Sarcomas constitute a rare heterogeneous group of tumors,including a wide variety of histological *** advances in our understanding of the pathophysiology of the disease,first-line sarcoma treatment options are still limited and new treatment approaches are *** H2AX phosphorylation is a sensitive marker for double strand breaks and has recently emerged as biomarker of DNA damage for new drug *** this study,we explored the role of H2AX phosphorylation at Ser139 alone or in combination with MAP17 protein,an inducer of DNA damage through ROS increase,as prognostic biomarkers in sarcoma ***,we proposed doxorubicin and olaparib combination as potential therapeutic strategies against sarcomas displaying high level of both *** evaluate retrospectively the levels of pH2AX(Ser139)and MAP17 in a cohort of 69 patients with different sarcoma types and its relationship with clinical and pathological *** found that the levels of pH2AX and MAP17 were related to clinical features and poor ***,we pursued PARP1 inhibition with olaparib to potentiate the antitumor effect of DNA damaging effect of the DNA damaging agent doxorubicin to achieve an optimal synergy in *** demonstrated that the combination of olaparib and doxorubicin was synergistic in vitro,inhibiting cell proliferation and enhancing pH2AX intranuclear accumulation,as a result of DNA *** synergism was corroborated in patient-derived xenografts(PDX)where the combination was effective in tumors with high levels of pH2AX and MAP17,suggesting that both biomarkers might potentially identify patients who better benefit from this combined therapy.

Background: The in vivo concentration of bicarbonate(HcO3-), one of the essential sperm capacitating effectors,varies greatly in the different environments sperm go through from cauda epididymis to the fertilisation site. On the contrary, porcine in vitro sperm capacitation and fertilisation media usually contains a standard concentration of25 mmol/L, and one of the main problems presented is the unacceptable high incidence of polyspermy. This work hypothesised that by modifying the HcO3-concentration of the medium, the output of in vitro sperm capacitation and fertilisation could be ***: Once exposed to the capacitation medium, the intracellular pH(pHi) of spermatozoa increased immediately even at low concentrations of HcO3-, but only extracellular concentrations of and above 15 mmol/L increased the substrates protein kinase A phosphorylation(pPKAs). Although with a significant delay, 15 mmol/L of HcO3-stimulated sperm linear motility and increased other late events in capacitation such as tyrosine phosphorylation(Tyr-P) to levels similar to those obtained with 25 mmol/L. This information allowed the establishment of a new in vitro fertilisation(IVF)system based on the optimization of HcO3-concentration to 15 mmol/L, which led to a 25.3% increment of the viable zygotes(8.6% in the standard system vs. 33.9%).conclusions: Optimising HcO3-concentrations allows for establishing an IVF method that significantly reduced porcine polyspermy and increased the production of viable zygotes. A concentration of 15 mmol/L of HcO3-in the medium is sufficient to trigger the in vitro sperm capacitation and increase the fertilisation efficiency in porcine.

Si-B-c ceramic composites were synthesized using SiB-6,B-4c,and phenolic resin as a carbon source by pressureless sintering in an Ar ***,the Si-B-c ceramic composites were fabricated to determine their potential for applications as high hardness and high temperature *** X-ray diffraction patterns of sintered bodies of SiB-6-B-4c with carbonized phenolic resin can be seen that SiB-6 and c changed to B-4c and *** this study,it is obtained that carbonized phenolic resin is good addition material as a reaction material comparing to carbon powder at 1683 K for 1 h by pressureless sintering in an Ar atmosphere.

Influence of biomass burning on ozone levels in the Megalopolis of central Mexico during the cOVID-19 lockdown

The massive reductions in anthropogenic emissions resulting from the cOVID-19 lockdown provided a unique opportunity to evaluate the effect of mitigation measures aiming to abate air *** Mexico,the total lockdown period took place during the dry-hot season when biomass burning activity is ***,we investigate the role of biomass burning emissions on regional ozone levels in the Megalopolis of central *** studied period covers the lockdown phases 2 and 3,and the first month of the New *** applied a factor separation technique and process analysis to estimate the pure and synergistic contributions of emission reductions under lockdown and that from biomass burning to daily ozone maximum concentrations in 7 metropolitan areas of different states in the *** results revealed that biomass burning plumes likely masked the effect of massive reductions from mobile emissions,impacted the PBL development during phase 3 and favored transition and mixed NO_(x)-limited and VOc-limited regional *** contributed to increased ozone production in the middle to lower PBL by changing the regional background levels which potentially could bias high ozone production efficiency *** the Megalopolis contribution to economic and societal development at national scale,our study suggests that ozone mitigation measures during the dry-hot season targeting mainly mobile emissions will likely be offset by biomass burning plumes.A regional and synergic policy aiming to control biomass burning would help to reduce the occurrence of high ozone levels in central Mexico with the co-benefit of tackling short-lived climate pollutants.

Resource manipulation reveals flexible allocation rules to growth and reproduction in a Mediterranean evergreen oak

Aims In plants,resource allocation to growth and reproduction may depart from trade-off expectations if(i)investment in growth and reproduction relies on different resource pools,(ii)allocation to reproduction is dependent upon reaching some growth threshold or(iii)reproduction is developmentally linked to growth,both functions relying on the same resource *** examined the effects of enhanced resource level on patterns of resource allocation to growth and reproduction in holm oak(Quercus ilex ***),a Mediterranean evergreen *** In the experimental year(2003),we manipulated the amount of soil nutrients in autumn(to increase nutrient uptake during shoot elongation in the following spring)and soil water in summer(to increase water uptake during acorn growth).Indicators of growth and male and female reproduction were estimated in the preexperimental(2002),experimental(2003)and post-experimental(2004)*** Findings Fertilized trees produced significantly longer shoots,but the number of female flowers per shoot was not affected by *** production of male catkins was also enhanced by *** did not affect the production of female flowers or abortion *** and female reproduction showed no consistent relationship in untreated trees,but resource addition elicited a growth-female reproduction trade-off in the experimental *** sign of this significant relationship changed in the post-experimental year,indicating the existence of lagged effects of resource manipulation on acorn ***,patterns of allocation to growth and reproduction varied as a function of sex,resource availability and year,a result consistent with extreme allocational plasticity in holm oak.

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOD-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOD-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOD-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. We then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.

The main objective of this work was to use reline deep eutectic solvent,containing Al(III)ions,for the electrochemical study of the nucleation and growth of aluminum onto a glassy carbon electrode at different temperatures and angular speeds(ω)of the working *** order to fulfill this,electrochemical and surface characterization techniques were *** was found that as temperature increased,the onset of the Al(III)DES reduction occurred at less negative potentials while the current peak of the voltammograms *** indicate that Al deposition thermodynamics and kinetics were ***,no anodic current was detected due to Al passivation by Al(OH)_(3)(s)andγ-Al_(2)O_(3)(s).Atω=0 r/min,the Al deposition chronoamperograms were analyzed by a theoretical model comprising Al 3D diffusion-controlled nucleation and growth and residual water ***,those recorded at different angular speeds were analyzed with a theoretical model where adsorption−desorption and diffusion-controlled nucleation−growth occurred *** deposits were characterized by SEM,EDX,XPS and ***ω=0 r/min,formation of well distributed nanoparticles((78.1±9.5)nm)was observed,while atω=900 r/min the deposit was formed by multiple 10μm diameter leaf-like flat microstructures,composed by Al,Al(OH)_(3)(s)andγ-A2O3(s).
