Lamin C protein deficiency in the primary fibroblasts from a new laminopathy case with ovarian cystadenoma

Background Laminopathies are a group of rare genetic disorders characterized by multiple-tissue *** describe a new laminopathy with ovarian cystadenoma and explore its molecular *** The case is a 15-year-old girl who presents the prominent progeroid disorders, multiple system degeneration and early-onset cystadenoma of the *** genes including LMNA, ZMPSTE24, PPAR G, INSR and WRN were sequenced to screen for DNA *** mRNA and protein expression levels of LMNA were examined in primary *** pathophysiological events such as morphologic alterations, cell senescence, cell proliferation,apoptosis and pRb as well as p53 protein expressions were also investigated in primary *** No mutation was identified in the candidate genes *** abnormalities including nuclear blebs,mislocalization of lamin A/C were evident in the patient ***, nucleus exhibited nuclear herniation and almost complete loss of peripheral *** addition, lamin C protein expression was markedly reduced whereas lamin A protein level was normal and no prelamin A was detected in the primary *** the senescence-associated β-galactosidase staining of patient＇ cells was negative, cells in S phase increased in accompany with a decrease in pRb protein ***, increases in apoptotic cell death and p53 expression were *** Our data suggest that selective deficiency of lamin C protein is associated with a case of laminopathy with ovarian *** abnormalities in nuclear structure and alterations in gene expression such as the decrease in pRb and increase in p53 may be responsible for the multiple tissue degeneration.

目的探讨胰岛素和格列齐特对2型糖尿病大鼠肝脏内质网应激和胰岛素敏感性的影响。方法高脂饮食联合链脲佐菌素建立2型糖尿病大鼠模型，随机分为糖尿病组、胰岛素组和格列齐特组并设正常对照组。胰岛素组和格列齐特组分别予以中效胰岛素和格列齐特治疗3周。检测肝脏免疫球蛋白结合蛋白（bip）、剪切的x盒结合蛋白1（XBP-1s）、磷酸化c-Jun（p-c-Jun）、丝氨酸磷酸化胰岛素受体底物1（p-IRS-1）、葡萄糖6磷酸酶（G6Pase）蛋白表达。结果与正常对照组比较，糖尿病组bip（0．28-4-0．07比0．90±0．10）、XBP-1s（0．41±O．07比0．95±0．07）、p-c-Jun（0．59±0．18比1．94±0．03）、p-IRS-1（1．73±0．18比5．324-0．22）和G6Pase（0．11±0．01比0．45±0．01）表达均增加，P值均〈0．01。胰岛素和格列齐特治疗后，与糖尿病组比较，胰岛素组和格列齐特组bip（0．90±0．10比0．25±0．04和0．53±0．02，P值均〈0．01）、XBP-1s（0．95±0．07比0．47±0．01和0．78±0．02，P〈0．01和P〈0．05）、P-c-Jun（1．94±0．03比0．50±0．10和1．33±-O．11，P值均〈0．01）、p-IRS-1（5．32±0．22比1．59±0．32和3．13±0．02，P值均〈0．01）以及G6Pase蛋白（0．45±0．01比0．15±0．02和0．25±0．01，P值均〈0．01）表达均下降，以胰岛素组下降更明显（P值均〈0．01）。结论胰岛素和格列齐特均能减轻肝脏内质网应激、下调c-Jun氨基端激酶活性以及改善胰岛素敏感性；以胰岛素的效应更强，提示除了降糖以外，胰岛素可能还有抗炎、抗氧化应激或促使脂质重分布等作用。

目的探讨早期胰岛素治疗对糖尿病大鼠肝脏NF—KB炎症信号通路的影响。方法通过实时定量PCR和Westernblot检测肝脏磷酸烯醇丙酮酸羧激酶（PEPCK）蛋白表达，胞质NF—KB抑制因子（IKBα）及核NF—KB蛋白表达，TNFα、IL-6、IL-1B基因表达，EliSA测定NF—KBp65DNA结合活性。结果与正常对照组比较，糖尿病大鼠肝脏PEPCK蛋白表达增加40％，早期胰岛素和格列奇特治疗后，PEPCK蛋白明显下降。与正常对照组比较，糖尿病大鼠肝脏IKBα蛋白水平降低，细胞核NF—KBp65蛋白及NF-KBp65DNA结合活性表达增加，TNFα和IL-1B基因表达分别增加1．75倍和0．47倍；早期胰岛素和格列奇特治疗使IKBα蛋白表达增加，NF—KBp65蛋白及NF—KBp65DNA结合活性表达下降，TNFα、IL-18基因表达下降。而晚期治疗未见早期治疗的变化。结论早期胰岛素治疗可抑制糖尿病大鼠肝脏活化的NF—KB信号通路和炎症因子，其机制与早期代谢控制改善糖脂毒性有关。

