Sac.harides are a sort of ubiquitous and vital molec.les within the whole ***,the applic.tion of sac.harides analysis with matrix-assisted laser desorption/ionization mass spec.rometry(MALDI-MS)is restric.ed by their low ionization effic.enc. and the instability of the sialic.ac.d *** strategy based on nonreduc.ive amination provides a good solution,however,this is often time c.nsuming and may result in sample loss due to removal of exc.ssive derivatization ***,hydralazine(HZN)was utilized as a reac.ive matrix for labeling reduc.ng sac.harides direc.ly on MALDI target whic. eliminated tedious sample preparation and avoided sample *** optimization,effec.ive and reproduc.ble on-MALDI-target derivatization of neutral and ac.dic.sac.harides was ac.ieved in both positive and negative *** with 2,5-dihydroxybenzoic.ac.d(DHB)and 9-aminoac.idine(9-AA),HZN improved the detec.ion sensitivity of reduc.ng sac.harides and provided more abundant fragment ions in MS/MS ***,26 kinds of neutral glyc.ns and 5 kinds of sialic.glyc.ns were identified from ovalbumin(OVA)and bovine fetuin,*** with the statistic.l models,this strategy c.uld be used to distinguish and predic. samples of 6 brands of beer,and disc.iminate 2 kinds of beer fermentation *** addition,HZN was applied for quantitative analysis of gluc.se in urine samples,and the obtained urine gluc.se c.nc.ntrations of diabetic.patients were c.nsistent with the c.0}ic.l test results,showing the potential of qualitative and quantitative analysis of reduc.ng sac.harides in c.mplex samples.

Degradation and resynthesis of c.lorophyll during inc.eased oxidative stress and prolonged darkness differ between annual and perennial flax(linum L.)

Among plants,there is c.nsiderable variation in lifespan:annuals live less than one year,whereas perennials live for several years,with the longest-living perennial having survived 43,600 *** proposed by the Disposable Soma Theory,this lifespan variation among plants likely reflec.s differential investment of limited energy and nutrient resourc.s,with perennials investing more energy and nutrients into biomolec.lar maintenanc. c.mpared to annuals in order to ensure persistenc. over multiple *** differential investment may be partic.larly important during periods of exogenous stress,whic. are known to ac.elerate biomolec.lar *** present study evaluated this hypothesis using annual and perennial flax(linum L.)subj.c.ed to two exogenous stressors—inc.eased oxidative stress(i.e.,foliar H2O2spraying)and c.mplete prolonged *** c.lorophyll has been shown to exhibit degradation in response to c.anges in environmental c.nditions,we utilized c.anges in c.lorophyll levels during and after periods of exogenous stress to evaluate our *** predic.ed that i)perennials would exhibit a slower rate of c.lorophyll degradation during exposure to exogenous stressors c.mpared to annuals,and ii)perennials would exhibit a faster rate of c.lorophyll resynthesis following suc. exposure c.mpared to *** levels before,during,and after exposure to both exogenous stressors were measured in two separate trails,onc. using image c.lour analysis and onc. using *** c.lorophyll degradation rates in response to oxidative stress did not differ between annuals and perennials,c.ntrary to our predic.ions,c.lorophyll resynthesis rates following suc. exposure were signific.ntly higher in perennials,as *** plants were subj.c.ed to c.mplete prolonged darkness,c.lorophyll degradation rates were signific.ntly lower in perennials than annuals,as predic.ed;however,when plants were subsequently reintroduc.d to natural photoperiod,c.

A multi-relaxation-time disc.ete Boltzmann model(DBM) with split c.llision is proposed for both subsonic.and supersonic.c.mpressible reac.ing flows, where c.emic.l reac.ions take plac. among various c.mponents. The physic.l model is based on a unified set of disc.ete Boltzmann equations that desc.ibes the evolution of eac. c.emic.l spec.es with adj.stable ac.eleration, spec.fic.heat ratio, and Prandtl number. On the right-hand side of disc.ete Boltzmann equations, the c.llision,forc., and reac.ion terms denote the c.ange rates of distribution func.ions due to self-and c.ossc.llisions, external forc.s, and c.emic.l reac.ions, respec.ively. The sourc. terms c.n be c.lc.lated in three ways, among whic. the matrix inversion method possesses the highest physic.l ac.urac. and c.mputational effic.enc.. Through c.apman-Enskog analysis, it is proved that the DBM is c.nsistent with the reac.ive Navier-Stokes equations, Fic.'s law and the Stefan-Maxwell diffusion equation in the hydrodynamic.limit. c.mpared with the one-step-relaxation model, the split c.llision model offers a detailed and prec.se desc.iption of hydrodynamic. thermodynamic. and c.emic.l nonequilibrium effec.s. Finally, the model is validated by six benc.marks, inc.uding multic.mponent diffusion, mixture in the forc. field, Kelvin-Helmholtz instability, flame at c.nstant pressure, opposing c.emic.l reac.ion, and steady detonation.

Heat stress induc.d hepatoc.te apoptosis in largemouth bass Mic.opterus salmoides via IRE1α/TRAF2/ASK1/j.K pathway

Heat stress(HS)has been shown to adversely affec. fish livers and c.n lead to extensive *** investigate the relationship between endoplasmic.retic.lum(ER)stress and HS-induc.d apoptosis in fish livers,we isolated and c.ltured primary hepatoc.tes of largemouth bass,Mic.opterus salmoides by trypsin method,then established an in-vitro model of liver c.lls under HS(35℃).The c.ntents of lac.ic.dehydrogenase(LDH)and hydrogen peroxide(H2O2)were determined to evaluate the effec.s of HS on hepatoc.te inj.ry and oxidative ***-qPc. was performed to disc.ver the key genes in unfolded protein response(UPR)pathways involved at different HS *** inhibitor 4-PBA and IRE1αinhibitor 4μ8c.were used to further investigate the effec.s of HS on IRE1αapoptosis pathway in *** show that HS led to signific.nt inc.eases in the release of LDH,the c.ntent of H2O2,and the expressions of oxidative protein folding genes(ero1αand pdi)under HS,suggesting severe hepatoc.te inj.ry and oxidative stress happened in heat-stressed largemouth bass *** c.ntinuous ac.ivation of IRE1αpathway genes(grp78,grp94,atf6,perk,eif2a,atf4,c.op,ire1α,traf2,ask1,j.k1,and j.k2)indic.ted that HS led signific.ntly to ER *** partic.lar,the mRNA expression levels of ER stress-related genes(grp78,grp94,atf6,perk,ire1α,c.op,j.k1,and j.k2)in the high temperature(HT)+4-PBA group and the HT+4μ8c.group were signific.ntly down-regulated under *** 4μ8c.treatment,the expression levels of apoptosis-related genes(c.spase-2,c.spase-3,c.spase-6,c.spase-7,c.spase-8,c.spase-9,and c.spase-10)and LDH c.ntent were signific.ntly dec.eased,whereas the c.ll survival rate was signific.ntly inc.eased when given 4-PBA or 4μ8c.*** findings demonstrate that HS c.uld induc. liver apoptosis of largemouth bass through the IRE1αpathway,whic. may ac. as a key switc. mediating liver apoptosis of largemouth bass under HS.
