Mitochondrial dysfunction is a significant pathological alte ration that occurs in Parkinson's disease(PD),and the Thr61.le(T61.)mutation in coiled-coil helix coiled-coil helix domain containing 2(CHCHD2),a crucial mitochondrial protein,has been reported to cause Parkinson's ***-ATPase participates in the synthesis of cellular adenosine triphosphate(ATP)and plays a central role in mitochondrial energy ***,the specific roles of wild-type(WT)CHCHD2 and T61.-mutant CHCHD2 in regulating F1.O-ATPase activity in Parkinson's disease,as well as whether CHCHD2 or CHCHD2 T61. affects mitochondrial function through regulating F1.O-ATPase activity,remain unclea ***,in this study,we expressed WT CHCHD2 and T61.-mutant CHCHD2 in an MPP^(+)-induced SH-SY5Y cell model of *** found that CHCHD2 protected mitochondria from developing MPP^(+)-induced *** normal conditions,ove rexpression of WT CHCHD2 promoted F1.O-ATPase assembly,while T61.-mutant CHCHD2 appeared to have lost the ability to regulate F1.O-ATPase *** addition,mass spectrometry and immunoprecipitation showed that there was an interaction between CHCHD2 and *** weeks after transfection with AAV-CHCHD2 T61.,we intraperitoneally injected 1.methyl-4-phenyl-1.2,3.6-tetrahydropyridine into mice to establish an animal model of chronic Parkinson's disease and found that exogenous expression of the mutant protein worsened the behavioral deficits and dopaminergic neurodegeneration seen in this *** findings suggest that WT CHCHD2 can alleviate mitochondrial dysfunction in PD by maintaining F1.0-ATPase structure and function.

Small-molecule photothermal agents(PTAs)with intense second near-infrared(NIR-Ⅱ,1.000 to 1.700 nm)absorption and high photothermal conversion efficiencies(PCEs)are promising candidates for treating deep-seated tumors such as *** date,the development of small-molecule NIR-Ⅱ PTAs has largely relied on fabricating donor–acceptor–donor(D–A–D/D′)structures and limited success has been ***,through acceptor engineering,a donor–acceptor–acceptor(D–A–A′)-structured NIR-Ⅱ aza-boron-dipyrromethene(aza-BODIPY)PTA(SW8)was readily developed for the 1.064-nm laser-mediated phototheranostic treatment of *** the donor groups to acceptor groups produced remarkable red-shifts of absorption maximums from first near-infrared(NIR-Ⅰ)regions(~808 nm)to NIR-Ⅱ ones(~1.064 nm)for aza-BODIPYs(SW1.to SW8).Furthermore,SW8 self-assembled into nanoparticles(SW8@NPs)with intense NIR-Ⅱ absorption and an ultrahigh PCE(75%,1.064 nm).This ultrahigh PCE primarily originated from an additional nonradiative decay pathway,which showed a 1.0-fold enhanced decay rate compared to that shown by conventional pathways such as internal conversion and vibrational ***,SW8@NPs performed highly efficient 1.064-nm laser-mediated NIR-Ⅱ photothermal therapy of osteosarcoma via concurrent apoptosis and *** work not only illustrates a remote approach for treating deep-seated tumors with high spatiotemporal control but also provides a new strategy for building high-performance small-molecule NIR-Ⅱ PTAs.

BACKGROUND: The pharmacological action of traditional Chinese medicine compound is the comprehensive effect of the various ingredients, and the interactions of various ingredients are closely correlated with the final effect. In order to reveal the compatibility mechanism of BHD's prescription in treating and preventing ischemic cerebrovascular disease, we needed explore the effect and relation of ingredients in the prescription. OBJECTIVE: To observe the effect of Buyang Huanwu decoction (BHD) and Astragalus mongholicus on the activity of platelet activating factor receptor (PAFR) in the platelet of rabbits in vitro, and investigate the mechanism of Astragalus mongholicus. DESIGN: A decomposed recipes study. SETTING: Guangzhou university of Traditional Chinese Medicine. MATERIALS: Five New Zealand rabbits, weighing 2-3.kg, both sexes, were used. BHD was composed of Sheng huang Qi 120 g, Dang Gui Wei 6 g, Chi Shao 4.5 g, Chuan Xiong 3.g, Di Long 3.g, Tao Ren 3.g, Hong Hua 3.g. The prescription for activating blood circulation consisted of Dang Gui Wei 6 g, Chi Shao 4.5 g, Chuan Xiong 3.g, Di Long 3.g, Tao Ren 3.g and Hong Hua 3.g. The prescription for invigorating qi consisted of 120 g Sheng huang Qi. The prepared herbal pieces were purchased from the traditional Chinese medicine Dispensary of Foshan Second People's Hospital, and appraised by Professor xu from Science of Chinese Materia Medica College, Guangzhou university of Traditional Chinese Medicine. 3.-PAF was supplied by Amersham Co., Ltd. (specific activity: 6. 475 TBq/mmol; batch number: 200402); PAF standard by Biomol Co., Ltd. (batch number: P13.8V). We report the discovery of a peculiar X-ray transient,EP240408a,by Einstein Probe （EP） and follow-up studies made with EP,Swift,NICER,GROND,ATCA and other ground-based multiwavelength *** new transient was first detected with Wide-field X-ray Telescope （WXT） on board EP on April 8th,2024,manifested in an intense yet brief X-ray flare lasting for *** flare reached a peak flux of 3.9×1.-9erg cm-2s-1.n 0.5-4 keV,3.0 times brighter than the underlying X-ray emission detected throughout the *** and more precise follow-up observations by EP/FXT,Swift and NICER confirmed the finding of this new *** X-ray spectrum is non-thermal in 0.5-1. keV,with a power-law photon index varying *** X-ray light curve shows a plateau lasting for4d,followed by a steep decay till becoming undetectable1. d after the initial *** on its temporal property and constraints from previous EP observations,an unusual timescale in the range of 7-23.d is found for EP240408a,which is intermediate between the commonly found fast and long-term *** counterparts have been found in optical and near-infrared,with the earliest observation at 1. h after the initial X-ray detection,suggestive of intrinsically weak emission in these *** demonstrate that the remarkable properties of EP240408a are inconsistent with any of the transient types known so far,by comparison with,in particular,jetted tidal disruption events,gamma-ray bursts,X-ray binaries and fast blue optical *** nature of EP240408a thus remains an *** suggest that EP240408a may represent a new type of transients with intermediate timescales of the order of1. *** detection and follow-ups of more of such objects are essential for revealing their origin.
