Serum proteins differentially expressed in gestational diabetes mellitus assessed using isobaric tag for relative and absolute quantitation proteomics

BACKGROUND As a well-known fact to the public,gestational diabetes mellitus(GDM)could bring serious risks for both pregnant women and *** this important investigation into the linkage between GDM patients and their altered expression in the serum,proteomics techniques were deployed to detect the differentially expressed proteins(DEPs)of in the serum of GDM patients to further explore its pathogenesis,and find out possible biomarkers to forecast GDM *** Subjects were divided into GDM and normal control groups according to the IADPSG diagnostic *** samples were randomly selected from four cases in each group at 24-28 wk of gestation,and the blood samples were identified by applying iTRAQ technology combined with liquid chromatography-tandem mass *** proteins and signaling pathways associated with GDM were identified by bioinformatics analysis,and the expression of key proteins in serum from 12 wk to 16 wk of gestation was further verified using enzyme-linked immunosorbent assay (EliSA).RESULTS Forty-seven proteins were significantly differentially expressed by analyzing the serum samples between the GDMgravidas as well as the healthy ones. Among them, 31 proteins were found to be upregulated notably and the rest16 proteins were downregulated remarkably. Bioinformatic data report revealed abnormal expression of proteinsassociated with lipid metabolism, coagulation cascade activation, complement system and inflammatory responsein the GDM group. EliSA results showed that the contents of RBP4, as well as ANGPTL8, increased in the serumof GDM gravidas compared with the healthy ones, and this change was found to initiate from 12 wk to 16 wk *** GDM symptoms may involve abnormalities in lipid metabolism, coagulation cascade activation, complementsystem and inflammatory response. RBP4 and ANGPTL8 are expected to be early predictors of GDM.

The cooperative visible-light mediated and NHC-catalyzed dearomative arylation-acylation of N-(2-bromobenzoyl)indoles with aldehydes was developed, affording the corresponding highly functionalized polycyclic indolines in moderate to good yields with exclusive diastereoselectivities. Mechanism investigations support the activation of CAr-Br bond via visible-light mediated energy transfer(EnT) and the following single-electron-transfer(SET) with Breslow intermediate under NHC catalysis to give key aryl radical and ketyl radical. This protocol demonstrates the cleavage of CAr-Br bond under mild cooperative EnT/NHC catalysis.

Cytochrome P45026A1 Contributes to the Maintenance of Neuropathic Pain

The cytochrome P450 proteins(CYP450s)have been implicated in catalyzing numerous important biological reactions and contribute to a variety of ***26A1,a member of the CYP450 family,carries out the oxidative metabolism of retinoic acid(RA),the active metabolite of vitamin *** we report that CYP26A1 was dramatically upregulated in the spinal cord after spinal nerve ligation(SNL).CYP26A1 was mainly expressed in spinal neurons and *** analysis displayed that the content of all-trans-RA(at-RA),the substrate of CYP26A1,was reduced in the spinal cord on day 7 after *** of CYP26A1 by siRNA or inhibition of CYP26A1-mediated at-RA catabolism by talarozole relieved the SNL-induced mechanical allodynia during the maintenance phase of neuropathic *** also reduced SNL-induced glial activation and proinflammatory cytokine production but increased anti-inflammatory cytokine(IL-10)*** RA receptors RARα,RXRβ,and RXRγwere expressed in spinal neurons and glial *** promoter of Il-10 has several binding sites for RA receptors,and at-RA directly increased Il-10 mRNA expression in ***,intrathecal IL-10 attenuated SNL-induced neuropathic pain and reduced the activation of astrocytes and ***,the inhibition of CYP26A1-mediated at-RA catabolism alleviates SNL-induced neuropathic pain by promoting the expression of IL-10 and suppressing glial ***26A1 may be a potential therapeutic target for the treatment of neuropathic pain.

High-purity germanium(HPGe)detectors,which are used for direct dark matter detection,have the advantages of a low threshold and excellent energy *** surface passivation of HPGe has become crucial for achieving an extremely low energy *** this study,first-principles simulations,passivation film preparation,and metal oxide semiconductor(MOS)capacitor characterization were combined to study surface *** calculations of the energy band structure of the -H,-OH,and -NH_(2) passivation groups on the surface of Ge were performed,and the interface state density and potential with five different passivation groups with N/O atomic ratios were accurately analyzed to obtain a stable surface *** on the theoretical calculation results,the surface passivation layers of the Ge_(2)ON_(2) film were prepared via magnetron sputtering in accordance with the optimum atomic ratio *** microstructure,C-V,and I-V electrical properties of the layers,and the passivation effect of the Al/Ge_(2)ON_(2)/Ge MOS were characterized to test the interface state *** mean interface state density obtained by the Terman method was 8.4×10^(11) cm^(-2) eV^(-1).The processing of germanium oxynitrogen passivation films is expected to be used in direct dark matter detection of the HPGe detector surface passivation technology to reduce the detector leakage currents.

