Potential of four marine-derived fungi extracts as anti-proliferative and cell death-inducing agents in seven human cancer cell lines

Objective:To evaluate the in vitro anticancer activity of crude ethyl acetate extracts of the culture of four marine-derived fungi Aspergillus similanensis KUFA 0013(E1),Neosartorya paulistemis KUFC 7897(E2),Neosartorya siamensis KUFA 0017(E4) and Talaromyces trachyspermus KUFC 0021(E3) on a panel of seven human cancer cell ***:Effects on cell proliferation,induction of DNA damage and cell death were assessed by MTT and clonogenic assays,comet assay and nuclear condensation assay,***:The proliferation of HepG2,HCTl 16 and A375 cells decreased after incubation with the extracts E2 and *** anti-proliterative effect was confirmed by morphologic alterations and by clonogenic *** extracts also induced cell death in HepG2 and HCT116 *** was used as a positive control and showed in vitro anticancer ***:This study demonstrated,for the first time,that extracts of Neosartorya paulistensis and Neosartorya siamensis have selective anti-proliferative and cell death activities in HepG2,HCT16 and A375 *** bioactivity of these extracts suggests a potential for biotechnological applications and substantiates that both should be further considered for the elucidation of the molecular targets and signal transduction pathways involved.

Agglomerates based on milk whey proteins and modified starch (MS) were developed for patients with dysphagia. Calcium caseinate (CaCas), whey protein isolate (WPI), concentrate (WPC) and hydrolysate (WPH) were used. The sources were agglomerated with the MS and an increase in the porosity and viscosity of the agglomerates were observed. In all the systems evaluated, the WPI agglomerate at a concentration of 112 g/L showed a viscosity between 2122 and 5110 cP, and the agglomerates of WPC and WPH between 1115?-?2880 cP and 2600 - 6651 cP, respectively. CaCas exhibited high values in water and milk of 3200 cP and 6651 cP, respectively, and low values of 640 cP in juice. In sensory tests, the 70% WPI: 30% MS juice obtained a score 6.97, an improvement in relation to the other agglomerates, but not differing (p = 0.681) from the commercial thickener, 6.91 (p = 0.380). Based on these results, the 70% WPI: 30% MS was suggested for use in the nutritional therapy of patients with dysphagia.

Background：Due to high neutral lipids accumulation in the cytoplasm,in vitro-produced embryos from Bos primigenius indicus and their crosses are more sensitive to chilling and cryopreservation than those from Bos primigenius *** objective of the present study was to evaluate the effects of trans-10,cis-12 conjugated linoleic acid（CLA） on the development and cryotolerance of crossbred Bos primigenius taurus x Bos primigenius indicus embryos produced in vitro,and cultured in the presence of fetal calf *** zygotes（n = 1,692）were randomly assigned to one of the following treatment groups：1） Control,zygotes cultured in Charles Rosenkrans 2 amino acid（CR2aa） medium（n = 815） or 2） CLA,zygotes cultured in CR2 aa medium supplemented with 100 μmol/L of trans-10,cis-12 CLA（n =877）.Embryo development（cleavage and blastocyst rates evaluated at days 3 and 8 of culture,respectively）,lipid content at morula stage（day 5） and blastocyst cryotolerance（re-expansion and hatching rates,evaluated 24 and 72 h post-thawing,respectively） were compared between ***,selected mRNA transcripts were measured by Real-Time PCR in blastocyst ***：The CLA treatment had no effect on cleavage and blastocyst rates,or on mRNA levels for genes related to cellular stress and *** the other hand,abundance of mRNA for the 1-acylglycerol-3-phosphate0-acyltransferase-encoding gene（AGPAT）,which is involved in triglycerides synthesis,and consequently neutral lipid content,were reduced by CLA treatment.A significant increase was observed in the re-expansion rate of embryos cultured with trans-10,cis-12 CLA when compared to control（56.3 vs.34.4%,respectively,P = 0.002）.However,this difference was not observed in the hatching rate（16.5 vs.14.0%,respectively,P=0.62）.Conclusions：The supplementation with trans-10,ds-12 CLA isomer in culture medium reduced the lipid content of in vitro produced bovine embryos by reducing the gene expression of 1-acylglycerol 3-phosphate 0

BACKGROUND The association between the intestinal microbiota and psychiatric disorders is becoming increasingly *** gut microbiota contributes to colorectal carcinogenesis(CRC),as demonstrated with colibactin-producing Escherichia coli(CoPEC).AIM To evaluate the association between CoPEC prevalence and anxiety-and depressive-like behaviors with both preclinical and clinical *** Patients followed after a CRC surgery and for whom the prevalence of CoPEC has been investigated underwent a psychiatric *** were compared according to the CoPEC *** parallel C57BL6/J wild type mice and mice with a CRC susceptibility were chronically infected with a CoPEC *** behavior was assessed using the Elevated Plus Maze test,the Forced Swimming Test and the Behavior recognition system PhenoTyper®.RESULTS In a limited cohort,all patients with CoPEC colonization presented with psychiatric disorders several years before cancer diagnosis,whereas only one patient(17%)without CoPEC *** result was confirmed in C57BL6/J wildtype mice and in a CRC susceptibility mouse model(adenomatous polyposis colimultiple intestinal neoplasia/+).Mice exhibited a significant increase in anxiety-and depressive-like behaviors after chronic infection with a CoPEC *** This finding provides the first evidence that CoPEC infection can induce microbiota-gut-brain axis disturbances in addition to its procarcinogenic properties.

