Aim:P-glycoprotein is localized at the apical brush-border membrane of the proxi-mal renal tubule and functions as extruding toxins and xenobiotics out of *** difference of P-glycoprotein’s function resulted from single nucleotide poly-morphisms in MDR1 (multidrug resistance gene encoding for P-gp) and may bethe cause of interindividual differences in susceptibility to end-stage renal dis-ease (ESRD).The purpose of this study is to compare the genotype frequency ofC3435T and G1199A polymorphisms in MDR1 between ESRD patients and healthycontrols in the Chinese population to determine whether the alteration of the P-gpfunction is associated with ***:Two hundred and eighty-four healthyChinese controls and 244 Chinese patients with ESRD were involved in this *** specific PCR and polymerase chain reaction-restriction fragment lengthpolymorphism assay were used to determine the genotype MDR1 G1199A andC3435T,***:The genotype distribution of 3435CC,3435CT,and 3435TT were 0.35,0.50,and 0.15,respectively,in the control group and 0.38,0.47,and 0.15 in the group with the ESRD *** variant allele 1199G>A wasfound in any of the *** value of serum creatinine for genotypes 3435CC,3435CT,and 3435TT in the ESRD patients were 753.8+276.0 μmol/L,849.6±342.2μmol/L,and 987.0±512.0 μmol/L,*** difference between 3435TTand 3435CC reached statistical significance (PA variant *** studies are needed to clarifythe cause and interindividual differences in the susceptibility for the risk of ESRD.

Objective:This cross-sectional study aimed to determine the epidemiology of olfactory and gustatory dysfunctions related to COVID-19 in ***:This study was conducted by 45 tertiary Grade-A hospitals in *** and offline questionnaire data were obtained from patients infected with COVID-19 between December 28,2022,and February 21,*** collected information included basic demographics,medical history,smoking and drinking history,vaccination history,changes in olfactory and gustatory functions before and after infection,and other postinfection symptoms,as well as the duration and improvement status of olfactory and gustatory ***:Complete questionnaires were obtained from 35,566 *** overall incidence of olfactory and taste dysfunction was 67.75%.Being female or being a cigarette smoker increased the likelihood of developing olfactory and taste *** received four doses of the vaccine or having good oral health or being a alcohol drinker decreased the risk of such *** infection,the average olfactory and taste VAS scores were 8.41 and 8.51,respectively;after infection,they decreased to 3.69 and 4.29 and recovered to 5.83 and 6.55 by the time of the *** median duration of dysosmia and dysgeusia was 15 and 12 days,respectively,with 0.5%of patients having symptoms lasting for more than 28 *** overall self-reported improvement rate was 59.16%.Recovery was higher in males,never smokers,those who received two or three vaccine doses,and those that had never experienced dental health issues,or chronic accompanying ***:The incidence of dysosmia and dysgeusia following infection with the SARS-CoV-2 virus is high in *** and prognosis are influenced by several factors,including sex,SARS-CoV-2 vaccination,history of head-facial trauma,nasal and oral health status,smoking and drinking history,and the persistence of accompanying symptoms.

Repair techniques for short-distance peripheral nerve defects, including adjacent joint flexion to reduce the distance between the nerve stump defects, ＂nerve splint＂ suturing, and nerve sle eve connection, have some disadvantages. Therefore, we designed a repair technique involving intraoperative tension-free application of a nerve elongator and obtained good outcomes in the repair of short-distance peripheral nerve defects in a previous animal study. The present study compared the clinical outcomes between the use of this nerve elongator and performance of the conventional method in the repair of short-distance transection injuries in human elbows. The 3-, 6-, and 12-month postoperative follow-up results demonstrated that early neurological function recovery was better in the nerve elongation group than in the conventional group, but no signif- icant difference in long-term neurological function recovery was detected between the two gro ups. In the nerve elongation group, the nerves were sutured without tension, and the duration of postoperative immobilization of the elbow was decreased. Elbow function rehabilitation was significantly better in the nerve elongation group than in the control group. Moreover, there were no security risks. The results of this study confirm that the use of this nerve elongator for repair of short-distance peripheral nerve defects is safe and effective.

AIM:To explore the effect of the Andrographis paniculata(***)polysaccharide on the proliferation and apoptosis of human retinoblastoma(RB)Y79 cells and its ***:The refined *** polysaccharide was obtained using techniques such as water extraction,ethanol precipitation,and decompression *** inhibition effect of the *** polysaccharide on the proliferation of Y79 cells was detected by cell proliferation *** cytometr y was used for the detection of cell apoptosis rate and cycle *** qunatitative polymerase chain reaction(RT q PCR)and Western blotting were used to detect the expression of cell apoptosis signal pathway-related factors(caspase-3,caspase-8,and caspase-9)and cell cycle signal pathwayrelated factors(CDK1 and cyclin B1)at the transcriptional and translational ***:Infrared and ultraviolet spectrum scanning showed that the extracted drug was a polysaccharide withhigh *** being treated with different concentrations of *** polysaccharide for different periods of time,the Y79 cells showed different degrees of proliferation *** cytometric observations showed that the cell apoptosis rate and the proportion of cells blocked in the G2/M phase were significantly increased after *** polysaccharide *** analysis revealed that the m RNA and protein expression of caspase-3,caspase-8,and caspase-9 in the *** polysaccharide treatment groups increased significantly compared with that in the control groups,while the expression of CDK1 and cyclin B1 decreased ***:The *** polysaccharide could inhibit the proliferation and induce apoptosis of Y79 *** possible mechanism is via the upregulation of caspase-3,caspase-8,and caspase-9 expression in the cell apoptotic signaling pathway and the downregulation of CDK1 and cyclin B1 expression in the cell cycle signaling pathway.

