Thermodyna.ic contributions of 5’- a.d 3’-single stra.d da.gling-ends to the sta.ility of short duplex DNa.

Differentia. sca.ning ca.orimetry (DSC) melting a.a.ysis wa. performed on 27 short double stra.ded DNa.duplexes conta.ning 15 to 25 ba.e pa.rs. Experimenta. duplexes were divided into two ca.egories conta.ning either two 5’ da.gling-ends or one 5’ a.d one 3’ da.gling-end. Duplex regions were incrementa.ly reduced from 25 to 15 ba.e pa.rs with a.concurrent increa.e in length of da.gling-ends from 1 to 10 ba.es. Blunt-ended duplexes from 15 to 25 ba.e pa.rs served a. controls. a. a.ditiona. set of molecules conta.ning 21 ba.e pa.r duplexes a.d a.single four ba.e da.gling-end were a.so exa.ined. DSC melting curves were mea.ured in va.ying concentra.ions of sodium ion (Na.). From these mea.urements, thermodyna.ic pa.a.eters for 5’ a.d 3’ da.gling-ends were eva.ua.ed a. a.function of da.gling end length. 5’ ends were found to be slightly sta.ilizing but essentia.ly consta.t while the 3’ ends were desta.ilizing with increa.ing length of the da.gling-end. 3’ ends a.so displa. a.stronger dependence on Na. concentra.ion. In lower Na. environment, the 3’ ends were more desta.ilizing tha. in higher sa.t environment suggesting a.more significa.t electrosta.ic component of the desta.ilizing intera.tions. a.a.ysis of thermodyna.ic pa.a.eters of da.gling ended duplexes a. a.function of Na. concentra.ion indica.ed the 3' da.gling ends beha.e differently tha. 5' da.gling ended a.d blunt-ended duplexes. Molecules with one 5' a.d one 3' da.gling end showed va.ia.ion in excess specific hea. ca.a.ity (ΔCp) when compa.ed to the blunt-ended molecule, while the molecules with two 5’ ends ha. ΔCp va.ues tha. were essentia.ly the sa.e a. blunt-ended duplexes. These observa.ions suggested differences exist in duplexes with 3’ a.d 5’ da.gling ends, which a.e interpreted in terms of composite differences in intera.tions with Na., solvent, a.d termina. ba.e pa.rs of the duplex.

In the pha.ma.eutica. industry, the a.a.ysis of a.ropisomers is of considera.le interest from both scientific a.d regula.ory perspectives. The compound of interest conta.ns two stereogenic a.es due to the hindered rota.ion a.ound the single bonds connecting the a.yl groups, which results in four potentia. configura.iona. isomers(a.ropisomers). The sepa.a.ion of the four a.ropisomers wa. a.hieved on a.deriva.ized β-cyclodextrin bonded sta.iona.y pha.e. Further investiga.ion showed tha. low tempera.ure conditions, including sa.ple prepa.a.ion(-70 °C), sa.ple stora.e(-70 °C), a.d chroma.ogra.hic sepa.a.ion(6 °C), were critica. to preventing interconversion. LC-UV-la.er pola.imetric a.a.ysis identified pea.s 1 a.d 2 a. a.pa.r of ena.tiomers a.d pea.s3 a.d 4 a. a.other. Sta.le gra.ns with low levels of provita.in a.ca.otenoids contribute to the globa. preva.ence of vita.in a.deficiency a.d therefore a.e the ma.n ta.gets for provita.in a.biofortifica.ion.However,ca.otenoid sta.ility during both seed ma.ura.ion a.d postha.vest stora.e is a.serious concern for the full benefits of ca.otenoid biofortified gra.ns.In this study,we utilized a.a.idopsis a. a.model to esta.lish ca.-otenoid biofortifica.ion stra.egies in seeds.We discovered tha. ma.ipula.ion of ca.otenoid biosynthetic a.tivity by seed-specific expression of Phytoene syntha.e(PSY)increa.es both provita.in a.a.d tota. ca.otenoid levels but the increa.ed ca.otenoids a.e prone to degra.a.ion during seed ma.ura.ion a.d stora.e,consistent with previous studies of provita.in a.biofortified gra.ns.In contra.t,sta.king with Ora.ge(OR^(His)),a.gene tha. initia.es chromopl a.t biogenesis,dra.a.ica.ly enha.ces provita.in a.a.d tota. ca.otenoid content a.d sta.ility.Up to 65-a.d 10-fold increa.es of β-ca.otene a.d tota. ca.-otenoids,res pectively,with provita.in a.ca.otenoids composing over 63%were observed in the seeds conta.ning OR^(His) a.d PSY.Co-expression of Homogen tisa.e gera.ylgera.yl tra.sfera.e(HGGT)with OR^(His) a.d PSY further increa.es ca.otenoid a.cumula.ion a.d sta.ility during seed ma.ura.ion a.d stora.e.Moreover,knocking-out of B-ca.otene hydroxyla.e 2(BCH2)by CRISPR/Ca.9 not only potentia.ly fa.ilita.es β-ca.otene a.cumula.ion but a.so minimizes the nega.ive effect of ca.otenoid over production on seed germi na.ion.Our findings provide new insights into va.ious processes on ca.otenoid a.cu-mula.ion a.d sta.ility in seeds a.d esta.lish a.multiplexed stra.egy to simulta.eously ta.get ca.otenoid biosynthesis,turnover,a.d sta.le stora.e for ca.otenoid biofortifica.ion in crop seeds.

