Helicobacter pylori and other Helicobacter species DNA in human bile samples from patients with various hepato-biliary diseases

AIm： To investigate the presence of Helicobacter species by nested PcR of 16S rRNA genes followed by the presence of Flelicobacter pylori （H pylori） 16S rRNA, ureA, cagA genes in bile obtained at endoscopic retrograde cholangio-pancreatography （ERcP） from 60 Indian subjects. mETHODS： Sixty bile samples were obtained from patients diagnosed with various hepato-biliary diseases and control subjects at ERcP. PcR analysis was carried out using primers for Helicobacter genus 16S rRNA gene and H pylori （16S rRNA, urea and cagA） genes. Gastric H pylori status was also assessed from biopsies obtained at endoscopy from patients with various hepatobiliary diseases and controls. The control group mainly consisted of subjects with gastric disorders. Sequencing analysis was performed to confirm that PcR products with 16S rRNA and cagA primers were derived from H pylori. RESULTS No Helicobacters were grown in culture from the bile samples. Helicobacter DNA was detected in bile of 96.7% and 6.6% of groups Ⅰ and Ⅱ respectively. Ten from group Ⅰ were positive for 16S rRNA and ureA and 9 were positive for cagA gene. In contrast of the 2 from the control, 1 amplified with 16S rRNA, ureA and cagA primers used. The sequences of the 16S rRNA genes and cagA were 99% similar to Helicobacter pylori. cONcLUSION： Helicobacters are associated with the pathogenesis of various hepato-biliary disorders.

In a wide variety of mechanical and industrial applications,e.g.,space cooling,nuclear reactor cooling,medicinal utilizations(magnetic drug targeting),energy generation,and heat conduction in tissues,the heat transfer phenomenon is ***’s law of heat conduction has been used as the foundation for predicting the heat transfer behavior in a variety of real-world *** model’s production of a parabolic energy expression,which means that an initial disturbance would immediately affect the system under investigation,is one of its main ***,numerous researchers worked on such problem to resolve this *** last,this problem was resolved by cattaneo by adding relaxation time for heat flux in Fourier’s law,which was defined as the time required to establish steady heat conduction once a temperature gradient is *** offered a material invariant version of cattaneo’s model by taking into account the upper-connected derivative of the Oldroyd ***,both models are combinedly known as the cattaneo-christov(cc)*** this attempt,the mixed convective mHD Falkner-Skan Sutterby nanofluid flow is addressed towards a wedge surface in the presence of the variable external magnetic *** cc model is incorporated instead of Fourier’s law for the examination of heat transfer features in the energy expression.A two-phase nanofluid model is utilized for the implementation of *** nonlinear system of equations is tackled through the bvp4c technique in the mATLAB software *** influence of pertinent flow parameters is discussed and displayed through different *** and important results are summarized in the conclusion ***,in both cases of wall-through flow(i.e.,suction and injection effects),the porosity parameters increase the flow speed,and decrease the heat transport and the influence of drag forces.

Sources and Ecological Risks of Heavy metals in Soils Under Different Land Uses in Bangladesh

Soil heavy metal pollution, influenced by both natural and anthropogenic factors, significantly reduces environmental quality. In this study, cr, Ni, cu, As, cd, and Pb in eight different land-use soils from Patuakhali District in Bangladesh were assessed. concentrations of cr, Ni, cu, As, cd, and Pb in soils were 1-87, 5-271, 4-181, 0-80, 0.2-24.0, and 5-276 mg kg-1, respectively, measured using an inductively coupled plasma-mass spectrometer. The enrichment factor, pollution load index (PLI), and contamination factor (cJ) of metal i were used to assess the ecological risk posed by metals in soils. The PLI ranged from 0.78 to 2.66, indicating baseline levels to progressive deterioration of soil due to metal contamination. However, c;of cd ranged from 1.8 to 12.0, which showed that the studied soils were strongly impacted by cd. considering the severity of the potential ecological risk of a single metal, the descending order was cd > As > Pb > cu > Ni > cr. Soils under all land uses showed moderate to very high potential ecological risk.

characterization of hepatic progenitors from human fetal liver during second trimester

AIm： To enrich hepatic progenitors using epithelial cell adhesion molecule （EpcAm） as a marker from human fetal liver and investigate the expression of human leukocyte antigen （HLA） and their markers associated with hepatic progenitor cells. mETHODS： EpcAm ＋ve cells were isolated using magnetic cell sorting （mAcS） from human fetuses （n = 10） at 15-25 wk gestation. Expression of markers for hepatic progenitors such as albumin, alpha-fetoprotein （AFP）, cD29 （integrin ~1）, cD49f （integrin c^6） and cD90 （Thy 1） was studied by using flow cytometry, immunocytochemistry and RT-PcR; HLA class Ⅰ （A, B, c） and class Ⅱ （DR） expression was studied by flow cytometry only. RESULTS： FAcS analysis indicated that EpcAm ＋ve cells were positive for cD29, cD49f, cD90, cD34, HLA class I, albumin and AFP but negative for HLA class Ⅱ （DR） and cD45. RT PcR showed that EpcAm ＋ve cells expressed liver epithelial markers （cK18）, biliary specific marker （cK19） and hepatic markers （albumin, AFP）. On immunocytochemical staining, EpcAm ＋ve cells were shown positive signals for cK18 and albumin. cONcLUSION： Our study suggests that these EpcAm ＋ve cells can be used as hepatic progenitors for cell transplantation with a minimum risk of alloreactivity and these cells may serve as a potential source for enrichment of hepatic progenitor.

