A systematic review of progenitor survival and maturation in Parkinsonian models

stem cell-based brain repair is a promising emergent therapy for Parkinson's disease based on years of foundational research using human fetal donors as a cell *** current therapeutic options for patients,this approach has the potential to provide longterm stem cell–derived reconstruction and restoration of the dopaminergic input to denervated regions of the brain allowing for restoration of certain functions to *** ultimate clinical success of stem cell–derived brain repair will depend on both the safety and efficacy of the approach and the latter is dependent on the ability of the transplanted cells to survive and differentiate into functional dopaminergic neurons in the Parkinsonian *** the pre-clinical literature suggests that there is considerable variability in survival and differentiation between studies,the aim of this systematic review was to assess these parameters in human stem cell-derived dopaminergic progenitor transplant studies in animal models of Parkinson's disease.A defined systematic search of the PubMed database was completed to identify relevant studies published up to March *** screening,76 articles were included in the analysis from which 178 separate transplant studies were *** these,graft survival could be assessed in 52 studies and differentiation in 129 ***,we found that graft survival ranged fromcells transplanted,with a median of 51%of transplanted cells surviving in the brain;while dopaminergic differentiation of the cells ranged from 0% to 46% of cells transplanted with a median of 3%.This systematic review suggests that there is considerable scope for improvement in the differentiation of stem cell-derived dopaminergic progenitors to maximize the therapeutic potential of this approach for patients.

Answering Reachability Queries on Incrementally Updated Graphs by Hierarchical Labeling schema

We proposed a novel solution schema called the Hierarchical Labeling schema （HLs） to answer reachability queries in directed graphs. Different from many existing approaches that focus on static directed acyclic graphs （DAGs）, our schema focuses on directed cyclic graphs （DcGs） where vertices or arcs could be added to a graph incrementally. Unlike many of the traditional approaches, HLs does not require the graph to be acyclic in constructing its index. Therefore, it could, in fact, be applied to both DAGs and DcGs. When vertices or arcs are added to a graph, the HLs is capable of updating the index incrementally instead of re-computing the index from the scratch each time, making it more efficient than many other approaches in the practice. The basic idea of HLs is to create a tree for each vertex in a graph and link the trees together so that whenever two vertices are given, we can immediately know whether there is a path between them by referring to the appropriate trees. We conducted extensive experiments on both real-world datasets and synthesized datasets. We compared the performance of HLs, in terms of index construction time, query processing time and space consumption, with two state-of-the-art methodologies, the path-tree method and the 3-hop method. We also conducted simulations to model the situation when a graph is updated incrementally. The performance comparison of different algorithms against HLs on static graphs has also been studied. Our results show that HLs is highly competitive in the practice and is particularly useful in the cases where the graphs are updated frequently.

A mathematical model for the dynamical transmission of polio is considered,with the aim of investigating the impact of environment *** model captures two infection pathways through both direct human-to-human transmission and indirect human-to-environment-to-human transmission by incorporating the environment as a transition and/or reservoir of *** derive the basic reproduction number R0^env We show that the disease free equilibrium is globally asymptotically stable(GAs)if R0^env1,there exists a unique endemic equilibrium which is locally asymptotically stable(LAs).similar results hold for environmental contamination free sub-model(without the incorporation of the indirect transmission).At the endemic level,we show that the number of infected individuals for the model with the environ mental-related contagion is greater than the corresponding number for the environmental contamination free *** conjunction with the inequality R0s the basic reproduction number for the environmental contamination free sub-model,our finding suggests that the contaminated environment plays a detrimental role on the transmission dynamics of polio disease by increasing the endemic level and the severity of the ***,it is natural to implement control strategies to reduce the severity of the disease by providing adequate hygienic living conditions,educate populations at risk to follow rigorously those basic hygienic rules in order to avoid adequate contacts with suspected contaminated ***,we perform numerical simulations to support the theory.

Design and mathematical analysis of activating transcriptional amplifiers that enable modular temporal control in synthetic juxtacrine circuits

The ability to control mammalian cells such that they self-organize or enact therapeutic effects as desired has incredible *** only would it further our understanding of native processes such as development and the immune response,but it would also have powerful applications in medical fields such as regenerative medicine and *** control is typically obtained by synthetic circuits that use synthetic receptors,but control remains *** synthetic juxtacrine receptors(sJRs)are widely used as they are fully modular and enable spatial control,but they have limited gene expression amplification and temporal *** these are integral facets to cell control,I therefore designed transcription factor based amplifiers that amplify gene expression and enable unidirectional temporal control by prolonging duration of target gene *** a validated in silico framework for sJR signaling,I combined these amplifiers with sJRs and show that these sJR amplifier circuits can direct spatiotemporal patterning and improve the quality of self-organization.I then show that these circuits can improve chimeric antigen receptor(cAR)T cell tumor killing against various heterogenous antigen expression *** amplifiers are flexible tools that improve control over sJR based circuits with both basic and therapeutic applications.

