Multi-length scale modeling is performed to(i)predict the carburized case depth of SAE8620 steel gears by solving the Fick’s second law of diffusion,(ii)model the martensitic microstructure evolution in a grain inside the carburized case as well as to study the effect of stress cycling on retained austenite(RA)and martensite using a 3d phase-field model,(iii)simulate the effect of carburization and different RA contents on macroscale fatigue behavior of SAE8620 steel spur gear using the finite element *** diffusion model predicts that the case depth increases with increasing heat treatment time and *** phase-field simulations show that RA can transform to martensite during fatigue loading,where the extent of the transformation will depend on the type of stresses applied,*** in a high stress regime or low stress regime of fatigue *** transformation of martensite to austenite is also observed in low RA sample under high stress *** macroscale simulations show that the carburized case with high RA gives rise to better fatigue life compared to that with low RA.

It is difficult to obtain in vivo digestion kinetics data of high protein ingredients using chickens. Collecting kinetics data requires repeated sampling of digesta from the small intestine during the digestion process,which is not easily accomplished due to the anatomical structure of chicken digestive tract. An in vitro technique is proposed for measuring the digestion kinetics of protein sources fed to chickens. The method has a 30 min gastric and 3 h intestinal phase. Five hundred milligram crude protein(CP)equivalent of each meal sample(CP = % N × 6.25) was digested with pepsin(28,260 units) in 50 m L polyethylene centrifuge tubes for 30 min in a shaking water bath(150 strokes/min; 30 mm stroke length)at 41C. The 6.5 mL pancreatin was selected as the enzyme concentration for the intestinal phase, during which time 500 m L aliquots were collected at 0, 15, 30, 45, 60, 90, 120, 150, 180 and 240 min. Samples were diluted 1:820 with HCl and sodium acetate buffer, and then mixed with ninhydrin reagent(2:1) at100 ± 2C for 15 min and spectrometric readings taken at 568 nm. To validate the assay, 5 replications of soybean meal(SBM), corn gluten meal(CGM), corn distillers dried grains with solubles(CddGS), porcine meal(PCM), fish meal(FM) and casein(CA) were digested. The digestion data were modeled with PROC NLIN procedure, and the intra coefficient of variation(CV) assessed using PROC MEANS of SAS 9.4. The digestion values at 180 min were SBM 95 ± 4, FM 93 ± 3, PCM 68 ± 4, CGM 82 ± 3 and CddGS 70 ± *** CV for SBM, CGM, CddGS, PCM and FM were 5%, 5%, 12%, 10% and 2%, respectively. The estimated fractional digestion rates for SBM, CGM, CddGS, FM and PCM were 0.023, 0.013, 0.009, 0.024 and 0.013,respectively. In conclusion, the proposed in vitro technique estimated the rate and extent of the digestion of CP for the meals with low intra CV.

Ischemia-reperfusion plays a major role in the injury experienced by the liver during transplantation. Much work has been done recently investigating the role of redox species in hepatic ischemia-reperfusion. As animal models are better characterized and developed, and more insights are gained into the pathophysiology of hepatic ischemia reperfusion injury in humans the questions into exactly how oxidants participate in this injury are becoming more refined. These questions include effects of cellular location, timing of injury, and ability of therapeutics to access this site are increasing our appreciation of the complexity of ischemia reperfusion and improving attempts to ameliorate its effects. In this review, we aim to discuss the various methods to alter redox chemistry during ischemia reperfusion injury and future prospects for preventing organ injury during hepatic ischemia reperfusion.

