This paper aims to describe the development of laboratory animal science in China on the basis of historical evidence and recent national survey data,and to identify the problems facing the adoption of Three R *** authors undertook a national survey in 2006,by means of a questionnaire sent to 31 provinces,municipalities and autonomous regions,and also compared data from a variety of sources,including several national surveys and published paper’*** animal science in China has developed rapidly over the past 30 years,as a result of a combination of economic,governmental and societal *** than 100,000 people work in the field of laboratory animal science,in 2,000 laboratory animal centres,institutes,universities,organisations,and *** the year of our survey,more than 19 million laboratory animals were produced from 320 licensed production *** 16 million laboratory animals were used in animal experiments,in 1530 facilities licensed for their *** scale of the market for the supply and use of laboratory animals is huge,and thus it is very important to improve the level of adoption of these alternatives,in education,research and *** China,this presents a significant economic and technological opportunity in the field of biosciences *** concept of the Three Rs first appeared in China in the 1980s,when the scale of laboratory animal sciences was starting to *** the 1990s,the Three Rs concept became commonly accepted among laboratory animal scientists,and began to appear in government *** the first decade of the 21 st century,the Three Rs principles have become increasingly applied in our day-to-day *** further time is still needed to achieve the full application of the Three Rs principles,especially the adoption of Three R *** paper describes the achievements in China relating to laboratory animal science,the use of Three R alternatives, and animal welfare,

Background: The time-related decline in regenerative capacity and organ homeostasis is a major feature of aging. Rehmannia glutinosa and Astragalus membranaceus have been used as traditional Chinese herbal medicines for enhanced immunity and prolonged life. However, the mechanism by which this herbal medicine slows aging is unknown. In this study, we investigated the mechanism of the herbal anti-aging ***: Mice were fed diets supplemented with R. glutinosa or A. membranaceus for 10 months; the control group was fed a standard diet. The phenotypes were evaluated using a grading score system and survival analysis. The percentages of the senescence phenotypes of hematopoietic stem cells(HSCs) were determined by fluorescence-activated cell sorting analysis. The function and the mechanism of HSCs were analyzed by clonogenic assay and the real-time polymerase chain ***: The anti-aging effect of R. glutinosa is due to the enhanced function of HSCs. Mice fed with R. glutinosa displayed characteristics of a slowed aging process,including decreased senescence and increased rate of survival. Flow cytometry analysis showed decreased numbers of Lin–Sca1^+c-kit–(LSK) cells, long-term HSCs(LT-HSCs) and short-term HSCs(ST-HSCs) in the R. glutinosa group. In vitro, clonogenic assays showed increased self-renewal ability of LT-HSCs from the R. glutinosa group as well as maintaining LSK quiescence through upregulated p18 expression. The R. glutinosa group also showed decreased reactive oxygen species levels and the percentage of β-gal^+ cells through downregulation of the cellular senescence-associated protein p53 and ***: Rehmannia glutinosa exerts anti-aging effects by maintaining the quiescence and decreasing the senescence of HSCs.

Revascularization and angiogenesis,as substrates of sustained collateral circulation,play a crucial role in determining the severity and clinical outcome of acute ischemic stroke(AIS)due to large vessel occlusion(LVO).Developing an adjunct biomarker to help identify and monitor collateral status would aid stroke diagnosis and *** screen the potential biomarkers,proteomic analysis was performed in this study to identify those distinct plasma protein profiles in AIS due to LVO with different collateral ***,we found that levels of Plasma Factor VII Activating Protease(FSAP)significantly increased in those AIS patients with poor collaterals,and were correlated with worse neurological ***,both in vitro and in vivo models of ischemic stroke were used to explore pathological mechanisms of FSAP in endothelial *** demonstrated that the FSAP inhibitor,high-molecular-weight hyaluronan(HMW-HA),enhanced the pro-angiogenic vascular factors,improved the integrity of brain blood barrier,and promoted newly formed cerebral microvessels in the ischemic penumbra,consequently improving neurological *** elucidate the pathways that might contribute to revascularization during LVO,we applied transcriptomic analysis via unbiased RNA sequencing and showed that Wnt signaling was highly involved in FSAP mediated endothelial ***,inhibition of Wnt5a largely reversed the protective effects from HMW-HA treatment,implying that FSAP might aggravate endothelial dysfunction and neurological deficits by regulating Wnt5a ***,FSAP may represent a potential biomarker for collateral status after LVO and a promising therapeutic target to be explored in the treatment of stroke.

Dear Editor,Coronavirus disease 2019(COVID-19)is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,which induces multiple cardiovascular complications including acute myocardial injury resulting from acute coronary syndrome,myocarditis,stress-cardiomyopathy,arrhythmias,cardiogenic shock,and cardiac *** incidence of myocardial injury in COVID-19 patients was up to 20%or higher in the subpopulation with existing cardiovascular diseases,accounting for 69.4%of mortality.1 Although the presence of angiotensin-converting enzyme 2(ACE2)in cardiomyocytes could result in viral myocarditis in COVID-19 patients,the cardiopathology involved in COVID-19 is still unclear.

Enterovirus A71(EV-A71) causes major outbreaks of hand,foot,and mouth disease(HFMD) in many countries,most frequently affecting children,and a small proportion of cases may lead to ***,no vaccine is available in most endemic regions,and no licenced treatments for EV-A71 infection are ***,we characterize a human monoclonal antibody(Hu MAb),E1,by screening a Fab antibody phage library derived from patients who recovered from EV-A71 infection.E1 exhibits strong neutralizing activity against EV-A71 virus in *** cryo-electron microscopy(cryo-EM) structures of the EV-A71 virion in complex with E1 Fab fragments demonstrated that E1 recognized an epitope formed by residues in the BC and HI loops of *** a mouse model,E1 effectively protected against lethal EV-A71 challenge in both prophylactic and therapeutic *** particular,E1 significantly reduces virus titers and muscle damage.E1 might represent a potential adjunct to EV-A71 treatment.

Ischemic stroke is caused primarily by an interruption in cerebral blood flow,which induces severe neural injuries,and is one of the leading causes of death and disability ***,it is of great necessity to further detailly elucidate the mechanisms of ischemic stroke and find out new therapies against the *** recent years,efforts have been made to understand the pathophysiology of ischemic stroke,including cellular excitotoxicity,oxidative stress,cell death processes,and *** the meantime,a plethora of signaling pathways,either detrimental or neuroprotective,are also highly involved in the forementioned *** pathways are closely intertwined and form a complex signaling ***,these signaling pathways reveal therapeutic potential,as targeting these signaling pathways could possibly serve as therapeutic approaches against ischemic *** this review,we describe the signaling pathways involved in ischemic stroke and categorize them based on the pathophysiological processes they participate *** approaches targeting these signaling pathways,which are associated with the pathophysiology mentioned above,are also ***,clinical trials regarding ischemic stroke,which potentially target the pathophysiology and the signaling pathways involved,are summarized in ***,this review elucidated potential molecular mechanisms and related signaling pathways underlying ischemic stroke,and summarize the therapeutic approaches targeted various pathophysiology,with particular reference to clinical trials and future prospects for treating ischemic stroke.
