The aim of this study is to assess the effects of DNA methylation and historic acetylation, alone or in combination, on the expression of several tumor-associated genes and cell cycle progression in two established human colon cancer cell lines: Colo-320 and SW1116. Treatments with 5-aza-2’-deoxycytidine (5-aza-dC) and trichostatin A, alone or in combination, were applied respectively. The methylation status of the CDKN2A promoter was determined by methyla-tion-specific PCR, and the acetylated status of the histones associated with the p21WAF1 and CDKN2A genes was examined by chromatin immunoprecipitation. The expression of the CDKN2A, p21WAF1, p53, p73, APC, c-myc, c-Ki-ras and survivin genes was detected by real-time RT-PCR and RT-PCR. The cell cycle profile was established by flow cytometry. We found that along with the demethylation of the CDKN2A gene promoter in both cell lines induced by 5-aza-dC alone or in combination with TSA, the expression of both CDKN2A and APC genes increased. 目的:研究中老年患者不同部位脑微出血(CMBs)的相关危险因素。方法:通过中山大学孙逸仙纪念医院医学大数据平台收集自2022年1月至2023年5月的住院患者共669例患者,先分成CMBs阳性组(123例)和CMBs阴性组(546例),再将CMBs阳性组细分成仅脑叶组、仅深部组和广泛组三个亚组,分别通过对比任意的CMBs阳性亚组与CMBs阴性组的病史资料和实验室检查结果进行单因素分析。再采用Logistics回归分析各个脑微出血亚组的独立危险因素。结果:在单因素分析中筛选出了一些备选的危险因素,后纳入Logistics回归分析,结果显示,饮酒、高血压、MCV、HbA1C、Hb是脑微出血的独立危险因素,因不同亚组有所不同。结论:脑微出血的相关危险因素较多,且因分布部位而异,戒酒、控制血压、控糖将可能对脑微出血有重要预防和治疗价值。

We here report a genome-editing strategy to correct spinal muscular atrophy(SMA). Rather than directly targeting the pathogenic exonic mutations, our strategy employed Cas9 and guide-sg RNA for the targeted disruption of intronic splicing-regulatory elements. We disrupted intronic splicing silencers(ISSs, including ISS-N1 and ISS + 100) of survival motor neuron(SMN) 2, a key modifier gene of SMA, to enhance exon 7 inclusion and full-length SMN expression in SMA iPSCs. Survival of splicing-corrected iPSC-derived motor neurons was rescued with SMN restoration. Furthermore, co-injection of Cas9 mRNA from Streptococcus pyogenes(SpCas9) or Cas9 from Staphylococcus aureus(SaCas9) alongside their corresponding sgRNAs targeting ISS-N1 into zygotes rescued 56% and 100% of severe SMA transgenic mice(Smn-/-, SMN2tg/-).The median survival of the resulting mice was extended to >400 days. Collectively, our study provides proof-of-principle for a new strategy to therapeutically intervene in SMA and other RNA-splicing-related diseases.

Clinical and angiographic features associated with coronary collateralization in stable angina patients with chronic total occlusion

Objective： Coronary collateral circulation is an alternative source of blood supply to myocardium in the presence of advanced coronary artery disease. We sought to determine which clinical and angiographic variables are associated with collateral development in patients with stable angina and chronic total coronary occlusion. Methods： Demographic variables, biochemical measurements, and angiographic findings were collected from 478 patients with stable angina and chronic total coronary occlusion. The presence and extent of collaterals supplying the distal aspect of a total coronary occlusion from the contra-lateral vessel were graded from 0 to 3 according to the Rentrop scoring system. Results： Low （Rentrop score of 0 or 1） and high （Rentrop score of 2 or 3） coronary collateralizations were detected in 186 and 292 patients, respectively. Despite similar age, cigarette smoking, and medical treatment, patients with low collateralization were female in a higher proportion and less hypertensive, and had higher rates of type 2 diabetes and dyslipidemia than those with high coUateralization （for all comparisons, P〈0.05）. In addition, patients with low collateralization exhibited more single-vessel disease, less right coronary artery occlusion, more impaired renal function, and higher serum levels of high-sensitivity C-reactive protein （hsCRP） compared with those with high col- lateralization. Multivariate analysis revealed that age of 〉65 years, female gender, diabetes, no history of hypertension dyslipidemia, moderate to severe renal dysfunction, single-vessel disease, and elevated hsCRP levels were inde- pendently associated with low coronary collateralization. Conclusions： Coronary collateralization was reduced in almost 40% of stable angina patients with chronic total occlusion, which was related to clinical and angiographic factors. The impact of coronary collateralization on outcomes after revascularization needs further investigation.