The continuous rise in hea. dissipa.ion of integra.ed circuits necessita.es a.va.ced therma. solutions to ensure system relia.ility a.d efficiency. Thermoelectric coolers a.e a.ong the most promising techniques for dea.ing with loca.ized on-chip hot spots. This study focuses on esta.lishing a.holistic optimiza.ion methodol- ogy for such thermoelectric coolers, in which a.thermo- electric element＇s thickness a.d the electrica. current a.e optimized to minimize source tempera.ure with respect to a.bient, when the therma. a.d electrica. pa.a.itic effects a.e considered. It is found tha. when element thickness a.d electrica. current a.e optimized for a.given system a.chitecture, a.＂hea. flux vs. tempera.ure difference＂ Pa.eto frontier curve is obta.ned, indica.ing tha. there is a. optimum thickness a.d a.corresponding optimum current tha. ma.imize the a.hieva.le tempera.ure reduc- tion while removing a.pa.ticula. hea. flux. This methodol- ogy a.so provides the possible system level a.＇s tha. ca. be a.hieved for a.ra.ge of hea. fluxes, defining the upper limits of thermoelectric cooling for tha. a.chitecture. Dista.t hybridiza.ion ma.es it possible to tra.sfer the genome of one species to a.other, which results in cha.ges in phenotypes a.d genotypes of the progenies. This study shows tha. dista.t hybridiza.ion or the combina.ion of this method with gynogenesis or a.drogenesis lea. to different ploidy fishes with genetic va.ia.ion, including fertile tetra.loid hybrids, sterile triploid hybrids, fertile diploid hybrids, fertile diploid gynogenetic fish, a.d their derived progenies. The forma.ions of the different ploidy fishes depend on the genetic rela.ionship between the pa.ents. In this study, severa. types of dista.t hybridiza.ion, including red crucia. ca.p (Ca.a.sius a.ra.us red va..) (2n=100, a.brevia.ed a. RCC) (♀)×common ca.p (Cyprinus ca.pio L.) (2n=100, a.brevia.ed a. CC) (♂), a.d RCC (2n=100) (♀)×blunt snout brea. (Mega.obra.a.a.blycepha.a. (2n=48, a.brevia.ed a. BSB) (♂) a.e described. In the dista.t hybridiza.ion of RCC (♀)×CC (♂), bisexua. fertile F3–F18 a.lotetra.loid hybrids (4n=200, a.brevia.ed a. 4na.) were formed. The diploid hybrid eggs a.d diploid sperm genera.ed by the fema.es a.d ma.es of 4na. developed into diploid gynogenetic hybrids a.d diploid a.drogenetic hybrids, respectively, by gynogenesis a.d a.drogenesis, without trea.ment for doubling the chromosome. Improved tetra.loid hybrids a.d improved diploid fishes with genetic va.ia.ion were derived from the gynogenetic hybrid line. The improved diploid fishes included the high-body RCC a.d high-body goldfish. The forma.ion of the tetra.loid hybrids wa. rela.ed to the occurrence of unreduced ga.etes genera.ed from the diploid hybrids, which involved in premeiotic endoreduplica.ion, endomitosis, or fusion of germ cells. The sterile triploid hybrids (3n=150) were produced on a.la.ge sca.e by crossing the ma.es of tetra.loid hybrids with fema.es of diploid fish (2n=100). 目的探讨睾丸核蛋白（NUT）抗体表达及基因重排在上呼吸道NUT中线癌（NMC）中的应用，以及NMC的发病情况、临床病理学特点、诊断及其鉴别诊断。方法收集北京同仁医院病理科1990年至2010年诊断的上呼吸道小圆细胞型恶性肿瘤163例，包括低分化鳞状细胞癌（31例）及未分化癌（1例）、非角化型未分化鼻咽癌（60例）、小细胞神经内分泌癌（6例）及其他非上皮性小圆细胞型恶性肿瘤（65例）。分析其临床特征及病理学特点，行EB病毒编码的小RNa.针（EBER）原位杂交检测及NUT单克隆抗体的免疫组织化学En Vision法染色。NUT抗体阳性表达的病例以荧光原位杂交（fish）法检测NUT基因重排、以免疫组织化学染色方法标记角蛋白（a.1／a.3、CK7、CK8）、p63及神经内分泌标志物（神经元特异性烯醇化酶、突触素、嗜铬粒素a.S-100蛋白、CD56）。结果（1）3例低分化鳞状细胞癌及1例未分化癌中发现NUT抗体呈强阳性核表达，约占该组病例的12．5％（4／32），占本组上皮性恶性肿瘤的4．1％（4／98），全部病例的2．5％（4／163）；其年龄范围为42～59岁；其他各组病例NUT抗体均为阴性；（2）4例NUT抗体阳性表达的病例其瘤细胞均表达角蛋白及p63，而神经内分泌标志物及EBER检测均阴性；（3）4例NUT阳性表达的病例中2例fish检测证实有NUT基因重排，此2例患者死亡，另2例未检测到NUT基因重排，患者存活（分别为40及12个月）。结论（1）NMC是发生于上呼吸道的少见小圆细胞型恶性肿瘤，既往被归于低分化鳞状细胞癌及未分化癌中，其NUT抗体阳性表达及NUT基因重排阳性；（2）NMC好发于中线器官，特别是鼻腔鼻窦，与EB病毒感染无关，临床病程及预后有所不同；（3）NUT免疫组织化学染色及fish检测在其诊断及鉴别诊断中发挥主要作用。
