mayflies at low elevation where climatic differences between latitudes are *** addition,TPCs did not differ between mayflies and their stonefly predators,except at tropical low *** results indicate that divergence in TPCs may be con strai ned in aquatic in sects except under the most differe nt ther・mal regimes,perhaps because of trade-offs that reduce thermal sensitivity and increase performance breadth.

An electropenetrography waveform library for the probing and ingestion behaviors of Culex tarsalis on human hands

Carbon storage and net primary productivity in Canadian boreal mixedwood stands

Canadian boreal mixedwood forests are extensive,with large potential for carbon sequestration and storage;thus,knowledge of their carbon stocks at different stand ages is needed to adapt forest management practices to help meet climate-change mitigation *** stocks were quantified at three Ontario boreal mixedwood sites.A harvested stand,a juvenile stand replanted with spruce seedlings and a mature stand had total carbon stocks(±SE)of 133±13 at age 2,130±13 at age 25,and 207±15 Mg C ha^-1 at age 81 *** the clear-cut site,stocks were reduced by about 40%or 90 Mg C ha^-1 at *** held 27,34 and 62%of stocks,while detritus held 34,29 and 13%of stocks at age 2,25 and 81,*** soil carbon stocks averaged 51 Mg C ha^-1,and held 38,37 and 25%of *** net primary productivity(±SE)in the harvested and juvenile stand was 2.1±0.2 and 3.7±0.3 Mg C ha^-1 per annum(p.a.),compared to 2.6±2.5 Mg C ha^-1 *** the mature *** mature canopies studied had typical boreal mixedwood composition and mean carbon densities of 208 Mg C ha^-1,which is above average for managed Canadian boreal forest ecosystems.A comparison of published results from Canadian boreal forest ecosystems showed that carbon stocks in mixedwood stands are typically higher than coniferous stands at all ages,which was also true for stocks in vegetation and ***,aboveground net primary productivity was typically found to be higher in mixedwood than in coniferous boreal forest stands over a range of *** from this study,together with those published from the other boreal forest stands demonstrate the potential for enhanced carbon sequestration through modified forest management practices to take advantage of Canadian boreal mixedwood stand characteristics.

Induction of Bone Matrix Protein Expression by Native Bone Matrix Proteins in C2C12 Culture

Objective To study the expression of bone matrix protein （BMP） induced by bovine bone morphogenetic proteins （BMPs） in vitro. Methods Type 1 collagen, osteopontin （OPN）, osteonectin （ON）, osteocalcin （OC）, and bone sialoprotein （BSP） were detected by immunohistochemistry in C2C12 cultured from day 1 to day 28. Results The signaling of bone matrix protein expression became weaker except for type I collagen, OC and BSP after 5 days. Fourteen days after culture, the positive signaling of type I collagen, OPN, ON, OC, and BSP was gradually declined, and could be detected significantly as compared with that of the negative control on day 28. BMP assay showed that the lkaline phosphatase （ALP） activity was higher in C2C12 culture than in the control during the 14-day culture. Also, total protein and DNA significantly increased during the 14-day culture. High levels of ALP were seen in preosteoblasts and osteoblsts in vivo and in differentiating osteoblasts in vitro. ALP was well recognized as a marker reflecting osteoblastic activity. Conclusion Native bovine BMP induces conversion of myoblasts into osteoblasts, produces type I collagen, and plays significantly role in osteoinduction and bone matrix mineralization of C2C 12 in vitro.

Molecular therapeutic strategies targeting pancreatic cancer induced cachexia

Pancreatic cancer(PC) induced cachexia is a complex metabolic syndrome associated with significantly increased morbidity and mortality and reduced quality of life. The pathophysiology of cachexia is complex and poorly understood. Many molecular signaling pathways are involved in PC and cachexia. Though our understanding of cancer cachexia is growing, therapeutic options remain limited. Thus, further discovery and investigation of the molecular signaling pathways involved in the pathophysiology of cachexia can be applied to development of targeted therapies. This review focuses on three main pathophysiologic processes implicated in the development and progression of cachexia in PC, as well as their utility in the discovery of novel targeted therapies. Skeletal muscle wasting is the most prominent pathophysiologic anomaly in cachectic patients and driven by multiple regulatory pathways. Several known molecular pathways that mediate muscle wasting and cachexia include transforming growth factor-beta(TGF-β), myostatin and activin, IGF-1/PI3 k/AkT, and JAk-STAT signaling. TGF-β antagonism in cachectic mice reduces skeletal muscle catabolism and weight loss, while improving overall survival. Myostatin/activin inhibition has a great therapeutic potential since it plays an essential role in skeletal muscle regulation. Overexpression of insulin-like growth factor binding protein-3(IGFBP-3) leads to increased ubiquitination associated proteolysis, inhibition of myogenesis, and decreased muscle mass in PC induced cachexia. IGFBP-3 antagonism alleviates muscle cell *** component of cachexia is profound systemic inflammation driven by pro-cachectic cytokines such as interleukin-6(IL-6), tumor necrosis factoralpha(TNF-α), and interferon gamma(INF-γ). IL-6 antagonism has been shown to reduce inflammation, reduce skeletal muscle loss, and ameliorate cachexia. Pb-Sn mixed perovskites are becoming increasingly popular as narrowbandgap(1.2–1.3 eV)light absorbers in single-junction perovskite solar cells(PSCs)and as bottom cells for all-perovskite tandem solar cells,for highefficiency,low-cost,lightweight,roll-to-roll printable photovoltaic(PV)*** the first report of planar Pb:Sn mixed PSCs in 2014,the power conversion efficiencies(PCE)have increased from 10%to 21%by the end of 2020 with an exponential growth in research conducted in this *** much effort,the performance and stability of Pb-Sn mixed PSCs are still limited,which constrains their long-term use in all-perovskite tandem *** review highlights the avenues explored in improving different aspects of Pb-Sn mixed PSCs and provides a comprehensive discussion of the interdependent factors affecting the device *** includes compositional engineering of the perovskite crystal,absorber layer fabrication and crystallization methods,bandgap tuning,Sn4+reduction,and surface passivation of the absorber layer,as well as the selection of interlayers and electrodes of the final PSC.

Development of internal COI primers to improve and extend barcoding of fruit flies (Diptera: Tephritidae: Dacini)

Accurate species-level identifications underpin many aspects of basic and applied biology;however,identifications can be hampered by a lack of discriminating morphological characters,taxonomic expertise or *** approaches,such as DNA"barcoding"of the cytochrome c oxidase(COI)gene,are argued to overcome these ***,nuclear encoding of mitochondrial genes(numts)and poor amplification success of suboptimally preserved specimens can lead to erroneous *** insect group for which these molecular and morphological problems are significant are the dacine fruit flies(Diptera:Tephritidae:Dacini).We addressed these issues associated with COI barcoding in the dacines by first assessing several"universal"COI primers against public mitochondrial genome and numt sequences for dacine *** then modified a set of four primers that more closely matched true dacine COI sequence and amplified two overlapping portions of the COI barcode *** new primers were tested alongside universal primers on a selection of dacine species,including both fresh preserved and decades-old dry ***,Bactrocera tiyoni mitochondrial and nuclear genomes were compared to identify putative *** numt clades were identified,three of which were amplified using existing universal *** contrast,our new primers preferentially amplified the"true"mitochondrial COI barcode in all dacine species *** new primers also successfully amplified partial barcodes from dry specimens for which full length barcodes were *** we recommend these new primers be incorporated into the suites of primers used by diagnosticians and quarantine labs for the accurate identification of dacine species.