The herbal medicine Tong luo Jiu Nao （TLJN） contains geniposide （GP） and ginsenoside Rgl at a molar ratio of i0：1. Rgl is the major component of another herbal medicine, panax notoginseng saponin （PNS）. TLJN has been shown to strengthen brain function in humans, and in animals it improves learning and memory. We have previously shown that TLJN reduces amyloi- dogenic processing in Alzheimer＇s disease （AD） mouse models. Together this suggests TLJN may be a potential treatment for patients with dementia. Because chronic damage of the central nervous system by formaldehyde （FA） has been presented as a risk factor for age-associated cognitive dysfunction, in the present study we investigated the protective effect of both TLJN and GP in neuron-like cells exposed to FA. FA-exposed murine N2a neuroblastoma cells were incubated with TLJN, its main in- gredient GP, as well as PNS, to measure cell viability and morphology, the rate of apoptosis and expression of genes encoding Akt, FOXO3, Bcl2 and p53. The CCK-8 assay, cytoskeletal staining and flow cytometry were used to test cell viability, mor- phology and apoptosis, respectively. Fluorescent quantitative real-time PCR （qRT-PCR） was used to monitor changes in gene expression, and HPLC to determine the rate of FA clearance. Treatment of N2a cells with 0.09 mmol L-1 FA for 24 h signifi- cantly reduced cell viability, changed cell morphology and promoted apoptosis. Both TLJN and GP conferred neuroprotection to FA-treated N2a cells, whereas PNS, which had to be used at lower concentrations because of its toxicity, did not. Our data demonstrate that TLJN can rescue neuronal damage caused by FA and that its main ingredient, GP, has a major role in this ef- ficacy. This presents purified GP as a drug or lead compound for the treatment of AD.

目的探究老年人网络使用与认知功能之间的关系。方法本文运用中国家庭追踪调查(CFPS)2020年数据，使用IADL量表、流动中心抑郁自评量表分别测量老年人失能状况和抑郁状况；采用二列相关分析相关性；构建多重中介效应模型，研究老年人使用网络对老年人认知功能影响的中介路径。结果在控制年龄、性别、婚姻状况、代际支持后，回归结果显示是否使用网络对老年人认知功能的直接影响显著(β=0.688,t=7.36,P<0.001);抑郁状况和失能状况显著影响老年人认知功能(β=-0.018,t=-2.69,P<0.05;β=-0.114,t=-4.83,P<0.001)。中介效应分析结果显示，抑郁状况和失能状况在网络使用和老年人认知功能之间的简单中介效应分别为0.044(95%CI:0.011 3～0.083 4)和0.040(95%CI:0.015 8～0.064 2),链式中介效应为0.006(95%CI:0.000 4～0.013 2)。结论抑郁状况和失能状况在是否使用网络和老年人认知功能之间存在中介效应，且抑郁和失能在是否使用网络影响老年人认知功能中存在链式中介效应。随着互联网的普及，应促进老年人互联网的使用，同时应注重老年人的身心健康，以此保护老年人认知功能，延缓或防止老年人认知障碍的发生。

Severe acute respiratory syndrome （SARS） is a life threatening, upper respiratory disease. Its cause is a coronavirus, SARS-CoV. Since its emergence in 2003 in China, SARS has affected more than 8000 patients and caused 776 deaths in 26 countries. A reemergence of SARS occurred in Guangdong province, in which the first case was confirmed on January 5, 2004 and three more reported the following month.

Series of 2-alkyl-1,3,4-trimethylbenzenes(ATMBs)were detected in most of crude oils and source rocks collected from various strata and locations of the Tarim *** appeared to have heavy carbon isotopic signatures(δ13C,up to~-16‰)compared to those hydrocarbons from oxygenic phototrophic organisms,indicating that the unequivocal source of green sulfur bacteria(GSB)and photic zone euxinia(PZE)existed in the original *** the high paleoproductivity,the PZE may have enhanced the preservation of organic matter,which triggered the formation of extremely organic-rich source rocks with TOC up to 29.8%for the Lower Cambrian Yuertus Formation(€1y).The coexistence of ATMBs and the diagnostic products from secondary alterations(e.g.,abundant 25-norhopanes,thiadiamondoids,and diamondoids)indicated a stronger ability of *** with the results of quantitatively de-convoluting mixed oil,the oil-source correlation based on ATMBs from a large number of Lower Paleozoic samples of the Tarim Basin suggested that the abundant deep crude oil resources co ntained a dominant contribution(>50%)from the€1y source ***,the ATMBs,as diagnostic biomarkers indicating unequivocal precursors under special habitat conditions,might provide important insights for the exploration of deep Lower Paleozoic crude oils in the Tarim Basin.