Butt-welding Residual Stress of Heat Treatable Aluminum Alloys

This study, taking three types of aluminum alloys 2024-T351, 6061-T6 and 7075-T6 as experimental materials, conducted single V-groove GTAW （gas tungsten arc welding） butt-welding to analyze and compare the magnitude and differences of residual stress in the three aluminum alloys at different single V-groove angles and in restrained or unrestrained conditions. The results show that the larger the grooving angle of butt joint, the higher the residual tensile stress. Too small grooving angle will lead to dramatic differences due to the amount of welding bead filler metal and pre-set joint geometry. Therefore, only an appropriate grooving angle can reduce residual stress. While welding, weldment in restrained condition will lead to a larger residual stress. Also, a residual stress will arise from the restraint position. The ultimate residual stress of weldment is determined by material yield strength at equilibrium temperature. The higher the yield strength at equilibrium temperature, the higher the material residual stress. Because of its larger thermal conductivity, aluminum alloy test specimens have small temperature differential. Therefore, the residual tensile stress of all materials is lower than their yield strength.

Detection of Breaking Waves Using X-Band Pulse Radar in the Nearshore Region

An X-band pulsed Doppler microwave radar has been used to determine the characteristics of breaking waves. Field experiments were conducted at the Shuang-Si estuary in the north of Taiwan in the winter of 2005. Analyses on maxima radar cross section and Doppler frequency shift are done to characterize wave breaking zones. Based on observations of breaking waves, the wave breaking zones are shown to be located at water depths of 1.8 to 2.2 m in the experimental site. In general, the results indicate that a radar system has the potential to delineate the spatial variation of breaking waves clearly and that this is sufficient to achieve a measurement operation for near-shore air-sea interaction events.

cotton is considered as a major cash crop of the world. It earns huge foreign exchange by its valuable products;fiber, lint, cotton seed oil, hull and a lot more. Being an important fiber crop, it earns huge foreign exchange by contributing to textile and seed oil industry. This review summarizes cotton biology, its diversity and domestication, genome assembly, constraints in its production and methods to improve cotton plant to fulfill the need of textile and oil industry. But cotton is facing enormous biotic and abiotic stresses with insect pests being most prominent. massive destruction caused by insects needs to be controlled for maintaining fruitful cotton crop production. conventional breeding approaches are limited to improving single trait and integrate stable genes within plant genome in approximately 7 - 8 years. Improved biotechnological procedures have paved new pathways to target genes specifically and improve cotton germplasm in lesser time than conventional breeding.

Genome Editing Tools: Need of the current Era

Genome editing is considered as the most widely used approach of the present era. It had become a basic need of the current micro and molecular biological experiments. Gene engineering finds its widespread applications in medical, industry and agricultural sector. Unlike previous genetic engineering practices to insert or delete a part of genetic material at random place, genome editing allows the precise manipulation of DNA at a specific location. Zinc Finger Nucleases (ZFNs), Transcription Activator like Effector Nucleases (TALENs), clustered Regularly Interspresed Short Palindromic repeats (cRISPR/cas system) and meganucleases (recombinases) are the prime tools for editing an organism’s genome. Genome editing tools have an advantage to selectively delete or to integrate specific genes at specific loci. Use of recombinases for specifying site has further reduced time to integrate genes site specifically. Site specific gene stacking by the use of recombinases coupled with ZFNs, TALENs, or cRISPR/cas genes have paved new pathways to target genes site specifically and to improve germplasm in lesser time than conventional breeding approaches.

mixing features in an electromagnetic rectangular micromixer for electrolyte solutions

In this study, a new magnetohydrodynamic（mHD） mixer for electrolyte solutions with pumping function is reported, and the mixing performance of the device for two different electrolyte solutions is numerically examined in a uniform magnetic field. Application of different potentials to different electrodes allows the current to be induced. The combination of the induced current and magnetic field yields Lorentz force, resulting in the fluid motion. The numerical simulation for the flows in the device is carried out with commercial software cFX. The validity of cFX code for the present numerical model is presented. The mixing performance of the fluids is investigated in many different cases with different combinations of input voltage of the electrode. This study shows that the mixing performance can be enhanced by applying spatially alternating positive and negative voltages to the electrodes. The present simulation results show that with a magnetic field intensity lower than 0.5 T, a voltage difference smaller than 2.0 V, and an electric conductivity of electrolyte solution of 1.5 S/m the pumping capabilities ranging 1.6×10^（-7）-3.6×10^（-6） kg/s and the mixing indexes higher than 0.90 can be obtained with sophisticated designs of the micromixer.