Background: Human donor sclera is commonly used in glaucoma implant surgery to cover the external portion of the silicone tube and prevent its erosion through the overlying conjunctiva. corneo-scleral graft is used in this study as an alternative material for its desirable properties of being transparent, higher in tensile strength, cheap and more readily available. Methods: Eighteen consecutive cases were implanted with the Ahmed glaucoma valve im-plant. The limbal portion of the silicone tube was covered by a comeo-scleral graft obtained from the corneo-scleral rim left over following corneal transplant operation. The effectiveness in protecting the tube from erosion or migration, the transparency of the patch graft and complications encountered after surgery were ***: At a mean follow-up of 6 months, all tubes were well protected and no complications related to the patch graft were encountered during intra and postoperative period. Overview of surface ozone variability in East Asia-North Pacific region during IGAc/APARE(1994—1996)

surface ozone(O\-3) was measured at Oki Island(Japan), cheju Island(south Korea), Lanyu Island(Taiwan Province, china), cape D'Aguilar(Hong Kong sAR) and Lin'an, Longfenshan, Waliguan(china mainland) during January 1994—December 1996 as a component of IGAc/APARE(International Global Atmospheric chemistry/East Asia-North Pacific Regional Experiment). This paper gave a joint discussion on the observational results at these stations over the study region. Investigations showed that the average of surface O 3 mixing ratios at the seven sites are 47.9±15.8, 48.1±17.9, 30.2±16.4, 31.6±17.5, 36.3±17.5, 34.8±11.5 and 48.2±9.5 ppbv, respectively. significant diurnal variations of surface O 3 have been observed at Oki, cheju, D’Aguilar, Lin'an and Longfenshan. Their annual averaged diurnal differences range from 8 to 23 ppbv and differ in each season. surface O 3 at Lanyu and Waliguan do not show strong diurnal variability. seasonal cycles of surface O 3 showed difference at the temperate and the subtropical remote sites. Oki has a summer minimum-spring maximum, while Lanyu has a summer minimum-autumn maximum. The suburban sites at D'Aguilar and Lin'an report high-level O 3 in autumn and low level O 3 in summer. surface O 3 remains high in autumn and low in winter at the rural site Longfenshan. For the global background station Waliguan, surface O 3 exhibits a broad spring-summer maximum and autumn-winter minimum. The backward air trajectories to these sites have shown different pathways of long-range transport of air pollution from East Asia continent to North Pacific Ocean. surface O 3 was found to be strongly and positively correlated with cO at Oki and Lanyu, especially in spring and autumn, reflecting the substantial photochemical buildup of O\-3 on a regional scale. It is believed that the regional sources of pollution in East Asia have enhanced the average surface O 3 concentrations in the background atmosphere of North Pacific.

Residues of Polychlorinated Biphenyls (PcBs) in Fish, Water and sediment from shing Mun River

The level and pattern of contamination by polychlorinated biphenyls (PcBs) were investigated in tilapia, Oreochromis mossambicus (Peters), sediment, and water from the shing Mun River. The range of total PcBs was 12.9 ng/g to 181.6 ng/g wet weight in tilapia, 12.7 ng/g to 46.0 ng/g freeze-dried weight in sediment, and 3.8 ng/L to 13.6 ng/L in water. The effect of biomagnification was also observed, PcB concentrations increased from water to sediment to tilapia. PcB congeners occurred in such a way that lower chlorinated PcBs comprised a higher fraction of the total PcBs in water, sediment, and tilapia muscle, whereas higher chlorinated PcBs were more commonly found only in tilapia.

Analysis and behaviour of FRP-confined short concrete columns subjected to eccentric loading

Fibre-reinforced polymer (FRP) composites were widely utilized in civil engineering structures as the retrofit of reinforced concrete (Rc) columns. To design FRP jackets safely and economically, the behaviour of such columns should be predicted first. This paper is concerned with the analysis and behaviour of FRP-confined Rc circular and rectangular short col- umns subjected to eccentric loading. A simple design-oriented stress-strain model for FRP-confined concrete in a section analysis was first proposed. The accuracy was then proved by two test data. Following that, a parametric study including amount of FRP confinement, FRP strain capacity, unconfined concrete strength and shape of column section is provided. some conclusions were obtained at the end of the paper. The work here will provide a comprehensive understanding of the behaviour of FRP-confined concrete columns. The simplicity of the model also enables a simple equivalent stress block to be developed for direct use in practical design.