阵发性睡眠性血红蛋白尿症(paroxysmal nocturnal hemoglobinuria,PNH)是一种由于磷脂酰肌醇聚糖A基因突变引起细胞膜上锚链蛋白糖基磷脂酰肌醇(glycosylphosphatidylinositol,GPI)表达异常所致的后天获得性克隆性造血干细胞疾病,临床常表现为血管内溶血、反复血栓形成及骨髓衰竭,也可出现肾功能异常、肺动脉高压、吞咽困难、胸痛、腹痛、勃起功能障碍等多系统受累的症状。由于PNH为罕见病,且临床表现异质性强,常需进行多学科协作诊疗。北京协和医院依托罕见病诊疗平台,邀请多学科临床专家,在PNH诊疗方面达成了统一意见,并形成《阵发性睡眠性血红蛋白尿症多学科诊疗专家共识(2024)》,以期促进PNH诊疗的标准化、规范化。

AIM:To clarify whether Mycophenolic acid(MPA)or its metabolite AcMPAG can cause gastrointestinal disturbanc- es,and to explore the effect of UGT2B7 SNP 211G＞T on the pharmacokinetic of AcMPAG. METHODS:Twenty-four renal transplant patients were enrolled in this *** study was per- formed on day 14 after transplantation and symptoms were recorded on the same *** blood samples were collected before dosing and 0.5,1,1.5,2,4,6,8,10 and 12 hours after morning *** concentrations of MPA,MPAG and AcMPAG were detected by HPLC. Genotype of UGT2B7 211G＞T was determined using PCR-RFLP method. RESULTS:No significant difference was observed be- tween patients with and without side effects for AUC of MPA and *** values of AUC/dose of MPA,MPAG and AcMPAG were(39.7±12.3), (1063.7±646.5)and(10.2±3.0)ng·h·mL·mg in TT genotype group and(42.0±20.0),(1466.5± 683.3)and(20.7±5.6)ng·h·mL·mgin GG gen- otype group,*** ratio of AUC(AcMPAG)(0-12))/ AUCCMPA(0-12)),which was used to describe the change of UGT2B7 enzyme activity,was 0.49±0.20,0.34±0.26 and 0.28±0.14 in recipients with 211GG,GT and TT genotype,respectively(P＜0.05). CONCLUSION:There was no significant differences in pharmacokinetics of MPA and its metabolites between side effect group and non-side effect *** seems that the polymorphism 211G＞T in the UGT2B7 gene results in the reduction of enzyme acitivity.

In this study,microcomputer image processing and pattern recognition technology,and the knowledge of morphology and optical characteristics of Cryptococcus neoformans were used for identification of Cryptococcus *** groups of mice were lethally infected with standard strain,Wuhan strain,American B-2643 strain and *** of the Cryptococcus *** samplescollected included mice brain,lung,kidney,liver,small intestine tissue and were observed under a light *** than 600 images of the fungus were input into a microcomputer.A system of computer for automatic identification of the Cryptoccocus neoformans was developed. The technique involved image preprocessing,imagesegmenting,coding of line-length on the edge,curve fitting,extracting of image feature,building of image library and feature data bank etc..And then,768 images of the clinical samples and other fungus samples whose morphological features tend to be confused with Cryptococcus neoformans were input into microcomputer and subjected to automatic *** Cryptococcus neoformans was accurately identified within 15 min,and the consistence rate with results of routine culture was 98%.

目的探讨人成纤维细胞生长因子受体1(fibroblast growth factor receptor 1,FGFR1)基因表达抑制通过线粒体途径影响血管瘤内皮细胞(hemangioendo⁃thelial cells,HemECs)凋亡。方法将HemECs分为空白组(仅加入脂质体)、对照组(转染阴性对照siRNA)和实验组(转染靶向抑制FGFR1的siRNA)。转染48 h,用流式细胞术检测细胞凋亡率,用蛋白质印迹法检测FGFR1、B淋巴细胞瘤-2相关X蛋白(B lymphoma-2 related X protein,Bax)、B细胞淋巴瘤/白血病-X基因长片段(B cell lymphoma/leukemia-X gene long fragment,Bcl-xl)、半胱氨酸天冬氨酸蛋白水解酶3(cysteine aspartate proteolytic enzyme 3,Caspase-3)和Caspase-9蛋白的表达水平。结果干预细胞48 h,实验组、对照组和空白组的细胞凋亡率分别为(19.27±1.67)%,(3.47±0.21)%和(3.62±0.24)%,FGFR1蛋白相对表达量分别为0.07±0.01,0.56±0.05和0.53±0.04,Bax蛋白相对表达量分别为0.35±0.03,0.12±0.02和0.11±0.01,Bcl-xL蛋白相对表达量分别为0.13±0.02,0.50±0.05和0.49±0.05,Caspase-3相对表达量分别为0.37±0.04,0.12±0.01和0.12±0.01,Caspase-9相对表达量分别为0.26±0.03,0.10±0.01和0.10±0.01,实验组的上述指标与对照组和空白组比较,差异均有统计学意义(均P<0.05)。结论抑制FGFR1基因表达可通过线粒体途径影响HemECs凋亡。