Hydrothermal carbonization （HTC）, utilizing high temperature and pressure, has the potential to treat agricultural waste via inactivating pathogens, antibiotic resistance genes （ARG）, and contaminants of emerging concern （CEC） in a environmental and economical manner. Livestock mortality is one facet of agricultural waste that can pose a threat to the surrounding environment. While several methods are utilized to treat livestock mortality, there remains a paucity of data on the elimination of microbially- derived dNA in these treatment practices. This dNA, most notably ARGs, if it survives treatment can be reintroduced in agricultural environments where it could potentially be passed to pathogens, posing a risk to animal and human populations. HTC treatments have been successfully utilized for the treatment of CECs, however very little is understood on how ARGs survive HTC treatment. This study aims to fill this knowledge gap by examining the survivability ofmicrobially-derived dNA in the HTC treatment of livestock mortality. We examined three treatment temperatures （100℃, 150℃, and 200℃） at autogenic pressures at three treatment times （30, 60, and 240 min）. We examined the amplification of a plasmid-bome reporter gene carried by Escherichia coli dH 10B introduced to both beef bone and tissue. Results indicate that while all three temperatures, at all treatment times, were suitable for complete pathogen kill, only temperatures of 150℃ and 200℃ were sufficient for eliminating microbial dNA. These results serve as the basis for future potential HTC treatment recommendations for livestock mortality when considering the elimination of pathogens and ARGs.

Introduction:The availability of essential nutrients,such as nitrogen(N)and phosphorus(P),can feedback on soil carbon(C)and the soil microbial *** cycles can be supplemented by agricultural fertiliser addition,and we determined whether the stoichiometry and nutrient limitation of the microbial biomass could be affected by an unbalanced nutrient ***:Samples were taken from a long-term trial(in effect since 1968)with annual applications of 0,15 and 30 kg P ha^(−1) with constant N and *** and microbial biomass CNP contents were measured and nutrient limitation assessed by substrate-induced *** regression and discriminant analyses were used to identify the variables explaining nutrient ***:Soil and biomass CNP increased with increasing P fertiliser,and there was a significant,positive,correlation between microbial biomass P and biomass C,apart from at the highest level of P fertilisation when the microbial biomass was over-saturated with *** molar ratios of C:N:P in the microbial biomass remained constant(homeostatic)despite large changes in the soil nutrient *** growth was generally limited by C and N,except in soil with no added P when C and P were the main limiting nutrients.C,N and P,however,did not explain all the growth limitation on the soils with no added ***:Increased soil C and N were probably due to increased net primary *** results confirm that C:N:P ratios within the microbial biomass were constrained(***)under near optimum soil *** with no added P were characterised by strong microbial P limitation and soils under high P by over-saturation of microorganisms with *** changes in biomass C:P can be indicative of nutrient limitation within a site.

X-linked hypophosphatemia (XLH) is a rare disease of elevated fibroblast growth factor 23 (FGF23) production that leads tohypophosphatemia and impaired mineralization of bone and teeth. The clinical manifestations of XLH include a high prevalence ofdental abscesses and periodontal disease, likely driven by poorly formed structures of the dentoalveolar complex, including thealveolar bone, cementum, dentin, and periodontal ligament. Our previous studies have demonstrated that sclerostin antibody (SclAb) treatment improves phosphate homeostasis, and increases long bone mass, strength, and mineralization in the Hyp mousemodel of XLH. In the current study, we investigated whether Scl-Ab impacts the dentoalveolar structures of Hyp mice. Male andfemale wild-type and Hyp littermates were injected with 25 mg·kg−1 of vehicle or Scl-Ab twice weekly beginning at 12 weeks of ageand euthanized at 20 weeks of age. Scl-Ab increased alveolar bone mass in both male and female mice and alveolar tissue mineraldensity in the male mice. The positive effects of Scl-Ab were consistent with an increase in the fraction of active(nonphosphorylated) β-catenin, dentin matrix protein 1 (dMP1) and osteopontin stained alveolar osteocytes. Scl-Ab had no effecton the mass and mineralization of dentin, enamel, acellular or cellular cementum. There was a nonsignificant trend towardincreased periodontal ligament (PdL) attachment fraction within the Hyp mice. Additional PdL fiber structural parameters were notaffected by Scl-Ab. The current study demonstrates that Scl-Ab can improve alveolar bone in adult Hyp mice.