目的探讨早期胰岛素治疗对2型糖尿病大鼠骨骼核因子（NF）-κB炎症信号通路的影响。方法建立高脂喂养联合小剂量链脲佐菌素诱导的2型糖尿病SD大鼠模型。糖尿病大鼠随机分为早期和晚期治疗组：早期治疗在血糖升高后3d开始，给予中效胰岛素或格列奇特治疗3周；晚期治疗在血糖升高后4周开始，给予中效胰岛素治疗3周。血糖控制目标是随机血糖〈8mmol／L。通过实时定量PCR和Western印迹检测骨骼肌葡萄糖转运子4（Glut4）基因和蛋白表达，胞质IκBα蛋白表达，肿瘤坏死因子（TNF）-α、白细胞介素（IL）-6、IL-1β基因表达，EliSA测定NF-κBP65DNA结合活性。结果糖尿病大鼠骨骼肌Glut4mRNA表达水平比正常对照下降59％，膜上Glut4蛋白水平下降69％。与未治疗组比较胰岛素和格列奇特治疗使Glut4mRNA表达增加了17％和13％，膜上蛋白表达增加23％和10％（0．12±0．02vs0．21±0.07和0．16±0．03，P〈0．05），并且使胞质内Glut4蛋白水平降低。糖尿病大鼠骨骼肌IKBα蛋白比正常对照降低（0．28±0．02 vs 0．36±0．06，P〈0．05）；NF-κBP65DNA结合活性增加，TNF-α、IL-1β、IL-6基因表达上调。早期胰岛素和格列奇特治疗使IKBα蛋白比未治疗组表达增加（0．41±0．06和0．39±0．05 vs 0．28±0．02，P〈0．05），NF-κBP65DNA结合活性下降，骨骼肌TNF-α基因表达下降。结论早期胰岛素治疗可抑制2型糖尿病大鼠骨骼肌细胞活化的NF-κB信号通路和炎症因子，其机制和早期代谢控制改善糖脂毒性有关。

目的探讨过氧化物增殖激活受体α（PPAR-α）激动剂非诺贝特对大鼠糖皮质激素受体（GR）表达调节作用。方法30只雄性SD大鼠随机分为对照组，FE1组（非诺贝特50mg·kg^-1·d^-1，1个月），FE2组（非诺贝特100mg·kg^-1·d^-1，1个月），FE3组（非诺贝特100mg·kg^-1·d^-1，2个月），MK886组[同时给予非诺贝特（100mg·kg^-1·d^-1）和PPARa抑制剂MK886（30mg·kg^-1·d^-1），1个月]。检测sD大鼠肝脏、肌肉、脂肪组织GR基因和蛋白表达水平，同时测定肾上腺11β-羟化酶（CYP11B1）基因表达水平，并以放免法测定大鼠血皮质酮水平。结果非诺贝特显著下调了大鼠肝脏组织GR基因和蛋白质表达水平，FE1、FE2和FE33组GRmRNA水平分别较正常组降低55％、54％、68％，蛋白质表达水平分别降低了28％、77％和99％；并升高肾上腺CYP11B1表达水平及血中皮质酮水平，而MK886完全逆转了非诺贝特的这些效应[正常组、FE1、FE2、FE3、MK886组皮质酮水平分别为（393±23）、（495±44）、（516±18）、（622±93）、（382±37）ng／ml]，提示非诺贝特可抑制肝脏GR表达，并促进肾上腺皮质酮合成增多，这些作用依赖于PPARα激活。结论PPARα激活剂非诺贝特可抑制肝脏GR表达。

We investigate high time resolution data obtained by the Gravitational wave high-energy Electromagnetic Counterpart All-sky Monitor(GECAM)during the flare event on 2022 April 21 at 01:52 *** subpeaks with durations of 4-6 s have been detected in the hard X-ray precursor phase,and the key feature is that they appear in pairs and seem like double-peak *** subpeaks are rarely observed in hard X-ray band and confirmed by the microwave obtained by Nobeyama Radio Polarimeters(NoRP)and Radio Solar Telescope Network(RSTN).While an exponential function can describe the continuum component of the time profile from the precursor to part of the impulsive *** periods of quasi-periodic pulsations(QPPs)are detected to be about 7.3 and 12.8 s for the precursor and impulsive phase,respectively,with at least 95%confidence *** paired QPPs are assumed to be double-peak QPPs and then the scenario of current loop coalescence model is found to be in good agreement with our *** precursor phase can be interpreted as the oscillating coalescence of two islands,while the impulsive phase can be interpreted as more islands to coalesce one by one to form larger islands.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

Background:The Ebola virus disease spread rapidly in West Africa in 2014,leading to the loss of thousands of *** engagement was one of the key strategies to interrupt Ebola transmission,and practical community level measures needed to be explored in the field and tailored to the specific context of ***:First,community-level education on Ebola virus disease(EVD)prevention was launched for the community’s social mobilizers in six districts in Sierra Leone beginning in November ***,from January to May of 2015,in three pilot communities,local trained community members were organized to engage in implementation of EVD prevention and transmission interruption measures,by involving them in alert case report,contact tracing,and social *** epidemiological indicators of transmission interruption in three study communities were ***:A total of 6016 community social mobilizers from 185 wards were trained by holding 279 workshops in the six districts,and EVD message reached an estimated 631680 *** three pilot communities,72 EVD alert cases were reported,with 70.8%of them detected by trained local community members,and 14 EVD cases were finally *** tracing detected 64.3%of EVD *** median duration of community infectivity for the cases was 1 *** secondary attack rate was 4.2%,and no third generation of infection was *** health worker was infected,and no unsafe burial and noncompliance to EVD control measures were *** community-based measures were modeled to reduce 77 EVD cases,and the EVD-free goal was achieved four months earlier in study communities than whole country of Sierra ***: The community-based strategy of social mobilization and community engagement was effective in casedetection and reducing the extent of Ebola transmission in a country with weak health system.
