Dipeptidyl peptidase-4: A key player in c.ronic.liver disease

Dipeptidyl peptidase-4 (DPP-4) is a membrane-assoc.ated peptidase, also known as c.26. DPP-4 has widespread organ distribution throughout the body and exerts pleiotropic.effec.s via its peptidase ac.ivity. A representative target peptide is gluc.gon-like peptide-1, and inac.ivation of gluc.gon-like peptide-1 results in the development of gluc.se intoleranc./diabetes mellitus and hepatic.steatosis. In addition to its peptidase ac.ivity, DPP-4 is known to be assoc.ated with immune stimulation, binding to and degradation of extrac.llular matrix, resistanc. to anti-c.nc.r agents, and lipid ac.umulation. The liver expresses DPP-4 to a high degree, and rec.nt ac.umulating data suggest that DPP-4 is involved in the development of various c.ronic.liver diseases suc. as hepatitis c.virus infec.ion, non-alc.holic.fatty liver disease, and hepatoc.llular c.rc.noma. Furthermore, DPP-4 oc.urs in hepatic.stem c.lls and plays a c.uc.al role in hepatic.regeneration. In this review, we desc.ibed the tissue distribution and various biologic.l effec.s of DPP-4. Then, we disc.ssed the impac. of DPP-4 in c.ronic.liver disease and the possible therapeutic.effec.s of a DPP-4 inhibitor.

c. ambrosioides is a c.ngolese medic.nal plant used in the treatment of several pathologies. This work was initiated to evaluate the analgesic.and antipyretic.effec. of the aqueous extrac. of leaves of c. ambrosioides. Analgesic.effec. was evaluated by using the ac.tic.ac.d-induc.d writhing, the Tilk Flic. test as well as the pain induc.d by formaldehyde. The results obtained showed that the aqueous extrac. signific.ntly reduc.s the number of abdominal writhes (p c. ambrosioides (400 and 600 mg/kg) c.mpared to c.ntrol group (saline water). This result suggests an antipyretic.effec.. c.emic.l sc.eening revealed the presenc. of alkaloids, flavones and muc.lage. The presenc. of alkaloids and flavones c.uld explain the analgesic.and antipyretic.effec.s attributed to this aqueous extrac..

Desmodium velutinum (D. velutinum) is a plant of the family Fabac.ae used in traditional c.ngolese medic.ne for the treatment of high blood pressure (HBP) or hypertension (HT). The present work evaluated the hypotensive ac.ivity and determined the phytoc.emic.l profile of the aqueous extrac. and frac.ions of Desmodium velutinun (Willd) D. c. (Fabac.ae) in the rat. The hypotensive effec. of the aqueous extrac. and different frac.ions (F1, F2, F3 and F4) was evaluated at the dose of 2.5 mg/kg in normotensive rats anesthetized with 15% urethane at 1.5 g/kg, i.p. (intraperitoneally). Our results showed that the F3 frac.ion after administration c.used a signific.nt dec.ease in blood pressure and heart rate whic. remained higher than that of the aqueous extrac. and frac.ions (F1, F2 and F4). The ric.ness in polyphenolic.and flavonoid c.mpounds as well as the separating c.pac.ty of the eluent used within the F3 frac.ion c.early justifies the benefic.al effec.s of the latter on blood pressure and heart rate c.mpared with the extrac. and frac.ions (F1, F2 and F4). In a sec.nd part we c.rried out the qualitative (thin-layer c.romatography) and quantitative (total polyphenols and flavonoids determination) analysis. The c.emic.l c.mpounds remained dominated by the polyphenolic.and flavonoid groups, the c.ntent of total polyphenols and total flavonoids meets the following inc.easing order: (extrac., F1, F2, F3 and F4). The results obtained c.uld c.rtify the use of Desmodium velutinun in traditional c.ngolese medic.ne.
