Platelet aggregation is affected by nitrosothiols in patients with chronic hepatitis: In vivo and in vitro studies

AIM: To investigate the relationship among the number of platelets and plasma levels of S-nitrosothiols (S-NO), nitrite, total non-protein SH (NPSH), glutathione (GSH), cysteine (cYS), malondialdehyde (MDA), 4-hydroxininenal (4HNE), tumor necrosis factor-alpha (TNFα) and interleukin (IL)-6 in patients with chronic hepatitis c (cH).METHODS: In vitro the aggregation of platelets derived from controls and cH patients was evaluated before and after the addition of adenosine diphosphate (ADP) and collagen, both in basal conditions and after incubation with nitrosoglutathione (GSNO). RESULTS: In vivo , S-NO plasma levels increased significantly in cH patients and they were significantly directly correlated with platelet numbers. Patients with platelet counts crease in S-NO, lower levels of GSH, cYS, NPSH, TNFα, and IL-6, and higher levels of nitrite, MDA, and 4-HNE relative to those of patients with platelet counts > 150 000/μL. In vitro, the ADP and collagen aggregation time was increased in platelets from patients and not from controls; in addition, platelets from cH patients but not from controls also showed a latency time after exposure to collagen. cONcLUSION: The incubation of platelets with GSNO improved the percentage aggregation and abolished the latency time.

Ignition and combustion modelling in a dual fuel diesel engine

A numerical simulation of a single cylinder research diesel engine fuelled by natural gas and diesel oil in dual fuel mode was conducted to test the reaction mechanism presented by Li and Williams in Ref.[1]for methane *** mechanism made of only 9 reactions can represent a good compromise between reduction of computational time and accuracy of *** reproduce test cases previously carried out experimentally and numerically with a simpler kinetic mechanism at three different premixed ratios(10%,15% and 22%).Finally,a last case characterized by a supply of methane consistent with the typical load levels for this kind of engines(80%),was investigated only *** the simulations were performed with the KIVA-3V solver on a geometry which includes open valve periods,intake and exhaust *** a comparison between experimental and numerical results,a calibration of the model has been performed and a quite good fitting of the models has been achieved.

Energy valorization of organic waste material is nowadays an assessed practice of circular *** heat and power(cHP)technologies based on biomass gasification represent viable substitutes to traditional energy conversion units based on combustion,whose need has recently experienced a huge growth,due to the increasing concerns about the release of greenhouse gases(GHGs)emissions and the related effects on climate *** present,only a few solutions have yet achieved a level of full development for *** of them is the system developed by cMD,the cMD EcO20,made of a gasifier,a syngas cleaning system and a spark ignition internal combustion engine working as a *** the present work,a numerical model is developed to study this system into detail and search for optimal controlling *** simulation relies on a combined use of the Thermoflex?environment and a proper one-dimensional(1D)model of the engine module built within GT-Suite?.An original contribution is given to the turbulent combustion model that accounts for the laminar flame speed of the specific *** numerical model,that covers the entire biomass-to-energy conversion process,is validated under real operative *** final purpose of the work is the optimization of input parameters,as the initial biomass moisture content,the equivalence ratio at the gasifier or the timing of spark advance,to maximize the system electrical energy output.

Probiotic VSL#3对于慢性肝病肝功能异常参数的有益影响

Objectives: To evaluate whether chronic therapy with probiotics affects plasma levels of cytokines and oxidative/nitrosative stress parameters, as well as liver damage, in patients with various types of chronic liver disease. Patients and Methods: A total of 22 nonalcoholic fatty liver disease (NAFLD) and 20 alcoholic liver cirrhosis (Ac) patients were enrolled in the study and compared with 36 HcV-positive patients with chronic hepatitis without (20, cH) or with (16, cc) liver cirrhosis. All patients were treated with the probiotic VSL#3. Routine liver tests, plasma levels of tumor necrosis factor alpha (TNF-α), interleukin (IL)-6 and -10, malondialdehyde (MDA), and 4-hydroxynonenal (4-HNE), S-nitrosothiols (S-NO), were evaluated on days -30, 0, 90, and 120. Results: Treatment with VSL#3 exerted different effects in the various groups of patients: in NAFLD and Ac groups, it significantly improved plasma levels ofMDA and 4-HNE, whereas cytokines (TNF-α, IL-6, and IL-10) improved only in Ac patients. No such effects were observed in HcV patients. Routine liver damage tests and plasma S-NO levels were improved at the end of treatment in all groups. conclusions: Results of the study suggest that manipulation of intestinal flora should be taken into consideration as possible adjunctive therapy in some types of chronic liver disease.
