The reference reach method is commonly used for natural stream design. In the reference reach approach, a stable reach is selected to serve as the design template for a design reach. The planform and profile of this reference reach must be scaled to fit the design site. currently, the leading method for scaling uses a simple ratio of cross-sectional areas. This paper presents a simple method that employs hydraulic scaling to size the design reach to accommodate the desired bankfull flow. The method introduced in this paper is easy to use, does not require additional survey data, and should lead to more robust channel design.

Background:Human embryonic stem cells(hEScs)represent a tremendous resource for cell therapies and the study of human development;however to maintain their undifferentiated state in vitro they routinely require the use of mouse embryonic fibroblast(MEF)feeder-layers and exogenous protein media ***:These well established requirements can be overcome and in this study,it will be demonstrated that phenotypic stability of hEScs can be maintained using a novel,human plasma protein-based hydrogel as an extracellular culture matrix without the use of feeder cell *** were resuspended in human platelet poor plasma(PPP),which was gelled by the addition of calcium containing DMEM-based hESc culture *** and genomic expression of the pluripotency markers OcT4,NANOG and SOX2 were measured using immunohistochemistry and qRT-PcR *** hESc morphology was demonstrated throughout in vitro culture and both viability and phenotypic stability were maintained throughout extended culture,up to 25 ***:PPP-derived hydrogel has demonstrated to be an efficacious alternative to MEF co-culture with its hydrophilicity allowing for this substrate to be delivered via minimally invasive procedures in a liquid phase with polymerization ensuing in *** this provides a novel technique for the study of this unique group of stem cells in either 2D or 3D both in vitro and in vivo.

Desert resource environments(***,food)are tied to limited,highly localized rainfall regimes which generate microgeographic variation in the life histories of ***,enhanced growth rates,reproduction and survivorship are observed in response to increased resource availability in a variety of desert plants and short-lived *** examined the thermal ecology and reproduction of US federally threatened Mojave desert tortoises(Gopherus agassizii),long-lived and large-bodied ectotherms,at opposite ends of a 250-m elevation-related rainfall cline within Ivanpah Valley in the eastern Mojave Desert,california,*** operative environments in both the upper-elevation,“cima,”and the lower-elevation,“Pumphouse,”plots corresponded with daily and seasonal patterns of incident solar *** received 22%more rainfall and contained greater perennial vegetative cover,which conferred 5°c-cooler daytime shaded *** a monitored average rainfall year,cima tortoises had longer potential activity periods by up to several hours and greater ephemeral *** resource availability in cima was associated with larger-bodied females producing larger eggs,while still producing the same number of eggs as Pumphouse ***,reproductive success was lower in cima because 90%of eggs were depredated versus 11%in Pumphouse,indicating that predatory interactions produced counter-gradient variation in reproductive success across the rainfall ***-use impacts on deserts(*** energy generation)are increasing rapidly,and conservation strategies designed to protect and recover threatened desert inhabitants,such as desert tortoises,should incorporate these strong ecosystem-level responses to regional resource variation in assessments of habitat for prospective development and mitigation efforts.

Applying dispersants to spilled crude oil results in an oil in water dispersion of microscopic oil droplets that soon dilutes to levels below 1 ppm oil. These levels are substantially below those known to induce acute toxicity in 96-hour tests. We show here that oil hydrocarbons are biodegraded very rapidly in such situations (50% loss in 4 days in this experiment), and that no increase in acute toxicity to mysids (Americamysis bahia) is seen during this biodegradation, or over the following 20 days as the oil is further degraded (78% loss of detectable hydrocarbons in this experiment).

A key control point in gene expression is the initiation of protein translation, with a universal stress response being constituted by in- hibitory phosphoryiation of the eukaryotic initiation factor 2α （el F2oL）. In humans, four kinases sense diverse physiological stresses to regulate elF2α to control cell differentiation, adaptation, and survival. Here we develop a computational molecular model of elF2α and one of its kinases, the protein kinase R, to simulate the dynamics of their interaction. Predictions generated by coarse-grained dynamics simulations suggest a novel mode of action. Experimentation substantiates these predictions, identifying a previously unrecognized interface in the protein complex, which is constituted by dynamic residues in both elF2α and its kinases that are crucial to regulate protein translation. These findings call for a reinterpretation of the current mechanism of action of the el F2α kinases and demonstrate the value of conducting computational analysis to evaluate protein function.