Fire is an exceptional action that may occur during the life of a ***,it must be considered when designing a building *** standards provide several types of design methods for that propose,used for single elements,parts of structure or the structure as a *** fire design of columns is important both for new project as for remodel buildings and also for verification of the residual resistance of columns that have suffered a fire *** this way,the aim of this work is to analyze numerically different ways of fire exposure to check the compressive strength of the columns when subjected to fire and the influence of the adjacent walls to a column in case of *** thermal advanced analysis of the sections columns was performed using the finite element software,Abaqus CAE,where the standard fire curve,ISO 834(International Organization for Standardization 834),was used,with 4 h of fire *** was possible,with the two methods used in this work,to compare them to verify which model is more conservative and which is closer to the advanced numerical model,for calculating temperatures in the column *** was checked that the walls act as thermal insulators,protecting part of the columns from the convection and radiation of the ***,the effects of raising the temperature over the compressive resistance of the reinforced concrete column,were reduced.

Oil palm (Elaeis guineensis Jacq.) is one of the agricultural crops with the greatest potentialfor vegetable oil production in Brazil. However, a disease of unknown etiology popularlyknown as Fatal Yellowin (FY) has caused damage to Brazilian farmers particularly in theeastern region of the Amazon. So, the objective of this study was to evaluate the spatialdependence of FY on three oil palm genotypes, grown for many years in an organic produc-tion system in the Amazon region. The study area had 4104 ha, divided into 139 plots. Ineach plot, the monthly incidence of disease was monitored forming a database. The num-ber of diseased plants per year, number of accumulated diseased plants, number of dis-eased plants per hectare, growth rate of diseased plants and incidence of accumulateddisease were evaluated. The results indicated spatial distribution of the variables adjustedto the gaussian, spherical and exponential models, with predominance of the first *** increases the purpose that FY is caused by biotic factors. The highest range achievedin the study was 2929 m indicating the susceptibility of the tested genotypes. Some plotsclose to the river had the highest incidence of the disease on the study, probably associatedwith higher soil moisture.

AIM： To investigate the effects of mangiferin on gas- trointestinal transit （GIT） in normal and constipated mice, together with the possible ***： Intragastrically-administered charcoal mealwas used to measure GIT in overnight starved Swiss mice. In the first experiments, mangiferin （3 mg/kg, 10 mg/kg, 30 mg/kg, and 100 mg/kg, po） or tegaserod （1 mg/kg, ip） were administered 30 min before the char- coal meal to study their effects on normal transit. In the second series, mangiferin （30 mg/kg） was tested on delayed GIT induced by several different pharma- cological agonists （morphine, clonidine, capsaicin） or antagonists （ondansetron, verapamil, and atropine） whereas in the third series, mangiferin （30 mg/kg, 100 mg/kg and 300 mg/kg） or tegaserod （1 mg/kg） were tested on 6 h fecal pellets outputted by freely fed mice. The ratio of wet to dry weight was calculated and used as a marker of fecal water content. RESULTS： Mangiferin administered orally significantly （P 〈 0.05） accelerated GIT at 30 mg/kg and 100 mg/kg （89% and 93%, respectively）, similarly to 5-hydroxytrypta- mine4 （5-H%） agonist tegaserod （81%） when compared to vehicle-treated control （63%）. Co-administered man- giferin （30 mg/kg） totally reversed the inhibitory effect of opioid agonist morphine, 5-HT3-receptor antagonist ondansetron and transient receptor potential vanilloid-1 receptor agonist capsaicin on GIT, but only to a partial extent with the GIT-delay induced by ~2-adrenoceptor agonist clonidine, and calcium antagonist verapamil. However, co-administered atropine completely blocked the stimulant effect of mangiferin on GIT, suggesting the involvement of muscarinic acetylcholine receptor activation. Although mangiferin significantly enhanced the 6 h fecal output at higher doses （245.5±10.43 mg vs 161.9±10.82 mg and 227.1±20.11 mg vs 161.9±10.82 mg of vehicle-treated control, at 30 and 100 mg/ kg, P 〈 0.05, respectively）, the effect of tegaserod was more potent （297.4±7.42 mg vs 161.9±1

Hybrid combinations of Eucalyptus have increased due to expansion of plantations into unconventional areas and to the search for higher quality ***,most of these species have difficulties surviving in vitro *** chlorine and sealing systems are often used to reduce contamination and increase gas *** aim of the present study is to evaluate the establishment,multiplication,elongation and adventitious rooting of *** × *** clones(C1 and C2) and four active chlorine concentrations(0.000%,0.001%,0.003%,and 0.005%) were tested in the establishment and multiplication *** sealing forms(W/M,1/M and 3/M) and the same four active chlorine concentrations were applied to the elongation *** luminosities(dark and light)and three sealings(W/M,1/M and 3/M) were tested during adventitious *** chlorine concentration of0.005% led to the lowest fungal contamination rate and to the highest in vitro *** chlorine concentration of 0.003% resulted in the greatest length and highest number of shoots per explant in the multiplication *** were no phytotoxicity problems and the quality of plants grown in an environment with active chlorine was maintained in comparison with those grown in an *** increase in gas exchange in ventilation systems had a positive impact on the in vitro growth and development of plants.