Study on tungsten sintering temperature field based on electromagnetic simulation and computational fluid dynamics

In order to study the influence of various sintering variables more intuitively and clearly,electromagnetic field analysis and computational fl uid dynamics are used to analyze the infl uence of gas flow,heating power and other process parameters on the temperature range of medium-frequency sintering in this *** results indicate that the uniformity of temperature fi eld is determined by the gas flow rate and heating *** the heating power is 50 kW,the stable temperature drops about 10 K for every 1 m^(3)·h^(−1)increase of hydrogen *** peak value of the maximum temperature diff erence on the tungsten rods gradually increases with the increase of the hydrogen flow rate,and it appears slightly *** temperature distribution of tungsten rods in furnace is similar with diff erent hydrogen flow *** the same time,the closer to the furnace wall,the higher the temperature of the tungsten *** temperature diff erence between tungsten rods mainly occurred within 1 h after the beginning of *** the heating power can improve the stable temperature,but has little eff ect on the heating *** heating power can increase the peak value of the maximum temperature diff erence on the tungsten rods,but has little eff ect on the occurrence time of the peak value of the maximum temperature diff erence on the tungsten *** the heating power will increase the temperature diff erence of a single tungsten rod,but the temperature diff erence of a single tungsten rod never exceeds 65 *** study can be used to guide the process optimization *** with numerical simulation calculation,the sintering parameters with low energy consumption and high temperature uniformity can be quickly and eff ectively found,thus improving the microstructure and properties of sintered tungsten products.

感染翻修术后早期假体周围感染再清创的疗效分析

目的：分析清创保留假体联合抗生素（DAIR）治疗感染翻修术后早期假体周围感染（PJI）的临床效果。方法：回顾性分析2011年1月至2020年12月新疆医科大学第一附属医院收治的采用DAIR治疗104例早期PJI患者的临床资料。根据既往有无翻修手术史分为翻修手术组25例和初次手术组79例，两组患者的年龄、性别、感染部位、体重指数（BMI）、红细胞沉降率（ESR）、C反应蛋白（CRP）比较，差异均无统计学意义（P均>0.05）。比较两组患者的治疗成功率、致病菌、美国膝关节协会评分（KSS）、Harris髋关节评分（HHS）及术后并发症发生情况。结果：翻修手术组患者获得0.3～127.0个月随访，平均（51.9±43.7）个月；初次手术组患者获得0.6～131.0个月随访，平均（63.5±40.0）个月。末次随访时，翻修手术组的治疗成功率为56.0%(14/25)，低于初次手术组86.1%(68/79)(P0.05）。翻修手术组患者KSS和HHS分别由术前（36.2±8.1）分和（41.4±6.5）分提高至（64.7±11.7）分和（70.7±11.1）分（P均<0.001）。初次手术组患者KSS和HHS由术前（39.4±7.6）分和（44.0±7.2）分提高至（80.2±8.2）分和（81.7±6.7）分（P均<0.001）。翻修手术组患者术后KSS和HHS均低于初次手术组（P均<0.05）。翻修手术组患者术后并发症发生率为56.0%(14/25)，高于初次手术组的24.1%(19/79)(P<0.05)。结论：DAIR治疗既往有翻修手术史的早期PJI效果欠佳，而对于2次及以上翻修术后的早期PJI，则不建议使用DAIR治疗。

目的·总结以神经系统损伤为首发症状的2例原发性干燥综合征(primary Sjögren's syndrome,pSS)患者的临床特点并复习相关文献。方法·对以神经系统受累为首发症状的2例pSS患者的临床表现、实验室检查结果、影像学检查结果、唇腺活检结果、治疗效果等进行总结和分析。结果·病例1,女性,自50岁起出现进行性行走不稳伴下肢远端感觉障碍,表现为痉挛步态、感觉性共济失调、传导束性感觉障碍和二便障碍;58岁起无法独立行走。实验室检查提示抗核抗体阳性(1∶1000着丝点型),抗干燥综合征抗原A(SSA)/Ro-52抗体、抗SSA/Ro-60抗体、抗着丝点抗体阳性;唇腺活检可见唾液腺组织、导管间及小叶内淋巴细胞浸润灶2个/4 mm^(2)(淋巴细胞>50个)。头颅磁共振成像可见“蛇眼征”;肌电图显示右侧腓神经运动纤维轴索损伤。在院期间予以大剂量糖皮质激素冲击、联合免疫抑制剂治疗后明显好转。3个月后可推助行器行走,日常生活基本自理。病例2,女性,自81岁起出现颈部不自主右斜,既往有轻度口干、眼干、伴膝关节疼痛1年。实验室检查提示抗SSA/Ro-52抗体、抗SSA/Ro-60抗体阳性,血红蛋白86 g/L;唇腺活检可见小唾液腺组织部分腺泡萎缩,间质内见淋巴细胞、浆细胞浸润灶2个/4 mm^(2)(淋巴细胞>50个)。周围神经电生理检查结果提示右侧正中神经传导速度减慢。经免疫抑制剂、解痉、肌松药物治疗后症状较前好转。结论·以复杂型痉挛性截瘫样表现、颈部肌张力障碍为首发症状的病例拓展了pSS的临床表型谱;临床上应注意鉴别继发于pSS的神经系统损伤与其他原发神经系统疾病;对于具有无明显原因的神经系统损伤症状的患者,应当筛查相关自身免疫抗体。
